тЧП Solid phase system - The antibodies can be attached to polystyrene tubes, glass beads,
nylon particles and magnetic particles which help in separation of bound and free
fractions.
Acceptability criteria of separation method -
тЧП It should be efficient to separate the two fractions without contamination of each other.
тЧП The separation does not cause any appreciable disturbance in the state of equilibrium
reached at that time.
тЧП It should be practical in terms of speed, simplicity, acceptability and cost.

Material required for ELISAs are :
тЧП polystyrene/polyvinyl chloride plates, typically having 96-wells coated to bind protein very
strongly.
тЧП a primary and/or secondary detection antibody. The primary detection antibody is a
specific antibody that only binds to the protein of interest, while a secondary detection
antibody is a second enzyme-conjugated antibody that binds a primary antibody that is
not enzyme-conjugated,
тЧП analyte/antigen, coating antibody/antigen,
тЧП buffer and
тЧП substrate/chromogen.

General steps of an ELISA immunoassay:
тЧП Coating of microtiter plate wells with antigen
тЧП Blocking (typically with the addition of bovine serum albumin [BSA]) of all unbound sites
to prevent false positive results
тЧП addition of primary antibody (e.g. rabbit monoclonal antibody) to the wells
тЧП addition of secondary antibody conjugated to an enzyme (e.g. anti-mouse IgG).
[Between each of these steps there are "wash" steps using a buffer such as
phosphate-buffered saline (PBS) and a non-ionic detergent, to remove unbound
material. The wells are washed two or more times during each wash step, depending on
the specific protocol being followed]
тЧП Detection - is carried out by the addition of a substrate to the enzyme that can generate
a color [The most commonly used enzymes are horseradish peroxidase (HRP) and
alkaline phosphatase (ALP). [The substrate for HRP is hydrogen peroxide and
tetramethylbenzidine (TMB) is used as a chromogen which results in a blue color
change. ALP measures the yellow color of nitrophenol after room temperature incubation
periods of 15 to 30 minutes and usually uses P-Nitrophenyl-phosphate (pNPP) as its
substrate].

Chemiluminescence ELISA - In this method a luminescent substrate is used and the chemical
reaction between the enzyme label and luminescent substrate emits light which is detected by
a luminometer.
Four major types of ELISA:
тЧП Direct ELISA (antigen-coated plate) - used for analyzing the immune response to an
antigen and to detect antigens.
тЧП Indirect ELISA (antigen-coated plate) - used to detect not only antigens, but also
endogenous antibodies and their concentration.
тЧП Sandwich ELISA (antibody-coated plate) - used to detect the presence of antigen in a
sample and for analysis of complex samples.

тЧП Competitive ELISA - used to detect antigen (small molecules and hormones)
concentration in a sample and most commonly used when only one antibody is available
for the antigen of interest. Direct, indirect and sandwich ELISAs can all be adapted to
this format.
