ABSTRACT- Systemic and Scientific investigation was carried out for identification, selection, characterization of popularly grown medicinal plants in and around Patna and their phytochemical analysis was carried out to assess their medicinal value potentialities. The traditional medicine involves the uses of different plant extracts or the phytochemical bioactive constituents, which provides the health application at an affordable cost. Secondary metabolites are responsible for medicinal activity of plants. Qualitative phytochemical analysis of these plants confirm the presence of various phytochemicals like saponins, terpenoids, steroids, anthocyanins, coumarins, fatty acids, tannins, leucoanthocyanins and emodins etc. The result suggest that the phytochemical properties for curing various ailments and possess potential antioxidant, which may leads to the isolation of new and novel secondary compounds for generation of new drugs. Knowledge of the phytochemical constituents of plants are desirable because such information will be of value for the synthesis of complex chemical substances. Out of 50 medicinal plants subjected to phytochemical screening 11 were found to be highly potential,18 moderately potential and 21 to be least potential on the basis of presence of phytochemicals in the leaf extract for secondary metabolites.

Lempuyang gajah (Zingiber zerumbet (L.) Smith), lempuyang pahit (Zingiber amaricans BL.), and
lempuyang wangi (Zingiber aromaticum Vahl.) are used as traditional medicine (jamu) in Indonesia. It is also
used for treatment of microbial infections, helps to increase appetite and stimulate digestion in chickens.
Information on their uses are available, but only limited in the scientific data on their bioactivity. The study was
conducted on the antibacterial effect of organic extracts of these plants with Mycoplasma gallisepticum as the
agent of chronic respiratory disease in chickens. Juice and extracts of fresh and dried rhizome are evaluated
through the disc diffusion assay and minimum inhibitory concentration. Oxytetracyclin (30 µg) are used as
standards. All extracts are individually exhibited as antibacterial activity against Mycoplasma gallisepticum (7
± 0.11 mm to 21 ± 0.86 mm). The minimum inhibitory concentration (MIC) determination of plants extracts are
ranged from 7.8 mg/ml to 31.2 mg/ml. The preliminary results suggested promising antibacterial properties of
wild ginger from Indonesia, and probably could be used in management of chronic respiratory disease in
chickens.

Pedalium murex showed broad spectrum antimicrobial activity
against three fungal strains Aspergillus niger, Aspergillus flavus,
Candida albicans and five bacterial strains Escherichia coli,
Staphylococus epidermis, Klebseilla pneumonia,Citrobactor
diverses, Enterococus faecalis.The Ethanolic extracts were tested
against selected test bacteria and fungi through disc diffusion assay
where amoxicillin was used as standard. The results showed that
alcoholic extract possess good antimicrobial activity against selected
test bacteria and fungi. The present results therefore offer a
scientific basis for traditional use of the various extract of Pedalium

Abstract: In this study Methanolic and chloroform leaf and root extract of Rauvolfia serpentina was studied for its antibacterial activity. Antibacterial activity of leaf and root extracts was assessed against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis and Klebsiella pneumonia by disc diffusion method. Methanolic extract of root was showed the maximum zone of inhibition for all test organisms than the leaf extract. According to observations of root extract of 50µl/ml concentration 15.4mm, 16.2mm, 12.3mm,10.1mm and 15.0mm zones of inhibition and for concentration of 100µl/ml 22.5mm, 23.1mm, 15.1mm, 18.0mm, 22.0mm zones of inhibition were formed against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis and Klebsiella pneumonia respectively. 50µl/ml concentration of leaf and root chloroform extracts showed no zone of inhibition against Staphylococcus aureus and Bacillus subtilis, maximum zone of inhibition was observed 15.0mm and 15.5mm against E. coli for leaf and root chloroform extract respectively. 100µl/ml concentration showed maximum zone of inhibition against all test organisms for both leaf and root extracts. All the bacteria were more susceptible to methanolic extracts than the chloroform extracts.

In the present work an attempt has been made to carry out screening for the preliminary antibacterial activity of different plants used by Sugali tribes of Yerramalis forest. Fifteen plants were selected for preliminary screening for their antibacterial potentiality, The antibacterial activity was done against four bacterial strains, viz., Bacillus cereus, Staphylococcus aureus, Klebsiella pneumoniae, Escherichia coli,.The preliminary screening experiment revealed that methanol extracts were more potent than the aqueous extracts. The most susceptible bacteria were K. pneumoniae and the most resistant bacteria were E. coli. Bauhinia racemosa L. exhibited remarkable antibacterial activity.

The use of plants in treatment of burns, dermatophytes and infectious diseases is common in traditional medicine. The development of new antimicrobial agents against resistant pathogens is increasing interest. Therefore, the methanolic extracts from different parts of four medicinal plants used locally in folk medicine were evaluated for antimicrobial activity. It was found that most plant extracts studied had antibacterial and antifungal activities. The methanolic extract of leaf of the plant Azadiracta indica, Acacia nilotica and Witania somnifera showed significant antibacterial activity against Bacillus subtilis, Escherchia coli, stphaylocuccus aureus and pseudomonas fluorescence. Azadiracta indica and A.tinolica showed significant antifungal activity against A. flavus, Ziziphus mauritiana. The rhizome extract of curcuma longa showed significant activity against all tested bacteria and showed higher anti fungal activity against Fusarium verticillioides

Medicinal plants are of great importance to the health of individuals and communities. The medicinal value of these plants lies in some chemical substances that produce a definite physiological action on the human body. The objective of the present study was to evaluate the phytochemical constitution and antioxidant activity of Aqueous extracts of three selected plant Boerhaavia diffusa, Euphorbia hirta and Amaranthus polygonoides. Preliminary phytochemical screening revealed the presence of phytochemicals like alkaloids, flavonoids, Steroids, phenols, tannin and carbohydrates in Boerhaavia diffusa and Euphorbia hirta where as in Amaranthus polygon ides many phytoconstituents like alkaloids, flavonoids, Steroids, terpenoids, phenols, saponin, tannin and carbohydrates were present. Antioxidants are the compounds which terminate the attack of reactive species and reduce the risk of diseases. The free radicals oxidants are species with very short half life, high reactivity and damaging activity towards macromolecules like proteins, DNA and lipids. The results of antioxidant activity of three aqueous extract showed maximum activity in different concentration of 50, 250, 500, 750 and 1000 µg ml. The percent inhibition of aqueous extract of Boerhavia diffusa , Euphorbia hirta , Amaranthus polygonoides was 176.15, 404.78 and 413.06 respectively. In the present work potent anti oxidant activity of aqueous extract of Boerhaavia diffusa was higher when compared to other two extracts. The present study revealed that the plant extract possessed good antioxidant activity and less quantity of toxic metals, which therefore can be used as a natural source of free radical scavenger. Attenuation of Pseudomonas aeruginosa Virulence by Some Indonesian Medicinal ...UniversitasGadjahMada
This study aims to discover quorum sensing inhibitors (QSI) from some Indonesian medicinal plants ethanol extract to analyze their inhibitory activities against QS-mediated virulence factors in P. aeruginosa using in-vitro experimental study-laboratory setting. Indonesian medicinal plant ethanolic extracts were tested for their capability to inhibit P. aeruginosa motility, biofilm formation using microtiter plate method, pyocyanin and LasA production using LasA staphylolytic assay. Statistical significance of the data were determined using one way ANOVA, followed by Dunnett’s test. Differences were considered significant with P values of 0.05 or less. The findings obtained showed that Ethanolic extract of T. catappa leaves and A. alitilis flower capable to inhibit P. aeruginosa motility as well as pyocyanin production and biofilm formation. Both extracts also showed capability in reducing LasA protease production. It is concluded that T. catappa and A. alitilis are an interesting sources of innovative plant derived quorum quenching compound(s), thus can be used in the development of new antipathogenic drug.

Antimicrobial activity of methanolic extract of leaves and flowers of Bauhinia x Blakeana was resolved against Gram-positive and Gram-negative microscopic organisms. Streptomycin was utilized as reference principles for examination of action. It was discovered that fluid concentrate has antimicrobial activity against Escherichia coli and Lactobacillus leaves have good activity than flowers.

Attenuation of Pseudomonas aeruginosa Virulence by Some Indonesian Medicinal ...UniversitasGadjahMada
This study aims to discover quorum sensing inhibitors (QSI) from some Indonesian medicinal plants ethanol extract to analyze their inhibitory activities against QS-mediated virulence factors in P. aeruginosa using in-vitro experimental study-laboratory setting. Indonesian medicinal plant ethanolic extracts were tested for their capability to inhibit P. aeruginosa motility, biofilm formation using microtiter plate method, pyocyanin and LasA production using LasA staphylolytic assay. Statistical significance of the data were determined using one way ANOVA, followed by Dunnett’s test. Differences were considered significant with P values of 0.05 or less. The findings obtained showed that Ethanolic extract of T. catappa leaves and A. alitilis flower capable to inhibit P. aeruginosa motility as well as pyocyanin production and biofilm formation. Both extracts also showed capability in reducing LasA protease production. It is concluded that T. catappa and A. alitilis are an interesting sources of innovative plant derived quorum quenching compound(s), thus can be used in the development of new antipathogenic drug.

Resistant or multi resistant strains are continuously appearing in the treatment of ailments and this necessitates the synthesis of new drugs especially from naturally occurring plants. Momordica charantia is a very common indigenous plant of tropical and sub-tropical regions of the world. Phytochemical screening, antimicrobial analysis were conducted using recommended procedures. Ethanol and water were used for the extraction and Ultraviolet Visible and Infrared Spectroscope were used to identify the functional groups. Alkaloids, flavonoids, saponins, tannins, anthraquinones were all present in both the ethanolic and aqueous extracts, terpenoids was only present in aqueous extract while cardiac glycosides was not present in both extracts. Concentration (200mg/ml) revealed the highest clear zone of inhibition in both aqueous and ethanol extracts. The zone of inhibition increases with increase in concentration of sample. The ethanol extract shows the higher clear zone of inhibition when compared to aqueous extract with a diameter of zones of inhibition of 28, 26, 24, 26, 26 and 24mm for Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginesa, Salmonella typhi and Klebsiella pneumoniae respectively. The aqueous extract has no effect at concentration (6.25mg/ml) because no clear zone of inhibition was measured. The antimicrobial activity for aqueous extract was lower than and not as effective as ethanol extract because ethanol is a phenolic compound and non-polar solvent when compared with water that is a polar solvent. Infrared spectra showed OH stretch due to phenol was observed at 3702 cm-1 , - C - H stretch (alkane) at 2972cm-1 , C - H bending vibration (alkane) at 1381cm-1 , - C - N and – C = N at 2077cm-1 , NH stretching at 3375cm-1 . The results indicate that it contained a phenolic, amine and amide substitute. Therapeutic drugs with antibiotic and antifungal activities can be synthesized from Mormodica charantia.

Anti Microbial Activities and Phytochemical Screening of the Premna Odorata B...ijtsrd
The researcher focused mainly to determine the antimicrobial properties and phytochemical screening of the Alagaw leaf extract. Specifically, this study was conducted to determine the percent yield, antimicrobial activities and the secondary metabolites of Alagaw leaf extract which was analyzed and it include alkaloid, anthraquinone, saponins and steroid. Findings of the study showed that the Alagaw leaf extract has a percent yield of 11.5 . Anti Microbial Activity was tested by petri disk on a plate Nutrient Agar streaked with the E. coli bacteria, the plates were incubated for 24hrs and 37oC. Results were observed for the presence of zone inhibition clear area around the test disk. Results from this study showed that the antimicrobial activity on E. coli as indicated as negative by the presence of Alagaw leaf extract. Furthermore, the result suggests that the Alagaw leaf extract did not suppress the growth of the E. coli bacteria, hence it indicates that it has no anti microbial effect to the test organism. While the secondary metabolites such as alkaloid, anthraquinone, saponins and steroid is found negative. Effect of Nitrobenzene granules and Seaweed extracts on biochemical contents ...Agriculture Journal IJOEAR
The present study is aimed to evaluate the effect of organic extracts (benzene, diethyl ether and water) of seaweeds (Halimeda gracilis, Ceramium rubrum and Cystophyllum muricatum) and nitrobenzene granuleson biochemical contents of Arachishypogea L. callusunder in vitro conditions. The callus of Arachishypogea L. was obtained from the leaf explants on MS medium containing 2, 4-D (1 mgL-1) and BAP (0.5 mg L-1). The mass multiplication of callus was achieved at 1mg L-1 of 2, 4-D and 0.5 mg L-1 of GA3. The calli were then treated with different concentrations (0.5, 1.0 and 1.5 mg L-1) of seaweed extracts and Nitrobenzene granules. Total carbohydrate, total protein and total chlorophyll contents were analyzed at 5, 10 and 15 days intervals. The total carbohydrate content was high (3.7mg/100mg) in callus treated with Benzene extract of Ceramium rubrumat 1.5 mg L-1 on 15th day. The total protein content was increased (6.9mg/100 mg) in callus treated with Benzene extract of Cystophyllum muricatum at 0.5 mg L-1 on 5th day and the total chlorophyll content was lower (0.36mg/100mg) in Nitrobenzene granules at 0.5mg L-1 in 5th day when compare to control. The present study reveals the positive role of different extracts of seaweeds on increasing the biochemical contents of callus culture of A.hypogea. The extracts can be further evaluated for their role on enhanced regeneration of plants from callus culture.

Its stem bark extracts obtained via soxhlet extraction using n-hexane, acetone and ethanol gare alkaloids, Steroids, Saponins, Cardiac glycosides, triterpenes and tannins when screened phytochemically. These extracts were further investigated for their anti-bacterial activity against staphylococcus aureus klebsiella, pneumoniae and pseudomonas aeruginosa with ampicillina, as standard. Highest anti- bacterial activity with acetone extract at a concentration of 300mg/ml and least activity with ethanol extract at a concentration of 100mg/ml. n- Hexane extract showed no inhibition on the tested micro-organisms.

The current study was carried out in District Abbottabad aimed to determine the common urinary
tract infections in local community to determine the epidemiology of significant diseases in asymptomatic patients
of renal disorder. In this study a total of 1000 urine samples were examined during 3rd February to 1st April 2015
from patients attending Ayub Teaching Hospital Abbottabad by using dipstick and microscopic analysis of urine.
There were 638 females and 362 males patients examined during this period. The range of age groups is between
1.5 years to 80 years. Results of this study was reported as Pyuria 11%, Proteinuria 21.1%, Hematuria 10.4%,
Epithelial Cells 8.2%, pH 7.8 %, Granular casts 7.3%, Triple phosphate 6.6%, Calcium oxalate 6.4%, Glycosuria
6.3%, Bacteria 6.2% and mucous 4.1%. This study concludes that routing urinalysis should be performed for all
individuals to diagnose the asymptomatic diseases that will help in simple therapeutic measurements as urinalysis
is a simple step to determine the root of Urinary tract disorders.

MATERIALS AND METHODS -This was a single institute study approved by the institutional ethics
committee. Prior informed consent was obtained from the parents. 100 consecutive patients admitted in
N.I.C.U. WITH gestational age < 36 weeks with respiratory complaints were enrolled. Chest x-ray was
obtained within few hours of admission and lung sonography was performed within 24 hours. Follow – up
sonography was performed as and when necessary. Sonography image was graded and correlated with chest

self-medication practices; to identify sources of medications used; to identify sources of information about

Aim: To study the clinico-haematological profile malaria in a rural hospital of Tripura.
Material and methods: A cross-sectional hospital-based study was done from at Kulai District
Hospital,Tripura. This hospital based cross sectional study was done on 60 confirmed cases of falciparum
malaria (either by peripheral smear or rapid diagnostic test) admitted in Kulai District Hospital. A case sheet
proforma was prepared and data (demographic profile,clinical feature, investigation, treatment, and
complication) from all indoor patients was collected and analyzed.
Result: Out of 60 patients, 40(66.6%) were males and 20 (33.4%) were females. Most of the patients were
between the age group 21-40 years with the highest prevalence between the age group of 21-30. Fever was the
most common symptom. Anemia was present in 42(70%) patients, out of which 6(10%) patients had severe
anemia. Thrombocytopenia was present in 36(60%) patients.Abnormal liver function tests were observed in
26(43.3%) subjects while abnormal kidney function tests were observed in16(26.6%) patients. All the 60
patients received Artemisinin based antimalarial drugs.
Conclusion: Early detection, prompt management, and adequate supportive therapy may reduce mortality due
to falciparum cerebral malaria.

The eggplant (Solanum melongena L.) is known to cause food allergies in some Asian countries, but detailed
studies on allergies caused by eggplant are lacking, however, it was highlighted the presence of allergens in
edible parts of eggplant with preponderance in the peel .
The purpose of this study was to propose an extraction method rapid, efficient and cost of natural dye from
waste products from the food industry, such as the peels of eggplant, from which it was extracted, isolated and
purified the nasunin,a colored molecule in red-fuchsia.
Nasusin was tested on 58 patients to evaluate the potential sensitizing effect on the skin. The results demonstrate
that allergenic effects are negligible and therefore the nasunin can be used as a colorant in various industrial
sectors with a certain safety margin

Patients and methods: A total of 94 pediatric patients with B-cell ALL, of whom 70 were the studied population
and 24 were the test population, were treated with four courses of HDMTX doses 2.5 gm/m2
(low-risk arm) or 5

(standard-/high-risk arm) given every other week by intermittent intravenous infusions over 24 hours as

Abstract : In Albania, many people erroneously think that tuberculosis (TB) is a disease of the past-an illness
that no longer constitutes a public health threat. Surveillance is an integral part of tuberculosis (TB) control.
Albania has a highTB notification rate and there are doubts about underreporting. The evolution of the
incidence of tuberculosis is presented, together with more detailed figures over the period 1998-2009. These
figures were obtained by the monthly forms (called 14/Sh) compared with the individual notification data.
Objective: To examine the distribution and sources of increased tuberculosis (TB) morbidity and reporting
system deficiencies in the Albania from 1998 through 2009. Metodology: The study is descriptive one conductet
during the period 1998-2009. The statistical analysis is based on data reported from regional level (regional
epidemiological departments) to the central level (Public Health Institute). Results: The main findings were:
discordance between the collected data (individual form) and reported data (monthly form); tuberculosis
incidence rate shows little oscillations which ranges from 6.67 to 9.2 cases/100.000 population; 50% of the
regions show a lack of information on the confirmation of diagnosis and laboratory examination type used for
confirmation. Conclusion: TB disease in high-risk populations where it is difficult to detect, diagnose, and treat;
limitations of current control measures and the need for new tests and treatments, including an effective
vaccine; improving information system, regulation of individual form and personnel training.
Total Phenol and Antioxidant from Seed and Peel of Ripe and Unripe of Indones...iosrphr_editor
Study on total phenol and antioxidantactivity ofsugar apple fruits of various solvent, part of fruits, and level of ripening. Solvent extraction used were 80% (v/v) methanol, 50% (v/v) acetone, boiling water, and 50% (v/v) ethanol. Part of fruits thatbeen used for samples were seed and peel which are normally by products of sugar apple processing, level of ripening were unripe, and ripe sugar apple fruits. Total phenol was determined by Folin-ciocalteau method. Total antioxidant was quantified by 1,1-diphenyl-2-picrylhydrazyl(DPPH) method.Therewas a difference in type of solvent, part of fruits, and level of ripeningon total phenol and antioxidant concentration of sugar apple fruits. Seeds have higher total phenol concentration than peels of this fruits. Unripe sugar apple fruits have higher total phenol and antioxidant than ripe fruit. The best solvent for phenol extraction was ethanol 50%butthe best solvent for antioxidant extraction was acetone 50%.

To comply with the requirements of GMP pharmaceutical industries should have an overall validation policy which documents how validation will be performed. The purpose of this validation is to show that processes involved in the development and manufacture of drug, production and analytical testing can be performed in an effective and reproducible manner. This review article provides guidance on how to perform validation characteristics for the analytical method which are utilized in pharmaceutical analysis.
A Cross Sectional Study of Ethnic Differences in Occurrence and Severity of A...iosrphr_editor
Non-steroidal anti-inflammatory drugs are the most widely used "over the counter" medication all over the world despite their complications in different major organs. Present studies envisaged for knowing the occurrence and severity of adverse drug reactions from NSAIDs in different ethnic communities of Sikkim. A cross sectional study was undertaken in the medicine outpatients department of a secondary and tertiary care hospital. The patients belonging to Nepalese, Bhutias, Lepchas ethnic communities and others community (settlers from other parts of India) were included to analyzed the data based on the age and gender, ethnicity and ADRs, drugs and ADRs. Severity assessment was done using Hartwing and Siegel scale and causality assessment by Naranjo scale. Total 109 cases of ADRs, predominating in female were detected. Nepalese were the most affected and Gastrointestinal tract (GIT) being the most affected organ in them. Diclofenac showed maximum number of ADRs in all the communities. Maximum number of cases occurred on single day use (40.36%) of drugs. All the cases were belonging to the "possible category" and the maximum being the mild (72.48%) in nature. It is advisable to consider the ethnic/racial differences equally with other factors, to improve the safety and efficacy of a drug.

Malaria is a major health problem in India with 1.04 million cases reported in 2012 leading to 504 deaths. The clinical spectrum depends on the infecting species, level of parasitemia and the immune status of the host. Malaria pathogenesis is based on extensive changes in hematological and biochemical parameters. The objective of this study was to study the clinical features, hematological and biochemical parameters in malaria patients and correlate them with the parasitic index (PI). Material and methods: We conducted a study on 300 malaria patients. The frequency of various symptoms and signs of malaria caused by various plasmodium species were determined. The degree of anemia, WBC count, platelet count serum bilirubin, liver enzymes and serum creatinine levels were studied and their variation depending on the parasitic index was documented. Results: 197 patients had vivax malaria, 76 patients had falciparum malaria and 27 patients had mixed infection. 171 patients had a PI of less than 2%, 100 patients had PI between 2 to 5%, 23 patients had PI between 5 to 10% and only 6 patients had PI of more than 10%. 72.3% of patients had thrombocytopenia, 46.66% had anaemia, 25% had increased bilirubin 29.66% showed increased liver enzymes and 7.66% had increased creatinine levels. Conclusion: There was a correlation between degree of parasitemia and severity of malaria in majority of cases. Derangements in hematological and biochemical parameters were more frequently seen in patients with higher PI. Hence PI can be used as an indicator by the clinician to know the severity of infection and plan appropriate treatment.

ABSTRACT: The root and latex of Calotropis gigantea were screened for its antimicrobial and
phytochemical activities. The solvents used for the roots and latex extraction were n-hexane, benzene, acetone,
ethanol, aqueous. The extract was tested against infectious disease causing bacterial such as Escherichia coli,
Pseudomonas aeruginosa, Staphylococcus aureus using the well diffusion method. The aqueous, ethanolic and
acetone extract of root of calotropis gigantea inhibition against all the test microbe ranging from 10 mm to 16
mm diameter inhibitory zone. The aqueous and ethanolic extract of latex of calotropis gigantea inhibition
against all the test microbe ranging from 10 mm to 18 mm diameter inhibitory zone. In present study, bacterial
extract showed a varying zone of inhibition of the growth of tested organism than n-hexane, benzene, ethanol,
and aqueous. Phytochemical properties of root and latex of Calotropis gigantea obtain from n- hexane,
benzene, acetone, ethanol and aqueous extracts were investigated. The results confirmed that presence of
antibacterial activity and phytochemical in the shade dried extract of Calotropis gigantea against the human
pathogenic organisms.

I. INTRODUCTION
A medicinal plant is any plant which, in one or more of it contains substance that can be used for
therapeutic purpose or which is a precursor for synthesis of useful drugs. The plants posses therapeutic
properties or exert Beneficial Pharmacological effects on the animal body are generally designated as
“Medicinal Plant”. In current scenario of medical and pharmaceutical advancement, microbes involve in the
change of their metabolism and genetic structure to acquire resistant against the drugs used in the treatment of
common infection disease. (1), (2)
The continued emergence or persistence of drug resistant organisms and the
increasing evolutionary adaptation by pathogenic organisms to commonly used antimicrobials have reduced the
efficacy of antimicrobial agent currently in use. (3)
Plant have the capacity to produce a large number of organic
chemicals called as phytochemicals. The accumulation of phytochemicals in the plant cell cultures had been
studied for than thirty years and the generated knowledge had helped in realization of using cell culture for the
production of desired phytochemicals. (4)
Calotropis belong to Asclepidaceae family. It is also known as Akada, Aak, Mandar, Aakh etc. It has
two species procera and gigantea .here we study about Calotropis Gigantea. (5)
The roots and leaves of
calotropis gigantea are used traditionally for treatment of abdominal, tumours boils, skin diseases, wound,
insect bites. A literature review showed that Calotropis Gigantea contained cardenolide, glucosides, a non
protein, amino acid, flavonoids and steroids. Calotropis gigantea in small dose are also useful in the treatment
of cold, cough, asthma inflammatory diseases and loss of digestive and analgesic property of Calotropis
Gigantea.(6) ,(7)
Milky sap is used in the treatment of boils, scabies, burns, bruises, cuts, sores, stopping blood,
and wound healing; Leaves are used in chest congestion and cardiovascular conditions. The roots and barks of
C. gigantea are in use for paralysis, fits, epilepsy, and convulsions in children. Shoot, leaf, roots flowers and
latex extracts are reported to have antibacterial and antifungal properties by researchers. (8)
This study amied to
find out the phytochemical content and its antimicrobial activity of root and latex extracts of Calotropis
gigantea.
II. MATERIALS AND METHODS COLLECTION OF PLANT MATERIAL:
Collection of plant material the leaves and fruits of Calotropis gigantea also known as crown flower;
was done from the areas around Bhabra of District Alirajpur, Madhya Pradesh. The identification of the whole
plant and its parts were done by Dr. Kishor Panwar department of Botany, Holkar Science College, Indore,
Madhya Pradesh.

III. EXTRACTION OF PLANT
Latex is a complex emulsion of polymer micro particles in an aqueous medium. The extractions from
latex are quit tricky in context with preparation of latex for the extraction purpose. For this, the latex was
collected from the injured tissues of plant C. gigantea, by making an incision on the stem part. The obtained
latex was then centrifuged at 5000 RPM in the laboratory and the supernatant aqueous phase was decanted and
the sticky rubber like matter is pulled out of centrifuge tube and properly dried for 2-3 days. The matter is then
processed into smaller particles in mortar pestle. The rest process for extraction is similar to those the extraction
for leaves, roots and fruits. 10 grams of dried latex was subjected to Soxhlet extraction with 200 ml of solvents
starting from pure distilled water, followed by extraction with other solvents Ethanol, Acetone n-Hexane and
Benzene in separate ways.
The twigs of the roots of Calotropis gigantea were washed and allowed to dry in shade for a week
and then grounded into fine powder in mixer grinder. Similar as the extraction for leaves, 10 grams of dried
powder of roots was subjected to Soxhlet extraction with 200 ml of solvents starting from n-hexane, followed by
extraction with other solvents Benzene and Acetone Ethanol and pure distilled water in separate ways.
Soxhlet process was allowed to carry out till the complete exhaustion of sample material use for extraction
with the maintenance of temperature below the boiling points of the solvents used. The extract so obtained is
subjected to evaporation of solvent to get the extract in crystalline/slurry form which were suitably diluted and
used for preliminary phytochemical analysis and studies of their antimicrobial activity.
IV. PHYTOCHEMICAL ANALYSIS OF THE EXTRACT:
A small portion of the dry extracts were subjected to the phytochemical test using Harbourne’s (1983)
methods to test for alkaloids, tannins, terpenoids, saponins, flavonoids and glycosides.
Test for alkaloids: About 0.2 g extract warmed with 2% H2SO4 for two minutes, filtered and few drops of
Dragendorff’s reagent added orange red precipitate indicates the presence of alkaloids. And or filtrates were
treated with Wagner’s reagent (Iodine in Potassium Iodide). Formation of brown/reddish precipitate indicates
the presence of alkaloids.
Test for glycosides: The extracts hydrolyzed with HCl solutions and neutralized with NaOH solutions. A few
drops of Fehling solution A and B were added. Red precipitate indicates the presence of glycoside. Another test
use was Benedict’s test, in which the filtrates were treated with Benedict’s reagent and heated gently. Orange
red precipitate indicates the presence of reducing sugars.
Test for tannins: Small quantity of extracts mixed with water, heated, filtered and ferric chloride added. A dark
green solution indicates the presence of tannins.
Test for saponins: About 0.2g of the extracts shaken with 5ml of distilled water and then heated to boil frothing
(appearance of creamy mix of small bubbles) shows the presence of saponins.
Test for flavonoids: Extract of about 0.2 g of the extracts shaken with 5ml of distilled water and then a few
drops of 10% lead acetate solution is added. A yellow or dirty white precipitate shows the presence of
flavonoids. (9)
V. CULTURE MEDIA AND INOCULUM PREPARATION:
Nutrient agar broth cultures of the pure culture isolates of Staphylococcus aureus, E. coli and
Pseudomonas aeruginosa were prepared by transferring a loop of culture into sterile nutrient broth and
incubated at 37o
C for 48hours. A loop full was taken from these broths and seeded onto sterile nutrient agar
plates through sterile cotton swab to develop diffused heavy lawn culture.
VI. ANTIMICROBIAL ACTIVITY:
The well diffusion method was used to determine the antibacterial activity of the extracts prepared
from the Calotropis gigantea roots and latex using standard procedure. In this method, first the test bacteria
broth of bacteria are used to inoculate on the nutrient agar plates with the help of sterile cotton swabs to develop
the lawn culture. Then to these plates 6 mm diameter well are punched in agar plates pre-inoculated with test
microorganisms Undiluted over night broth cultures should never be used as an inoculam .routine direct
application of suitably diluted extracts are poured into the well. The plates were incubated at 37o
C for 24 hr. and
then examined for clear zones of inhibition. Sterile water was used as control. (10)

RESULTS AND DISCUSSION:
PHYTOCHEMICAL ANALYSIS OF BIOACTIVE COMPOUND IN DIFFERENT SOLVENT
EXTRACTS OF CALOTROPIS GIGANTEA
The plant root extracts in different solvent were screened for the presence of various bioactive
phytochemical compounds. The aqueous roots extracts of Calotropis gigantea were found highly rich in
alkaloids and glycosides, although terpenoids are totally absent in them. Saponins are the only which are present
in all the root extract under evaluation. The n-hexane extracts were poor in their phytochemical contains.
The plant latex extract in different solvent were screened for the presence of various bioactive phytochemical
compounds the acetone and ethanolic latex extract of Calotropis gigantea were found rich in alkaloids,
glycosides, tannins and saponins . Saponins are the only which are present in all the letax extract under
evaluation. The n-hexane and benzene extracts were poor in their phytochemical contains. This were
documented in Table 1&2.

ANTIMICROBIAL ACTIVITY OF DIFFERENT ORGANIC SOLVENT EXTRACTS OF
CALOTROPIS GIGANTEA
Antimicrobial activity of different solvent extract of calotropis gigantea is shown in table that the
aqueous, ethanolic and acetonic extracts of Root of C. gigantea impart sufficient inhibitory actions against the
test microbe ranging from 10 mm to 16 mm diameter inhibitory zones. And out of the hexane and benzene
extracts only limited inhibition was observe in benzene extract against the E. coli only.
The ethanolic and aqueous extracts of latex of C. gigantea impart sufficient inhibitory actions against
the test microbe ranging from 10 mm to 18 mm diameter inhibitory zones. The aqueous extract of latex has
maximum zone of inhibition against the Staphylococcus aureus the common Gram positive pathogenic
microorganism and this is the maximum inhibitory potential out of the all extracts viz. root and latex. But the
acetonic, hexane and benzene extracts of latex has no inhibition against any organism. The result of antibacterial
activity are shown in table 3 & 4 and figure 1 & 2

Antibactrial activity of different solvent extacts of C. gigantea showed vayring degrees of antibacterial activity
against all microorganisms tested.(11)
There are many reports of plants in the family Asclepiadaceae possessing anti-
microbial activity. (12, 13)
From this study it can be said that, actone ethanol and aqueous shade dried root extract and ethanol
and aqueous shade dried latex extract of C. gigantea showed wide range of antibacterial activity can be used and
administered in the ethano medical practice. (14, 15)
The results obtained from our study showed an effective inhibition against
the test organism which justify the traditional use of the plant for infection diseases.

VII. CONCLUSION:
The encouraging results indicate that the aqueous, ethanolic, acetonic, n-hexane and benzene extracts
of various parts of Calotropis gigantea might be exploited as a natural drug for the treatment of several
infectious diseases caused by these organisms and could be useful in understanding the relations between
traditional cures and current medications. Our results showed that in present work that extracts obtained
different parts of the plant Calotropis gigantea using various solvents are rich sources of potent phytochemicals
especially the roots and latex extract and has inhibitory effects on the experimental microbes. From previous
studies and the current work it is clear that the plant is rich source of alkaloids, glycoside, tannins, saponins,
flavonoids, terpenoids etc. These bioactive complex phytochemicals can be used for the development of potent
drugs, medicines or antimicrobial agents that can be used for various purposes for human welfare upon further
extensive & systematic studies.
