The initial macroscopic examination of semen includes observing liquefaction, viscosity, appearance, volume, and pH. Liquefaction normally occurs within minutes at room temperature and may contain jelly-like granules. Viscosity is estimated by allowing a sample to drop and observing the length of any thread formed, with >2cm indicating high viscosity. Appearance can provide clues to conditions like infection or illness. Normal volume is 2-5mL. pH should be measured 30 minutes after liquefaction and typically ranges from 6.0-7.0.
