WHAT WAS THE PROTOCOL?
1. Samples were frozen and stored at -80°C until sections were cut at 12 μm thick and collected
onto gelatin-coated slides (Menzel GmbH, Braunschweig, Germany).
2. For immunofluorescence labelling, serial sections of DRG were hydrated in PB buffer (pH=7.4)
for 15 mins.
3. Cell membranes were permeabilised in PB containing 0.1% Triton X-100 and afterwards
incubated for 1 h in 10% normal goat serum (NGS, Jackson ImmunoResearch Laboratories,
West Grove, USA) in PB containing 0.4% Triton X-100 (blocking solution).
4. Sections were incubated for 48 hrs at 4°C in a humid chamber with the respective primary
antibodies (FAAH, St. John’s laboratory; 1:50).
5. After three washes in PB, triple immunofluorescence was revealed by incubation at 4°C in for
24 hrs in the dark with the appropriate fluorochrome-conjugated secondary antibody.
6. Sections were washed 3 times for 15 mins with PB and coverslipped with Aquatex mounting
medium (Merck, Darmstadt, Germany).
7. The sections processed for immunofluorescence were studied with a fluorescence microscope
(DM RXA2) with confocal scanner (TCS SL) (Leica Microsystems GmbH, Manheim,
Germany) equipped with the following lasers: Ar 488, He-Ne 543, He-Ne 633.

Equilibrative nucleoside transporter 1 (ENT1) is a protein that in humans is encoded by the SLC29A1 gene. Multiple alternatively spliced variants, encoding the same protein, have been found for this gene. This gene is a member of the equilibrative nucleoside transporter family. The gene encodes a transmembrane glycoprotein that localizes to the plasma and mitochondrial membranes and mediates the cellular uptake of nucleosides from the surrounding medium. p70S6 kinase or p70S6K is a serine/threonine kinase that acts downstream of PIP3 and phosphoinositide-dependent kinase-1 in the PI3 kinase pathway. As the name suggests, its target substrate is the S6 ribosomal protein. Phosphorylation of S6 induces protein synthesis at the ribosome. The phosphorylation of P70S6K at threonine 389 has been used as a hallmark of activation by mTOR and correlated with autophagy inhibition in various situations. However, several recent studies suggest that the activity of P70S6K plays a more positive role in the increase of autophagy. Phosphorylation of Thr229 in the catalytic domain and Thr389 in the linker domain are most critical for kinase function. Phosphorylation of Thr389, however, most closely correlates with p70 kinase activity in vivo. Prior phosphorylation of Thr389 is required for the action of phosphoinositide 3-dependent protein kinase 1 (PDK1) on Thr229.
