This document describes the method for determining the unknown concentration of histamine using guinea pig ileum. Guinea pig ileum is highly sensitive to histamine due to the presence of H1 receptors. The assay involves setting up guinea pig ileum in an organ bath with tyrode solution and measuring the contractile response to additions of histamine of known and unknown concentrations. The responses are used to construct a dose-response curve and calculate the concentration of the unknown histamine sample. Proper experimental controls and techniques are required to obtain accurate and reproducible results from this bioassay method.
