Antimicrobial Screening of Vanga Vennai and Mathan Thailam for Diabetic Foot ...IOSR Journals
Siddha medicine is one of the ancient traditional science which is specialized in external medicines (Pura marunthukal). Diabetic patients develop foot ulcers as a major complication of Diabetes mellitus due to high susceptibility to infection which often leads to amputation. Diabetic foot infection is a combination of both aerobic and anaerobic organisms. Thus there is a need for effective and selective treatment strategies. This article focuses on antimicrobial screening of Vanga vennai (VV), Mathan Thailam (MT) and combination of both (VV+MT) which are the classical Siddha preparations. The methodology involved in the study is Agar well dilution method for anti microbial activity. Thus it paves a new way in treating infections of Diabetic foot ulcers. This preliminary study reveals these Siddha formulations as an effective therapeutic strategy for the management of Diabetic foot ulcer.

Evaluation of Oxidative Stress and Stress Induced Serum Protein Changes in Ps...Premier Publishers
Psoriasis is a non-infectious, chronic inflammatory disease of the skin. This study aimed to explore the oxidative stress and stress induced protein changes in psoriasis and to analyse the possible interrelationship between these factors among psoriasis patients. This was a hospital-based case control study which included 50 cases of psoriasis and 50 healthy controls. Serum malondialdehyde was measured by thiobarbituric acid reactivity assay method. Protein carbonyl (PC) was measured by Levine’s method. Protein bound sialic acid (PBSA) was measured using modified Aminoff’s method using a spectrophotometer. Data analysis was done using SPSS software version 16.0. Our results showed significant elevation in the oxidant status (MDA, PC and PBSA) among the psoriasis cases compared to the controls. Multiple linear regression analysis showed a linear relationship between MDA, PBSA and duration of psoriasis. Our study provides evidence to increased ROS production, indicated by increased lipid peroxidation in serum and stress induced protein changes in psoriasis. The findings of our study also suggest that as the duration of psoriasis increases the level of oxidative stress also increases which is indicated by the increase in MDA and PBSA levels.
Background: Chronic Obstructive pulmonary disease (COPD) is an increasing cause of morbidity and mortality world-wide. MMP 9 is an acute phase reactant secreted by the liver in response to infection, inflammation or tissue damage. Methods: This case-control study was conducted on 35 healthy controls and 40 COPD patients at a tertiary care hospital in north India. MMP 9 levels were measured in serum by ELISA Kit. Results: The present study showed that mean MMP 9 levels in serum was significantly higher in COPD group as compared to control group (p<0.0001) and the levels increased with the increasing severity of the disease. Conclusion: Our study confirms that MMP 9 levels were significantly higher in COPD patients as compared to controls and their levels increased with the increasing severity of the disease. Measuring MMP 9 levels in combination with other biochemical markers can be helpful in monitoring disease outcome and management of the disease. Diabetes is a very serious, chronic, and complex metabolic disorder that is based on multiples aetiologies with both acute and chronic. There are two types of diabetes, one types 1 DM and the other is type two DM. Both T1DM and T2DM are the most serious type of chronic conditions that are typical cannot be cured. Insulin stimulates adipocytes, myocytes, and hepatocytes, which uptake glucose from the circulatory system. Antidiabetic drugs show useful effects through decreasing glucose absorption in the intestines increasing insulin levels in the body or increasing the bodys sensitivity or decreasing its resistance to insulin. Most of these plants contain bioactive compounds. Such compounds are alkaloids, flavonoids, glycosides, terpenoids, carotenoids, etc., which are frequently implicated as having an Antidiabetic effect. The different types of phytochemicals include flavonoids, saponins, phenolic acids, alkaloids, tannin, stillness, and polysaccharides. They have two types of drug design structure based drug desingand ligand based drug design. The protein is a structure based ligand, which is based on drug design for the role of significantly quantitative structure activity relationship QSAR and pharmacophore analysis. Ligand based pharmacophore model generation depends on the information regarding the known biological activity without any structural information depends on the macromolecular target. The ligand based VS is more expensive due to structure based methods. Based on the opposing chemical functionalities, and the geometric arrangement about each other, where the interactions are observed there is the pharmacophore features are placed on the ligand side. In patients with chronic kidney disease (CKD), along with progression of CKD,
abnormalities of mineral and bone metabolism develop, which result in altered serum levels of minerals
such as calcium and phosphorus, as well as abnormalities in parathyroid hormone (PTH) or vitamin D
metabolism. Chronic Kidney Disease-Mineral Bone Disease (CKD-MBD) is a serious burden because of
increased cardiovascular mortality thus making therapeutic improvements essential in CKD-MBD. The
present study was aimed at evaluation of pharmacological management of CKD-MBD.
Methods:A retrospective study including 180 patients divided into two groups of 90 each (diabetes
mellitus and non-Diabetes) was performed in the Department of Nephrology, SVIMS, Tirupati. Patients
who were on follow up for at least 3 years (2015-2017) were considered, serum parameters were measured at every six months with a total of 6 visits. First visit was taken as baseline and sixth visit as
conclusion.
Results:The disease incidence of CKD-MBD is more common in male patients i.e. 67.8%. Serum calcium
levels were significantly increased and eGFR was significantly decreased in all patients with CKD at
conclusion compared to baseline.Further, Serum calcium levels were significantly increased at conclusion
in CKD patients without DM and eGFR was significantly decreased at conclusion compared to baseline
in CKD patients with DM. The proportion of untreated patients is high for all the drugs except vitamin D
analogues in both subgroups of CKD patients.
Conclusion:Pharmacological intervention in CKD patients helps in the effective management of mineral
bone disease by maintaining serum calcium, phosphate and calcium phosphorous product status.

Multiple sclerosis is a chronic inflammatory neurodegenerative disorder which directly affects Central Nervous System CNS . People with MS suffer with an episodic reversible memory loss during the initial stages and later it leads to the neurological deterioration. Number of research and studies has been done on the natural compounds and phytochemical compounds in order to develop the particular drug for the treatment of MS in vivo andin vitro. The present study focuses on the inhibitory effect of Rosmarinic acid against the effect of CD2 Associated protein with the help of Molecular Docking. Molecular Docking basically screens the ligand and the target protein and shows the interaction between them on the basis of the minimum binding affinities and drug likeliness properties. In our research, docking was performed between CD2 Associated protein and selected ligands with the help of docking software. Ligands were selected on the basis of their minimum Binding affinities and finally by their drug likeliness properties. Rosmarinic acid BA 5.6 was the resultant ligand of our recent study. It showed the perfect interaction with CD2 Associated protein. Therefore, we may conclude that Rosmarinic acid may act as a compound which may be used as a drug for the treatment of multiple sclerosis fromfurther in vitro and in vivostudies in future. People with MS suffer with an episodic reversible memory loss during the initial stages and later it leads to the neurological deterioration. Number of research and studies has been done on the natural compounds and phytochemical compounds in order to develop the particular drug for the treatment of MS in vivo andin vitro. The present study focuses on the inhibitory effect of Rosmarinic acid against the effect of CD2 Associated protein with the help of Molecular Docking. Molecular Docking basically screens the ligand and the target protein and shows the interaction between them on the basis of the minimum binding affinities and drug likeliness properties. In our research, docking was performed between CD2 Associated protein and selected ligands with the help of docking software. Ligands were selected on the basis of their minimum Binding affinities and finally by their drug likeliness properties. Rosmarinic acid BA 5.6 was the resultant ligand of our recent study. It showed the perfect interaction with CD2 Associated protein. Therefore, we may conclude that Rosmarinic acid may act as a compound which may be used as a drug for the treatment of multiple sclerosis fromfurther in vitro and in vivostudies in future. ABSTRACT- This study was undertaken to evaluate the serum levels of Oxidant (MDA) & antioxidant (SOD & Vitamin E) and compare oxidative stress (MDA) level among normotensive and hypertensive subjects. Oxidative stress has been relationship with mechanisms of EH (essential hypertension). A total number of 70 subjects were taken including both sex (Men and Women) between the ages of 35-70 years taken in this study. Exclusion criteria were chronic diseases, alcohol consumer, obesity, smoking/tobacco consumer and current use of any medication. Antioxidant enzymes activity and lipid peroxidation (malondialdehyde) were determined in serum. In 70 subjects out of 35 were found as an controls normotensive individuals and the cases 35 hypertensive patients. Serum MDA levels were highly significantly elevated in hypertensive patients in compared to normotensive individuals (4.39±0.98 µmol/l vs 1.51±0.70µmol/l and p < 0.0001). SOD acts as an antioxidant was highly significantly decrease in hypertensive patients in compared to normotensive individuals (0.44±0.06U/mg protein/min vs 0.96±0.04 U/mg protein/min and p <0.0001).>< 0.001). These findings demonstrate the strong association of SOD and Vitamin E level decrease in hypertensive patients and by MDA level increase in hypertensive patients. Oxidative stress in hypertensive patients increasing over time may play a role in the improvement of atherosclerosis and cardiovascular disease, should be considered in further research.

Background and objectives: the 5’nucleotidase (5’NT) is one of
hydrolytic enzymes present in different organs which catalyze
hydrolysis of 5’ nucleotides to ribonucleosides and phosphate.
Malondialdehyde (MDA) is the end product of lipid peroxidation
by oxidative stress (free radicals).
The aim of present study was to measure the serum activity of
5’NT, and MDA concentration in breast tumors.
Material and method: A prospective study was carried out from
May to December 2013 by clinical biochemistry department in
College of Medicine-Hawler Medical University on (30) healthy
female individuals, (group 1) and (30) females with breast tumor
(group 2).
Results: The mean value of serum MDA was significantly higher
in females with breast tumors (group 2), than that of healthy
female individuals, (group 1) (p<0.01),><0.01).
Conclusion: Based on findings of the present study it can be
concluded that breast tumors can cause release of the enzyme
5’NT from tumor cells, and lipid peroxidation by reactive oxygen
species (ROS), which cause elevation of MDA.
ABSTRACT- Background: Tuberculosis is one of the commonest chronic infectious diseases; highly endemic in India kills five lakh patients every year. Oxidative Stress plays important role in inflammatory and degenerative diseases including pulmonary tuberculosis. There is hardly any one study available in literature correlating oxidative stress, lipid profile values and antioxidant status together with the pulmonary tuberculosis; so we decided to conduct this study.
Methods: Study group included newly diagnosed 50 cases of pulmonary tuberculosis and control group included 50, age and sex matched healthy volunteers and employees. All the cases were subjected to complete physical and systemic examinations, routine investigations including Sputum for AFB by Ziehl-Neelsen staining, AFB culture and Chest X-ray and special tests like Erythrocyte sedimentation rate (ESR), Malondialdehyde (MDA) and Adenosine deaminase (ADA) and findings recorded and statistically analysed.
Results: In the study group with 33 males and 17 females, we were found Serum MDA mean ± SD 2.91±0.99; Serum ADA 38.15±13.47, while the mean levels of pleural fluid MDA and ADA in tubercular patients were found to be 1.65+0.53 nmoles/ml and 56.88+22.1 U/L respectively. While in controls with 61 males and 39 females, these values were 1.72±0.45 nmoles/ml (MDA), 20.15± 6.70 U/L (ADA) respectively.
Conclusion: Malondialdehyde (MDA) and Adenosine deaminase (ADA) were found statistically significantly higher in study group when compared with control, (p <0.001). Antioxidant plays important role for prevention of pulmonary Tuberculosis.

Induced Biochemical Changes and Associated Complications In Smokersiosrjce
Worldwide more than 3 million people currently die each year from smoking. Smoking of tobacco is
practised worldwide & common in the developing countries like India. The study is designed to determine the
lipid peroxidation and antioxidant status along with serum calcium level in smokers (cigarette/bidi) with
reference to the normal healthy non-smokers. 50 smokers(Cigarette/Bidi) of mean age 35± 5 years were
enrolled for the study & were compared to 50 normal healthy non-smokers of the same age. Blood samples were
obtained by puncture of antecubital vein and the following tests were performed i.e. P-MDA, Serum-SOD andSCalcium.
Increased plasma lipid per oxidation (P<0.05) /><0.05) in serum SOD & serum Calcium were observed in smokers when compared to
normal/healthy control subjects. The increased level of lipid per oxidation products in smokers reflects that
there is increased cell wall damage that may further leads to several complications such as COPD, Stroke and
the variety of lung and other cancers. It was also found that cigarette / bidi smoke alters the antioxidants status
as well as minerals imbalance in smokers and increases atherogenecity.
ABSTRACT- Reactive oxygen species (ROS) are products of normal cellular metabolism and are known to act as second messengers. Physiological conditions, ROS participate in maintenance of cellular ‘redox homeostasis’ in order to protect cells against oxidative stress through various redox-regulatory mechanisms. Oxidative stress resulting from enhances free-radical formation and/or a defect in antioxidant defences has been implicated in the pathogenesis of diabetes and its associated complications. Diabetes mellitus comprises a group of metabolic disorders that share the common phenotype of hyperglycemia, association with the biochemical alteration of glucose and lipid peroxidation. Increase level of oxidative stress along with deranging different metabolisms; one of the Long term complications of diabetes mellitus is diabetic retinopathy, which is a leading cause of acquired blindness. Many of the recent landmarks in scientific research have shown that in human beings, oxidative stress has been implicated in the progression of major health problems by inactivating the metabolic enzymes and damaging important cellular components, oxidizing the nucleic acids, leading to cardiovascular diseases, eye disorders, joint disorders, neurological diseases (Alzheimer’s disease, Parkinson’s disease and amyotrophic lateral sclerosis), atherosclerosis, lung and kidney disorders, liver and pancreatic diseases, cancer, ageing, disease of the reproductive system including the male and female infertility etc. In this review we have the importance of endogenous antioxidant defense systems, the intense medical management; these strategies include dietary measures (antioxidants) their relationship to several pathophysiological processes and their possible therapeutic implications in vivo condition. Doxorubicin (DOX) is a chemotherapeutic agent used for treatment of different cancers and its clinical usage is hindered by the oxidative injury-related cardiotoxicity. This work aims to declare if the harmful effects of DOX on heart can be alleviated with the use of Coenzyme Q10 (CoQ10) or L-carnitine. The study was performed on seventy two female Wistar albino rats divided into six groups, 12 animals each: Control group; DOX group (10mg/kg); CoQ10 group (200mg/kg); L-carnitine group (100mg/kg); DOX+CoQ10 group; DOX+L-carnitine group. CoQ10 and L-carnitine treatment orally started 5days before a single dose of 10mg/kg DOX that injected intraperitoneally (IP) then the treatment continued for 10days. At the end of the study, serum biochemical parameters of cardiac damage, oxidative stress indices, and histopathological changes were investigated. CoQ10 or L-carnitine showed a noticeable effects in improving cardiac functions evidenced reducing serum enzymes as serum interleukin-1 beta (IL-1 β), tumor necrosis factor alpha (TNF-α), leptin, lactate dehydrogenase (LDH), Cardiotrophin-1, Troponin-I and Troponin-T. Also, alleviate oxidative stress, decrease of cardiac Malondialdehyde (MDA), Nitric oxide (NO) and restoring cardiac reduced glutathione levels to normal levels. Both corrected the cardiac alterations histologically and ultrastructurally. With a visible improvements in α-SMA, vimentin and eNOS immunohistochemical markers. CoQ10 or L-carnitine supplementation improves the functional and structural integrity of the myocardium.

radicals represent a class of molecules derived from the
metabolism of oxygen and exists inherently in all aerobic
organisms (Yoke, 2003). Oxidants such as hydrogen
peroxide, superoxide radicals, hydroxyl radicals and lipid
peroxides (LOOH) play an ever more important role in
human diseases (Thompson, 1995). Mononuclear and
polymorphonuclear leucocytes have the potential to
synthesize and release a variety of potentially toxic agents
including ROS into the extracellular environment where
they may injure cells and tissues, when in large quantities.
This study was designed to investigate the extent of
involvement of oxidative stress in typhoid salmonellosis
as well as compare its effect on total lipid and cholesterol
levels in both infected and healthy individuals, being a
measure of the health status of an individual.
MATERIALS AND METHOD
Sera obtained from the blood of one hundred and fifty two
(152) clinically confirmed typhoid patients in the
University Medical Centre, University of Nigeria, Nsukka
over a period of four months as well as from donor
healthy individuals were assayed for MDA (lipid
peroxidation product), total lipid and total cholesterol
levels. Subjects were of varying ages, sex, weight and
different stages of typhoid salmonellosis infection. The
design and conduct of the study were in accordance with
the Helsinki declaration. Lipid peroxidation product,
MDA was assayed using the method described by Wallin
et al., 1993 . This Principle is based on the reaction of
thiobarbituric acid (TBA) with malondialdehyde (MDA)
under slightly acidic conditions to produce a pink or red
colour complex with absorbs at 532nm (Anosike et al.,
2008). The concentration of MDA is proportional to the
level of peroxidation.
Total lipid levels were assayed using the method of
Zollner and Kirsch (1962) which is based on the principle
of colour reaction determined as absorbance at 530nm
(Romeu-Moreno and Mas 1999), of unsaturated lipids
with phosphoric acid-vanillin reagent. A modification of
the Zak (1957) reaction technique was employed in the
estimation of total cholesterol levels. It required that
cholesterol be extracted from serum with ethanol. This
extract is then reacted with a solution of FeCl3 dissolved
in phosphoric acid, and the resulting colour read-off
in a spectrophotometer at 550nm against a known
concentration that was treated in the same manner.
Normal values for cholesterol tend to increase with age,
but the average values range from 100 to 200mg/dl. Assay
results were expressed as mean±standard deviation. Paired
sample t-tests were used for comparisons of variables
between typhoid and healthy individuals. Correlations
were determined by Spearman correlation coefficient
method. Results were considered statistically significant
when P <0.05.
RESULTS
The mean total lipid levels in typhoid patients were
128.98±3.39 and 154.95±11.40 in healthy individuals
(Fig1). The mean total cholesterol levels in typhoid
patients were 142.22± 29.83 and that of healthy
individuals were 211.19±22.02 (Fig.2). The mean
malondialdehyde (MDA) levels were observed as
1.073±0.51 and 0.820±0.53 in typhoid and healthy
individuals respectively (Fig.3). Comparisons of the total
lipid levels, total cholesterol and MDA levels showed a
statistical significance between concentrations of total
cholesterol (P<0.01) and total lipids (P<0.00). No
statistical significance were observed in concentrations of
MDA (P<0.16).

DISCUSSION
Comparing healthy to presenting patients in Nsukka urban
area, decline was observed in the mean total lipid levels
(Fig1.) and mean total cholesterol levels (Fig. 2.) but the
mean MDA levels indicated an elevation (Fig. 3). Lipid
peroxidation results in oxidative modification of lipids and
ultimately leads to the development of ulcers (Dennis,
1992). Typhoid fever is associated with inflammation and
ulceration of the gut and liver of which oxidative lipid

modification plays an important role in exacerbating the
disease.
One of the frequently used biomarkers giving an
indication of lipid peroxidation is the serum concentration
of MDA – a product of lipid peroxidation processes.
MDA levels were observed to be higher in typhoid
patients than in healthy individuals. This compares with
previous works by Pranzy et al., 1999 showing higher
MDA levels in malaria patients when compared to healthy
individuals. The increased MDA levels could be due to
the activation of the immune system which increases
inflammation of the bowel as well as activation of
phagocytes, mononuclear and polymorphonuclear
leukocytes (PMNs) that have the potentials to synthesize
and release potentially toxic agents such as reactive
oxygen species (ROS). Furthermore, the study of Pranzy,
1999 on oxidative stress in malaria patients showed that
cholesterol levels decreased tremendously in malaria
patients when compared with healthy individuals. In this
study, this decrease in cholesterol levels and total lipids
were also observed, and may be attributed to lipid
peroxidation, loss of appetite and diarrhoea which are
classics in typhoid patients and partially due to the
biosynthesis and activation of immune responses like
prostaglandins, interleukins, interferons, tissue necrotic
factor (TNF), etc. (Pranzy et al., 1999). The decrease in
the level of total cholesterol concentration in typhoid
patients agrees with previous works done by (Khosla et
al., 1991) in which low levels of HDL-cholesterol where
observed in enteric fever patients. Results obtained
therefore demonstrate the implication of oxidative stress
in typhoid salmonellosis.
