Q.1. Describe the experiments of Griffith and Avery & co workers related to transforming

Ans. Griffith (1928) demonstrated the effect of rough, smooth and heat-killed smooth and
mixture of rough and heat-killed smooth strains of Pneumococci on rats. When the rats were
injected with the virulent form of Pneumococci (smooth or S strain) the rats died because S
strains are covered by a polysaccharide sheath which protects the bacteria from the immune
system of rat. When another strain of pneumococcus without the polysaccharide coating (rough
or R strain) was injected into rats the rats didn't die. Similarly, the rats didn't die when injected
with heat killed S strains. However, the rats died when injected with a mixture of R strain and
heat-killed S strain bacteria. Griffith found that the blood of the dead rats contained live and
virulent S strains only. So he concluded that the harmless R cells must have been transformed
into virulent S cells and this transformation must have been brought about by some principle
(transforming principle) present in the heat-killed S cells. Avery and coworkers (1944) found that
this transforming principle was DNA. Heating might have killed the S strain bacteria but DNA
survived which was somehow taken by the non-virulent R cells and then R cells became
capable of producing protective polysaccharide coating using the gene of heat-killed S strain
and thus transformed into virulent S strain.

Ans. DNA replication is the process by which DNA makes a copy of itself during cell division.
First the double helix structure of DNA molecule is unzipped or unwinded by the action of
enzyme helicase resulting in separation of two single strands of DNA and formation of a 'Y'
shapped replication fork. SSB protein (single stranded binding protein) binds to the two
separated strands and prevents annealing (reformation of double helix) . The two strands are
oriented in different directions and are therefore replicated differently but replication of both the
strands proceeds from 5' to 3' end. The strand oriented in the 3' to 5' direction (towards the
replication fork) is called leading strand and the replication of this strand proceeds continuously
whereas in the other strand oriented in 5' to 3' direction (away from the replication fork) the
replication is discontinuous and is called the lagging strand.
A primer (a short piece of RNA synthesized by enzyme 'primase') binds to the end of the leading
strand and it serves as the starting point for DNA synthesis. Now new complementary
deoxyribonucleotide bases are added to the strand of DNA in 5' to 3' direction with the help of
enzyme DNA polymerase. The newly incoming nucleoside triphosphate forms a phosphodiester
linkage with the 3'-OH group of the growing polynucleotide chain. The nucleotide precursors
align opposite the strand by virtue of specific base pairing (A-T; G-C). The new strands are proof
read to ensure that there were no mistakes in the new DNA sequence. If there is a wrong
nucleotide incorporated then it is excised by 3'-5' exonuclease activity of DNA polymerase and
then the polymerase function resumes and DNA replication progresses.
When replication of one strand has progressed for some length, replication of the second strand
commences in the lagging strand. However, in the lagging strand numerous RNA primers bind
at various points along the lagging strand. This results in synthesis of short polynucleotide
chains (1000-2000 bases) in a discontinuous manner. These chunks of DNA are called Okazaki
fragments. When this process of DNA synthesis is completed then enzyme exonuclease strips
away the primers and the Okazaki fragments are joined by enzyme DNA ligase and thus making
a continuous strand.
The result of DNA replication is two DNA molecules each consisting of one new and one old
chain of nucleotides. This is why DNA replication is called semiconservative. Following
replication the new DNA automatically winds up into a double helix.

Ans. Transcription is a process by which a particular segment of DNA is copied into an RNA
strand. For transcription, DNA serves as a template. Enzyme RNA polymerase creates a
transcription bubble at a specific site with the help of one or more transcription factors, which
separates the two strands of DNA helix by breaking the hydrogen bonds between
complementary DNA nucleotides. RNA polymerase adds matching ribonucleotides to the
complementary nucleotides of one DNA strand in presence of precursor RNA nucleotides,
Mg++ and Mn++ resulting in RNA-sugar-phosphate backbone to form an RNA strand. There are
termination signals where RNA polymerase stops and releases the newly synthesized RNA. The

4. newly synthesized RNA is further processed by polyadenylation, capping and splicing. This
transcribed gene (RNA) may be a messenger RNA, ribosomal RNA or transfer RNA.

Ans. The relationship between the sequence of bases in DNA or mRNA and the sequence of
amino acids in proteins specified by them is called genetic code. It is a triplet of bases (codon:
three adjacent nucleotides) which specify an amino acid.
Characteristics:
1. Non-overlapping -- any base can be part of a codon and is not used in another codon and
each base is "read" only once.

6. 2. Degeneracy -- means one amino acid is coded for by more than one triplet (e.g. serine is
coded for by UCA, UCG, UCC, UCU, AGU, AGC). Only methionine (AUG) and tryptophan
(UGG) are coded by a single triplet each. Any triplet, however, codes for a single amino acid
only. The code is not universal but applicable to almost all the species.
3. There is a codon called initiation codon (AUG) which initiates the polypeptide synthesis and
there are three termination codons viz. UAA, UAG, UGA which are not read by any aminoacyl

Ans. Since mRNA carries messages for the synthesis of several proteins, the initiation codon
(AUG) serves as start signal for translation of a protein. When this AUG is present in the interior
of the mRNA it functions as codon for methionine but when present in the beginning it codes for
N-formyl methionine. In some cases the initiation codon is GUG. When a protein is fully
translated then translation stops due to presence of termination codons UAA, UAG and UGA
because these codons are not read by any aminoacyl-tRNA. These termination codons are
recognised by specific proteins called release factors which release the completed polypeptide.

incorporated in the daughter DNA during replication, it will not form proper hydrogen bonding
with the base of the template strand and the polymerase activity of DNA polymerase is inhibited
and the 3'-5' exonuclease activity of polymerase I would cut or excise the wrong nucleotide and
then the polymerase activity resumes and DNA replication continues.

Ans. Ribosome is a large complex of proteins and ribosomal RNA consisting of a large and a
small subunit which are snuggled together. However, these subunits move apart slightly during
translation. The particle mass of the most extensively studied ribosome of the prokaryotic E. coli
is approximately 2.5 ×106
Dalton with sedimentation coefficient 70S (subunits are 50S and 30S).
Likewise the eukaryotic ribosome (80S) consists of 60S and 40S subunits and their constituent
proteins and RNAs are different from those of E.coli ribosome. However, functionally both
prokaryotic and eukaryotic ribosomes are the same. Each ribosome has an aminoacyl binding

This is the first step in which amino acids are attached to their specific tRNAs resulting in the
formation of aminoacyl-tRNA (aa-tRNA; charged RNA). This is catalyzed by specific enzymes
called aminoacyl-tRNA synthetases. The aa-tRNA synthetases are remarkably specific as they
recognise not only the correct amino acid but also the appropriate tRNA molecule from the
whole range of tRNAs

Initiation of polypeptide chain formation:
In the 2nd step the charged tRNAs (aa-tRNAs) diffuse to the ribosome. The initiation of
polypeptide chain begins with the binding of initiator tRNA bearing methionine (Met-tRNA) in
eukaryotes and N-formyl methionine (FMet-tRNA) in prokaryotes, to start codon of mRNA
(AUG) already bonded to the P site of 30S subunit of ribosome which has all the necessary
enzymes for translation. The anticodon of the FMet-tRNA (CAU) binds to the codon AUG on
mRNA. The initiation stage is completed by binding the larger ribosomal unit to the smaller unit
resulting in the formation of the initiation complex. This is followed by binding appropriate
aa-tRNA to the matching codon at site A (aminoacyl binding site) which means the aminoacyl
units lie side by side for the formation of peptide bond in the next step.

9. Elongation of polypeptide chain:
In the elongation step the first amino acid (methionine) is joined to the second amino acid on the
second tRNA (aa2-tRNA) on A site. This reaction is catalyzed by peptidyl transferase. The free
tRNA on the P site leaves the mRNA and mRNA moves one codon over. The third charged
tRNA then gets attached to the matching third codon on the A site which was vacated by
leftward movement of one codon. Then again a peptide bond is formed between the Met-aa2
and the aa3 amino acid followed by shifting of mRNA one codon leftward from A site to P site as
the free tRNA2 dissociates. This process is repeated over and over again till the required chain
length is achieved.
Termination of polypeptide chain formation:
In the termination step once the ribosome has moved down to the chain terminating codon
(nonsense codon or STOP codon), the polypeptide is released and the ribosome can be reused
for synthesis of another protein. The polypeptide chain leaves the ribosome to assume its
unique secondary, tertiary and quaternary structures.

Ans. In prokaryotes the protein biosynthesis is regulated mostly at the transcriptional level. The
regulation of protein synthesis is based on the principle of "as and when required". Taking the
example of induction of β-galactosidase (lactase) in Escherichia coli. These bacteria synthesize
very small amount of galactosidase in a glucose containing medium but when these bacteria are
transferred to a lactose containing medium they start synthesizing this enzyme so that they
could utilize lactose and grow well. This ability of bacteria to synthesise a particular enzyme as
and when required is regulated by a set of genes and their control elements referred to as "Lac
Operon"(The collection of structural genes and their control elements is called an "Operon").
Lac Operon is responsible for the synthesis of three enzymes, namely β-galactosidase,
galactoside permease and thiogalactoside transacetylase and the corresponding three
structural genes for these enzymes are continuously located and referred to as Z, Y and A
genes respectively. These structural genes are preceded by two control elements, the O
(operator) and the P (promotor) and also a regulatory gene (I). RNA polymerase binds to the P
(promotor) during transcription.
When bacteria are grown in glucose medium the I gene produces I-mRNA which combines with
the ribosome and its translation results in the synthesis of a protein called Repressor. This
Repressor binds to the O gene and this prevents the transcription of Z, Y and A genes.
In presence of an inducer (lactose) the Repressor binds to the inducer (lactose) to form an
inducer - Repressor complex which has no affinity for the O gene. Therefore, the structural
genes Z, Y and A are transcribed to give lac-mRNA and thereby the synthesis of

inducer-Repressor interaction is reversible, when the concentration of inducer (lactose)
decreases the Repressor gets free and binds to the O gene and transcription of lac-operon is

Retroviruses: are RNA viruses which carry genetic information on RNA instead of DNA. When
such a virus enters a host cell it uses its own enzyme called reverse transcriptase to convert its
RNA into DNA which integrates into the host DNA which then transcribes and translates the viral
gene to produce the proteins required for making new copies of the virus. Examples: Ron's
sarcoma virus, HIV.

SSB proteins: The single stranded binding (SSB) proteins bind to the two separated DNA
strands during replication to prevent the premature annealing to prevent the two separated DNA

15. strands from forming the double helix again before the completion of DNA replication. These
SSB proteins are ubiquitous in all living organisms.
DNA-helicase: DNA replication begins with unwinding of its double helix resulting in separation
of the two single strands of DNA. This is done by binding of helicase to specific sites in the DNA
molecule. There are many helicases which function in all processes in which strand separation
must be catalyzed such as DNA replication, DNA repair and recombination, and transcription of
RNA.
3'-5' exonuclease activity: DNA polymerase I also possesses some 3'-5' exonuclease activity
which is important for "proof reading" during during DNA replication. If a nucleotide is wrongly
incorporated during DNA replication it is excised by the 3'-5' exonuclease activity. After excision
of the wrong nucleotide the normal activity of DNA polymerase for DNA replication resumes.
DNA-ligase: This enzyme is used to join the Okazaki fragments formed by the lagging strand
during DNA replication to make a continuous strand.
