RADIOISOTOPES AND THEIR USE IN BIOCHEMISTRY
हम जानते हैं कि atoms एक element क
े सबसे छोटे घटक (component) हैं और एक atom में nucleus होता
है, जो protons (+ve charge) और neutrons (no charge) से बना होता है, जिसक
े चारों ओर negatively
charged electrons घूमते रहते हैं I nucleus में प्रोटॉन की संख्या (P) element का atomic number (Z) तथा
प्रोटॉन और neutron (N) की संख्या का क
ु ल योग atomic mass/mass number (A) कहलाता है I
Number of protons (P) = Number of electrons (E) = Atomic number (Z)
Mass number (A) = (P) + (N)

ऐसे atoms को समस्थानिक (isotopes) कहते हैं I इस तरह Isotopes are the atoms of the same element
that have the same number of protons in their atomic nuclei (same atomic number) but differing
in numbers of neutrons (different atomic masses) अर्थात एक ही तत्व क
े वो परमाणु जिनकी परमाणु
संख्या समान होती है किन्तु परमाणु भार अलग - अलग हों I उदाहरण: Hydrogen क
े तीन isotopes होते हैं
क्योंकि उनमें neutrons की संख्या अलग - अलग होती है I
जब किसी atom में neutrons की संख्या protons की संख्या से अधिक होती है यानि N:P ratio 1 से अधिक
होता है तो atom अस्थिर (unstable) हो जाता है और ऐसे atoms क
े nucleus में अतिरिक्त energy होती है I
इन unstable isotopes को radioisotopes, radionuclides या radioactive isotopes (रेडियोधर्मी
समस्थानिक) कहते हैं I ऐसे radioisotopes स्थिर (stable) होने क
े लिए radioactive decay प्रक्रिया से गुज़रते
हैं और particles (α,β) or electromagnetic radiations (gamma radiations) उत्सर्जित करते हैं I इस
उत्सर्जन को radioactivity (रेडियोधर्मिता) या ionizing radiations कहते हैं I
Radioactive decay एक स्वतःप्रवर्तित प्रक्रिया (spontaneous process) है और इसकी दर अलग - अलग
isotopes क
े लिए अलग - अलग होती है I किसी radioactive material की half-life (t ½) उतना समय होता है
जिसमें वो material अपनी original value का आधा रह जाता है I Biological studies में उपयोग होने वाले क

Radioactive decay may occur in different ways:
1. Decay by negatron emission -
In this type of decay, a neutron is converted into a proton by ejection of a negatively charged
beta (β) particle called a negatron (β –ve)

2. Decay by alpha particle emission -
High atomic number वाले elements क
े isotopes सामान्यतः alpha (α) particle क
े emission द्वारा
decay होते हैं I इस emission क
े परिणामस्वरुप nucleus 2 protons और 2 neutrons खोता है जो कि एक
helium nucleus (containing 2 protons and 2 neutrons) क
े समान होता है I
Alpha emitters का are biological work में बहुत ही कम उपयोग होता है क्योंकि ये अपने अधिक mass और
ionizing power क
े कारण बेहद toxic होते हैं I
3.Decay by emission of gamma rays -
यह एक electromagnetic radiation है जो alpha और beta particles क
े emission होने क
े साथ हुए electron
excitation क
े कारण निकलते हैं I इनक
े निकलने से atomic number या mass में कोई परिवर्तन नहीं होता I
Gamma radiation की ionizing power कम होती है किन्तु penetrating power अत्यधिक होती है I Gamma
rays human body से पूरी तरह pass हो सकती हैं और जब ये शरीर से pass होती हैं तो ionization द्वारा
tissues और डीएनए को क्षति पहुँचती हैं I

4.Decay by electron capture -
इस प्रकार क
े decay में एक proton सबसे अंदरूनी k shell में परिक्रमा कर रहे electron को अधिकृ त (capture)
कर लेता है और यह proton एक neutron में परिवर्तित हो जाता है I तब “missing” electron एक बाहरी shell क
े
electron द्वारा प्रतिस्थापित होता है और x-ray उत्सर्जित होती है जिसकी energy दोनों electron shells की
energy क
े अंतर क
े बराबर होती है I x rays की energy gamma rays की energy से कम होती है और इसलिए
ये gamma rays क
े मुकाबले कम penetrating होती हैं I

Units Of Radioactivity
The radioactivity of a substance can be measured using different units.
● Becquerel is the SI unit for measurement of radioactivity. It is defined as the number of
disintegration per second (d.p.s.)

● Curie is the commonly used unit. It is defined as the quantity of radioactive material
having nuclear disintegration similar to that of 1g of radium i.e. 3.7 ×1010

dps or 1 Curie = 37 billion Bq
Milli Ci = Ci × 10- 3
Micro Ci = Ci × 10- 6
● Specific activity of a substance is defined as activity per unit weight or volume (e.g.,
Bq/gm or B/l).
● Counts per minute (c.p.m) is disintegration detected by a radiation counter.
Measurement of radioactivity -

े लिए सामान्यतः तीन विधियाँ इस्तेमाल होती हैं :
1. Ionisation of gases - इस विधि में जब radiation air या inert gas से भरे एक detector क
े अंदर pass
होता है तो यह gas atoms का ionization करता है जिससे atoms positive ions और electrons में अलग -
अलग हो जाते हैं I ये अलग हुए electrons और positive ions electrodes की ओर आकर्षित होते है जिससे एक
current का बहाव होता है और यह electric signals में परिवर्तित हो जाता है जिसे radiation की मात्रा क
े रूप में
मापा जाता है I उदाहरण - Geiger Muller (GM) counter जिसका इस्तेमाल radioactive laboratory में हुए
contamination की जाँच, Qualitative analysis of radioactivity, Quick screening of gels &
chromatographic fractions for labeled components आदि क
े लिए किया जाता है I
2. Excitation of solids and liquids - इस विधि में जब radiation किसी scintillator क
े through pass होता
है तो scintillator molecules excite हो जाते हैं किन्तु वे जब वापस अपने original ground state पर आते हैं
तो light emit करते हैं जिसे एक photomultiplier द्वारा amplify करक
े electric signal में परिवर्तित करक

radiation को मापा जाता है I Scintillation counters दो प्रकार क
े होते हैं :
(i) Solid scintillation counter - जिसमें solid fluorescing substance (scintillator) का इस्तेमाल होता है,
जैसे - zinc sulfide for α radiation, anthracene for β and sodium azide for gamma radiation.
(ii) Liquid scintillation counter - जिसमें एक scintillation 'cocktail' का इस्तेमाल होता है और यह बनता है
एक solvent (जैसे - xylene या toluene) और एक या अधिक fluors (scintillator) से I इसका इस्तेमाल weak

Uses of radioisotopes in medicine -
● Thyroid uptake studies by 131
I are used to detect functional derangements of thyroid
gland. In hyperthyroidism there will be increased uptake and in hypothyroidism there will
be decreased uptake when 131
I is given intravenously.
