• It has become staple food in many parts of world.
4. TISSUE CULTURE PROPAGATION OF
POATATO.
Plants can be propagates in vitro by two different methods
single node citting
shaken liquid culture
6. PROCEDURE.
• operation carried out under aseptic conditions.
• quickly flame the mouth of tube and carefully remove plantlet using a pair of
forceps.
• in petri dosh carefully remove leaves drom stem and cut stem into single node
segment.
• transfer node onto surface of fresh propagation medium.
• don’t damage single node cutting with the point of the sealpal
• single node cutting begin to grow rapidly into new rooted plantles.

• the tubes are incubated in growth root to allow plantlets to grow.
• ideal growth temperature os 20-22c with 16 hours of light supply.
• it’s also possible to grow plantlets in front of window.
9. LIQUID SHAKEN CULTURE.
• this method is expensive to setup since electric shaker is required.
• it’s much more rapid and produces more plantlets ready for sub culture after
3weeks.
• stem segments along with axillary buds transfer 3,4 of these stems segments
into flask on liquid medium.
• liquid murashige & skoog medoum is used.
10. CONT...
• ga has been added.
• flask is placed, at 20c for 16 hours.
• when plantlets are fully grown the flask will contain material for further
production of indiviual rooted in-vireo plantlets.
