discussions, policies, practices or goals of the organization" (Freeman, 1984: 25) In the case of
Nasty Gal, their stakeholders comprise of a number of publics that they interact with and are linked
with (Esman, 1972; Grunig and Hunt, 1984:141) including current/potential customers, staff
members, investors/stockholders, women, media and political groups. According to the boohoo
website, Nasty Gal's target customer demographic is 18–27 year olds. (Boohoo Group, 2017) The
products are niche and designed for a specific demographic and young girls want to be a part of this
community.
Stakeholder Approach In its rapid growth, Nasty Gal placed an extreme focus on the needs of the
shareholders to the detriment of its stakeholders. The boohoo group is potentially making the same
mistake as by taking on the task of reviving the brand's reputation it places pressure on them to
communicate positive projected financial returns whilst simultaneous managing the expectations of
Nasty Gal's existing customer base. As a result, the company has taken the soft approach in
communicating with the existing customers of the brand by not addressing the pre–existing issues

could also look into beverage choices. While food is often looked into for water footprints,
beverages can also have a significant impact. If each member of the dinner party consumes one
serving of wine and one serving of beer, each person is adding 52 gallons for a total of 520 gallons
of water for those servings of beverages alone. Instead of looking to alcoholic or other high water–
footprint liquids, water itself could be consumed or an alternative such as tea where one serving is

The discovery of transformation by Frederick Griffith in Streptococcus pneumonia has played an
important role in how we are now able to introduce plasmid DNA molecules into cells.
Transformation is the uptake of DNA molecules released from the donor cell by the recipient cell. It
is one of the three ways bacteria are able to exchange genetic material. In Griffith's experiment he
introduced mice to two different forms of S. pneumonia, one smooth, pathogenic and encapsulated
and the other rough, nonpathogenic and noncapsulated (Snustad, 193). The mice were injected with
live rough strain and heat killed smooth strain. The deaths of the mice lead Griffith to conclude that
some genes of the killed smooth strain were transformed to the rough strain and the bacteria became
encapsulated and pathogenic, therefore leading to the death of the mice (Snustad, 193).
Plasmids are small circular DNA molecules. They are not essential for survival of the host bacteria.
Some carry genes that allow resistance to antibiotics (Anderson). Plasmid pUC18 is a circular DNA
molecule. It contains portions of the E. coli Lac Z gene, which encodes for the first 146 amino acids
of β – galactosidase. E. coli contains the Lac Z gene, which encodes β – galactosidase . The E. coli
Lac operon digests lactose. Once E. coli is transformed with pUC18, complementation occurs. E.
coli produces active β –galactoidase. The active β –galactoidase hydrolyzes the substrate, X gal ,

The type of abuses known to children are: child sexual abuse, physical child abuse, emotional child
abuse, neglect of a child, domestic violence, and shaken baby syndrome We know that of late, child
trafficking, which is the exploitation of children under 18 years of age is getting worse. "The US
Department of Justice has supported CASA advocacy since 1985 through its Office of Juvenile
Justice and Delinquency Prevention (OJJDP), it has changed the lives of many"

A wildtype and lacZ– gene were used. This experiment involved checking different time point for
the different types of strains. For the wildtype graph, as the time point increased the ONP produced
decreased and also the beta galactosidase activity decreased. For the mutant lacZ gene when the
time point increased the enzyme activity and the ONP produced also increased but it got to a time
point of 135 minutes where there was no ONP produced, this indicted that there was no enzyme
activity. This result indicates that after a long time the inducer IPTG that was being used to continue

IPTG added the higher the amount of ONP produced this indicate there was also higher activity of
the beta galactosidase enzyme. This indicate that IPTG is a good gene expression inducer, also the
untested solution had the lowers enzyme activity and less ONP produced. When no inducer added
just a repressor they were low enzyme activity, also when glucose was added, and no inducer little
expression is indicated. This indicates that a promoter responds negatively when there no inducer
and positively when an inducer is present. A lot of glucose cause a cAMP–CAP complex which
causes little expressions of the lacZ gene, therefore there is little ONP produced. For the X gal plate
strain 1 and 2 and 3 shows that beta galactosidase cleaved to the X gal because of the deep blue

CASA/GAL Volunteer Cases
Court Appointed Special Advocates and Guardian Ad Litem's are just regular people who are doing
their best to make a difference in the life of a child who is involved in the court system because they
have been declared by the court to be neglected, abused or dependent. The CASA/GAL volunteer is
required to go through extensive training before they are ever given a case to handle. They are
expected to commit to at least 15 hours a month per a case and to handle 1–2 cases at a time. The
cases can last up to two years and sometimes can last longer than that. In the beginning they may put
in more hours because of all the court hearings and meeting they need to attend. The Judge who is
overseeing the case is the one who appoints a CASA/GAL volunteer. There are not enough
CASA/GAL volunteers to handle all cases at this time so they are usually used in just the most
difficult cases. The CASA program was first started in Seattle in 1977 and now has over 75,000
volunteers. There are around 1,000 CASA programs at this time in 49 different states but not in

Engineering has become the most important and widely used technique, creating life saving
antibiotics and medicines. E.coli is generally used in these procedures because of its ability to adapt

Paul begins by defending his credibility as an apostle. He writes a brief autobiographical history,

formed. To begin, the student extracts a plasmid, which is a circular strand of DNA found within

of nitrogenous bases. The enzyme also cuts above and below the gene of interest and then ligase
bonds the plasmid to the gene of interest and you now have a strand of recombinant DNA. Such a
strand is useful because scientists can import useful genes into bacteria and these can be used to
create such substances such as insulin. This makes life much easier for diabetics who cannot
produce sufficient quantities on their own. On the opposing view, people may not accept this way of
producing proteins as natural due to the fact that the DNA was genetically engineered. Eventually

Leave the tubes on a tube rack at room temperature for 10 minutes.
12.Remove 100 µL of cells from the tube and spread them on the plates with their corresponding
plasmid. (i.e. tubes with +plasmid go to the +plasmid agar plates and vice versa.)
13. Let the plates rest for several minutes to allow the cell suspensions to become absorbed into the
agar.
14. Use a sterile pipette to add 100 µL of cell suspension from the +plasmid to each appropriate
plate, and spread.
15.Seal the plates with tape, and incubate upside down for 24–36 hours at 37 degrees Celsius.

Saint–Joseph de Cluny, the religious congregation that Javouhey founded with four of her biological

  • 79. A Safe Haven For Orphans After the loss of the majority of France's colonies in the Americas, the Restoration government occupied itself with colonizing attempts in Senegal and French Guiana. During this period of colonial expansion in French Guiana, the French government endorsed several attempts to colonize Mana, an area along the fertile, but poorly known Mana River. However, none of their attempts were successful. In 1827, the French government granted Anne–Marie Javouhey and the Sœurs de Saint–Joseph de Cluny, the religious congregation that Javouhey founded with four of her biological sisters, a tract of land for the establishment of an independent commune. Javouhey initially planned to use Mana to create a safe haven for orphans. Although she asked the French government to financially support the establishment, the colony was to remain independent in terms of governance and its economic livelihood. By 1832, before the orphans that she had envisioned could materialize, Javouhey's attempt had failed. According to Sarah Curtis, of the original fifty–six colonists in 1828, "five had died, eight had left for other parts of Guiana, and twenty–nine had returned to France." In 1836, Javouhey returned to Mana. This time, she found success working for the moralization and gradual emancipation of slaves.
