1. ESTIMATION OF NITRATE IN
DRINKING WATER

2. Why nitrate test Is needed?
Determine the presence of nitrate ion.
Identify nitrate - nitrogen concentration is acceptable-
10mg/L.
Excess nitrate –eutrophication.
High level of nitrate - over use of chemical fertilizers,
improper disposal.

3. EFFECTS IN HUMAN
 High of nitrate can be dangerous to health.
 Cause methemoglobinemia – blue baby syndrome.
 Carcinogenic effect- brain tumor, pancreas tumor.

4. REGENT USES
 Sodium chloride – catalyst
 Sulphuric acid – oxidize reaction
 Brusin sulphanilic acid – colour intensity measurement
 Potassium nitrate – strong oxidizing agent

5. PROTOCOL
Required number of test tubes
10ml of sample (if saline, add 2ml of 30% NaCl)
Cold water bath at 0-100 C
10ml of sulphuric acid to all test tubes and mix
0.5ml of Brusine sulphanilic acid

1000 C Water bath for 25 minutes
cold water bath – thermal equilibrium
Read at 410 nm

Graph plotted.
Absorbance vs Nitrate concentration.
