Protocol
Treatment of Materials: Freshly dissociated human bladder smooth muscle cells (hbsmc). Seeded on PLL covered glass
coverslips. Fixed with 4% PFA on buffer phosphate-saline for 20 min. Rinse 3X with PBS.
Blocking: BSA 1%, Triton 0.1%, Tween 20 0.05% in PBS for 20 min.
Primary antibody incubation: primary antibody in blocking solution and incubated at 4˚C for overnight.
Wash: PBS 3X 10 min each
Secondary antibody incubation: secondary antibody in blocking solution and incubated at RT for 2 hrs.
Wash: PBS 3X 10 min each
Visualization: Coverslips mounted in ProLong Antifade on glass slides. Dry overnight at RT. Confocal imaged.

The enzyme-linked immunosorbent assay (ELISA) uses antibodies and a solid-phase enzyme immunoassay to detect the presence of a specific antigen in a liquid or wet sample. Antigens from the sample are immobilised on a solid support either non-specifically or specifically. A specific antibody is then applied over the surface so it can bind to the antigen. This antibody is linked to an enzyme or can itself be detected by an enzyme-linked secondary antibody. In the final step, a substance containing the enzyme's substrate is added. The subsequent reaction produces a visible signal, most commonly a colour change in the substrate, which indicates the quantity of antigen in the sample. When the presence of an antigen is analysed, the name "direct ELISA" refers to an ELISA in which only a labelled primary antibody is used, whereas the term "indirect ELISA" refers to an ELISA in which the antigen is bound by the primary antibody which then is detected by a labelled secondary antibody.

After graduating with an honours degree in Biochemistry Dr Hills worked for several years as a Clinical Scientist at St Bartholomew's hospital in London. He was awarded his PhD in 2002 from the faculty of Medicine at Queen Mary University of London. He continued his interest in reproductive science at Imperial College London where he worked as a postdoctoral researcher before joining Middlesex in 2004 as a lecturer. Dr Hills has published many high profile original research articles on various aspects of obstetric pathology including pre-eclampsia, recurrent miscarriage, preterm labour and fetal distress as well as several articles in the area of assisted reproduction. Currently, he is research interests include investigating the role of glycosaminoglycans and proteoglycans on the development of placental pathology and breast cancer. Dr Hills teaches a range of topics in biomedical science including clinical biochemistry, cellular and developmental biology as well as statistical analysis. He is author of around 30 peer-reviewed scientific articles and has refereed manuscripts for a variety of journals in the area of reproduction and endocrinology. Dr. David Rimm is a Professor in the Department of Pathology at the Yale University School of Medicine. He completed an MD-PhD at Johns Hopkins University Medical School followed by a Pathology Residency at Yale and a Cytopathology Fellowship at the Medical College of Virginia. Dr. Rimm is the Director of Translational Pathology and the Director of Yale Pathology Tissue Services. His lab group (15 researchers) focuses on quantitative pathology using the AQUA® technology invented in his lab with projects related to predicting response to therapy in breast and lung cancer and predicting recurrence or metastasis in breast and lung cancer. The technology has also been used in a series of efforts related to biospecimen science. He also has a group working on primary tumor culture using the conditionally reprogrammed cell method. He is a member of a number of correlative science committees for multi-institutional breast cancer clinical trials including SWOG, ALLTO, and TEACH. He also serves on the Molecular Oncology committee for the College of American Pathologists (CAP). He is currently supported by grants from both public and private sources. He is an author of over 280 peer-reviewed papers and 8 patents. He has served on advisory boards for Genentech, Novaritis, BMS, Perkin Elmer, Dako, ACD, Avida and Genoptix. He was a scientific co-founder of HistoRx, a digital pathology company (sold to Genoptix in 2012) and Metamark Genetics, a prognostic determinant company. Avacta Life Sciences Exhibits Affimers at Global Protein Engineering Summit Avacta Life Sciences exhibited recently at the Global Protein Engineering Summit ("PEGS") where it presented its Affimer technology. You can read more about Affimer technology here http://www.avactalifesciences.com PEGS is considered to be the essential protein engineering meeting where commercial and academic progress in protein engineering is showcased and this year it attracted over 1800 delegates from across the globe to Boston. Avacta Life Sciences presented its Affimer technology for the first time at a PEGS meeting with technical exhibits and a presentation by the CSO, Paul Ko Ferrigno, entitled "Biological Recognition: Beyond the Antibody."* The exhibition booth was busy with over 80 delegates talking to the Avacta Life Science management team over the four days of the summit. The feedback on the Affimer technology was very positive, in particular, the short development times and excellent stability were highlighted by delegates as key advantages of Affimers over antibodies. There was also a strong interest in Affimers from the management of companies developing biological therapeutics who were keen to learn more about the potential of Affimers as novel therapeutics. In addition, several companies were interested in the use of Affimers as an alternative to antibodies in diagnostic devices, mainly because they could generate binders against new biomarkers much more quickly and evaluate them in higher numbers. The benefits of Affimer microarrays for biomarker discovery also resonated with diagnostic developers who appreciated the advantage of being able to evaluate significantly larger numbers of potential biomarkers more cost and time effectively than by mass spectrometry. The potential of the arrays for multiplexed solutions for clinical diagnosis and monitoring during drug trials was also something that generated interest amongst those delegates. Matt Johnson, Chief Technical Officer of Avacta Life Sciences commented: "It was great to experience face to face the level of interest in Affimers. The majority of people I spoke to were either having problems raising antibodies to their target of interest or just couldn't use antibodies because of the type of assays they wanted to perform. Many of the presentations focused around the use of antibody fragments for intra-cellular studies which is a rapidly growing area that holds great interest for drug and diagnostics developers. It is an area where there are clear advantages for Affimers over antibody fragments which don't behave well in the cytoplasm. "The general enthusiasm around Affimers was very encouraging and the amount of interest generated by the potential of Affimers as therapeutics and by the Affimer arrays for biomarker discovery only reinforces my excitement around this new technology."

Glial fibrillary acidic protein is an intermediate filament (IF) protein that is expressed by numerous cell types of the central nervous system (CNS) including astrocytes and ependymal cells. GFAP has also been found to be expressed in glomeruli and peritubular fibroblasts taken from rat kidneys Leydig cells of the testis in both hamsters and humans, human keratinocytes, human osteocytes and chondrocytes and stellate cells of the pancreas and liver in rats. Tubulin in molecular biology can refer either to the tubulin protein superfamily of globular proteins, or one of the member proteins of that superfamily. α- and β-tubulins polymerize into microtubules, a major component of the eukaryotic cytoskeleton. Microtubules function in many essential cellular processes, including mitosis. Tubulin-binding drugs kill cancerous cells by inhibiting microtubule dynamics, which are required for DNA segregation and therefore cell division. In eukaryotes there are six members of the tubulin superfamily, although not all are present in all species (see below).[2][3] Both α and β tubulins have a mass of around 50 kDa and are thus in a similar range compared to actin with ~42 kDa. In contrast, tubulin polymers (microtubules) tend to be much bigger than actin filaments due to their cylindrical nature.
