Estimating Nitrate Levels in Drinking Water
1. ESTIMATION OF NITRATE IN
DRINKING WATER
Submitted by
R. Anbarasan,
2nd M.Sc Biotechnology,
Annamalai University.
2. Why nitrate test Is needed?
Determine the presence of nitrate ion.
Identify nitrate - nitrogen concentration is acceptable-
10mg/L.
Excess nitrate –eutrophication.
High level of nitrate - over use of chemical fertilizers,
improper disposal.
3. EFFECTS IN HUMAN
High of nitrate can be dangerous to health.
Cause methemoglobinemia – blue baby syndrome.
Carcinogenic effect- brain tumor, pancreas tumor.
5. PROTOCOL
Required number of test tubes
10ml of sample (if saline, add 2ml of 30% NaCl)
Cold water bath at 0-100 C
10ml of sulphuric acid to all test tubes and mix
0.5ml of Brusine sulphanilic acid
6. Cont…..
1000 C Water bath for 25 minutes
cold water bath – thermal equilibrium
