2. Terminologies in Chromatography
1) Eluent/ eluent
Origin: Latin ēluēns, ēluent-, present participle of ēluere, to wash out
The liquid used to extract one material from another in chromatography is called eluent.
It is the "carrier" portion of the mobile phase which moves the analytes through the
chromatograph.
Note: The eluent in liquid chromatography is a liquid solvent whereas ingas chromatography is
a carrier gas.
2) Eluate
Origin: Latin ēluere to wash out; see elute. -ate
Eluate is a solution of solvent and dissolved matter resulting from elution.
The eluate is the analyte material that emerges from the chromatograph.
Note: It specifically includes both the analytes and solutes which are passed through the
column.
3) Elution
Origin: elute » from Latin ēluere, ēlūt-, to wash out: ē-, ex-, ex- + -luere, to wash.
The process of extracting a substance which is adsorbed to another by washing it with a
solvent.
A chromatography method where the stationary bed is within a tube and the mobile phase
moves continuously through the column and the analyte is detected as it emerges from
the column (Royal Society of Chemistry)
Example
Working with column chromatography to separate out amino acids (arginine, phenylalanine,
tyrosine, tryptophan) from the mixture using BAW (Butan-1-ol, acetic acid, and sterile water)
as solvent. In this example BAW solvent is eluent, amino acids which are separated are eluate
and this process of separation is elution.
4) Difference between Chromatogram and Chromatograph
Chromatogram Chromatograph
The visual output from a chromatograph.
Usually a graphical display or histogram
The chromatograph is the instrument that is
used to generate the chromatogram.
