AUA DNA Barcoding 2018 Group 1
DNA barcoding was used to identify plant species collected from Mount Obama in Antigua. DNA was successfully extracted from 15 plant samples using a plant DNA extraction kit. PCR amplification using rbcLa primers was successful for 6 samples as shown by bands on an agarose gel. The 6 samples were sent for sequencing and the sequences matched to 6 plant species in the NCBI database: Morinda pubescens, Mimosa pudica, and Turnera ulmifolia. This demonstrated that DNA barcoding can be used to identify plant species found on Mount Obama.

Of the 18 samples tested, DNA was extracted from 15 and 11 species were identified through DNA sequencing and database analysis. There was no overlap between the species found at the peak and those previously collected at the base. The experiment proved DNA barcoding can accurately identify biodiversity in Antigua.

DNA barcoding help to recognize the plant based on short, gene sequences in a rapid, accurate, and cost effective manner. Current focus is on the investigation of phytomedicinals and herbal product integrity and authenticity through DNA barcoding with the goal of protecting consumers from potential health risks associated with product substitution and contamination. Recent reports reveal that DNA barcoding can be used for the assignment of unknown specimens to a taxonomic group, authentic identification of phytomedicinals, and in plant biodiversity conservation. Research indicates that there is no single universal barcode candidate for identification of all plant groups. Hence, comparative analysis of plant barcode loci is essential for choosing a best candidate for authenticating particular medicinal plant genus/families. Currently, both chloroplast/nuclear regions are used as universal barcodes for the authentication of phytomedicinals. A recent advance in genomics has further enhanced the progress in DNA barcoding of plants by the introduction of high-throughput techniques like next generation sequencing, which has paved the way for complete plastome sequencing that is now termed as super-barcodes. Hence, current focus is on the investigation of phytomedicines and herbal product integrity and authenticity through DNA barcoding with the goal of protecting consumers from potential health risks associated with product substitution and contamination. These approaches could improve the traditional ethnobotanical and scientific knowledge of phytomedicines and their safe use.

Pigeonpea is an important crop for food security in India and other parts of the world as it is drought tolerant and a source of protein and nutrients. It has a genome size of 858 million base pairs. The first draft genome sequence of the popular pigeonpea variety Asha was completed in 2012 using 454 sequencing, assembling over 500 million base pairs across 11 chromosome pairs. A new reference genome was published in 2018 using Illumina sequencing, assembling over 605 million base pairs across 72.7% of the 833 million base pair genome. The new genome provides insights into drought tolerance genes and will help improve pigeonpea varieties.

Abstract
In recent times, there is an extensive interest in these Alcoholic extracts due to the emergence and spread of new drugresistant human pathogens to existing antimicrobials. The emergence of medicine opposing pathogens is one of the mostcritical threats to booming treatment of bacterial diseases. Mode of action of Methanolic extracts likely involves fairly a lotof targets in the cell due to huge number of active components and also their hydroplillicity helps them to screen in the cellmembrane, rendering them permeable , leading to leakage of cell contents. This calls for a transformed effort to identifyagents efficient against disease causing bacteria to present antimicrobials. Seed extracts of two different plants viz. Phoenixdactylifera and Annona squamosa, were prepared by methanol extraction method at the ratio of 1:2 using 100ml volume of methanol and stock concentration of 50mg/ml in dimethyl sulfoxide (DMSO) of each extract was made.The extracts and fractions were tested for antimicrobial activity against standard microbial strains of Klebsiella pneumoniae (gramnegative), Staphylococcus aureus (gram-positive), Escherichia.coli (gram-negative), Salmonella typhi (gram-negative) , Enterococcus faecalis (grampositive), Pseudomon aerugenosa (gram-negative),and Salmonella paratyphi (gram-negative)by means of Agar-Disc Diffusion Method and minimal inhibitory concentration (MIC) was noted .. The test culture of standard microbial cultures was 3 X 105 CFU/ml, and standard antibiotic used is Ampicillin with clavulanic acid. In this context, two extract from traditional plants, Custard Apple (Annona squamosa) and Dates (Phoenix dactylifera) were used alone or in combination to assess their antimicrobial efficacy against both Gram negative and Gram positive bacterial clinical isolates .Antimicrobial test was completed by agar disc diffusion method. Although, both extract were found to be effective in inhibiting pathogens to varying degrees to the tested organisms, the Annona squamosa extract is found to be more effective than Phoenix dactylifera.When both extracts were used in combination, they have shown strong synergistic effect against all the pathogens tested in the present studyexcept for the P.aerugenosa and S. Para typhi. Bactericidal abilities displayed by the seed extracts signified their remarkable potential for exploration for effective natural antimicrobial agents against standard pathogenic bacteria. The extracts have shown the synergistic effects even at their MIC against E.fecalis, indicating that with further researches these extracts can be used for treating enteric diseases.

Current focus is on the investigation of phytomedicinals and herbal product integrity and authenticity through DNA barcoding with the goal of protecting consumers from potential health risks associated with product substitution and contamination. Recent reports reveal that DNA barcoding can be used for the assignment of unknown specimens to a taxonomic group, authentic identification of phytomedicinals, and in plant biodiversity conservation. Research indicates that there is no single universal barcode candidate for identification of all plant groups. Hence, comparative analysis of plant barcode loci is essential for choosing a best candidate for authenticating particular medicinal plant genus/families. Currently, both chloroplast/nuclear regions are used as universal barcodes for the authentication of phytomedicinals. A recent advance in genomics has further enhanced the progress in DNA barcoding of plants by the introduction of high-throughput techniques like next generation sequencing, which has paved the way for complete plastome sequencing that is now termed as super-barcodes. Hence, current focus is on the investigation of phytomedicines and herbal product integrity and authenticity through DNA barcoding with the goal of protecting consumers from potential health risks associated with product substitution and contamination. Pigeonpea is an important crop for food security in India and other parts of the world as it is drought tolerant and a source of protein and nutrients. It has a genome size of 858 million base pairs. The first draft genome sequence of the popular pigeonpea variety Asha was completed in 2012 using 454 sequencing, assembling over 500 million base pairs across 11 chromosome pairs. A new reference genome was published in 2018 using Illumina sequencing, assembling over 605 million base pairs across 72.7% of the 833 million base pair genome. The new genome provides insights into drought tolerance genes and will help improve pigeonpea varieties.

In recent times, there is an extensive interest in these Alcoholic extracts due to the emergence and spread of new drugresistant human pathogens to existing antimicrobials. The emergence of medicine opposing pathogens is one of the mostcritical threats to booming treatment of bacterial diseases. Mode of action of Methanolic extracts likely involves fairly a lotof targets in the cell due to huge number of active components and also their hydroplillicity helps them to screen in the cellmembrane, rendering them permeable , leading to leakage of cell contents. This calls for a transformed effort to identifyagents efficient against disease causing bacteria to present antimicrobials. Seed extracts of two different plants viz. Phoenixdactylifera and Annona squamosa, were prepared by methanol extraction method at the ratio of 1:2 using 100ml volume of methanol and stock concentration of 50mg/ml in dimethyl sulfoxide (DMSO) of each extract was made.The extracts and fractions were tested for antimicrobial activity against standard microbial strains of Klebsiella pneumoniae (gramnegative), Staphylococcus aureus (gram-positive), Escherichia.coli (gram-negative), Salmonella typhi (gram-negative) , Enterococcus faecalis (grampositive), Pseudomon aerugenosa (gram-negative),and Salmonella paratyphi (gram-negative)by means of Agar-Disc Diffusion Method and minimal inhibitory concentration (MIC) was noted .. The test culture of standard microbial cultures was 3 X 105 CFU/ml, and standard antibiotic used is Ampicillin with clavulanic acid. In this context, two extract from traditional plants, Custard Apple (Annona squamosa) and Dates (Phoenix dactylifera) were used alone or in combination to assess their antimicrobial efficacy against both Gram negative and Gram positive bacterial clinical isolates .Antimicrobial test was completed by agar disc diffusion method. Although, both extract were found to be effective in inhibiting pathogens to varying degrees to the tested organisms, the Annona squamosa extract is found to be more effective than Phoenix dactylifera.When both extracts were used in combination, they have shown strong synergistic effect against all the pathogens tested in the present studyexcept for the P.aerugenosa and S. Para typhi. Bactericidal abilities displayed by the seed extracts signified their remarkable potential for exploration for effective natural antimicrobial agents against standard pathogenic bacteria. The extracts have shown the synergistic effects even at their MIC against E.fecalis, indicating that with further researches these extracts can be used for treating enteric diseases.

This document discusses genetic diversity analysis of grape (Vitis vinifera L.) germplasm in India using microsatellite markers. A total of 42 grape genotypes were analyzed using 7 microsatellite markers. A total of 45 alleles were detected among the genotypes. The microsatellites grouped the genotypes into two main clusters (A and B) based on morphological and genetic characteristics, with subclusters differentiating seeded vs seedless fruits and pigmented vs non-pigmented fruits. The study found high genetic variability among the Indian grape germplasm and that microsatellite markers are a reliable tool for diversity and breeding programs.

The plants used in the present study were Ocimum sanctum (Tulsi), Withania somnifera (Ashwgandha), Santalum album (Chandan), Aloe vera (Aloe barbadensis), and shatavari (Asparagus racemosus). The extract from the leaves of these plants (are) used in malaria, bronchitis, gastric disorders, cough, cold etc. To test efficiency of some common plants extract against E. coli, Salmonella typhi, Proteus vulgaris, Staphylococcus aureus. Contrary to the synthetic drugs, antimicrobials of plant origin are not associated with many side effects and have an enormous therapeutic potential to heal many infectious diseases. The present investigation is therefore, undertaken to test the efficiency of some of the common plant extracts against some plants and human pathogens, i.e. E. coli and S. aureus. In this project work, we studied the different parts of medicinal plants of Latur, Osmanabad region used for curing different type of diseases specially skin diseases. Some plants have active components which show antimicrobial activity. These Herbal plants are beneficial to human being in therapeutic practice. Skin diseases are difficult conditions to live with, to save the very least. Though some skin diseases may cause minimal discomfort, the visual effects of the conditions can cause significant self esteem and confidence issues. The majority of skin diseases cause scarring or disfigurement. Skin diseases run the gambit from barely noticeable to fatal.

Antibacterial Activity of Citrus Sinensis X against Pathogenic Bacterias
Citrus sinensis was tested for antibacterial activity against bacteria strains such as E.coli, pseudomonas aeruginosa, klebsiella pneumoniae, and staphylococcus aureus. The extraction was done with water and ethanol. Agar disc diffusion was used to test antibacterial activity in vitro method. On Escherichia coli, the antibacterial action of aqueous extract demonstrated a zone of inhibition. On the other organisms, it showed little or no zones of infection, although being seven millimeters in diameter. Inhibition with a diameter of 0 3mm. little inhibitory zones were also visible in the ethanol extract. On test organisms with diagram ranging from 1 3mm the inhibitory concentration that must be met MIC . Using a two flow serial dilution method, no zones of contamination were found in the ethanol and aqueous extract. Genetic characterization of morphological and yield traits in ten genotypes of Celosia argentea L. was evaluated
at the Research Farm of the Department of Botany, University of Ibadan, Nigeria. The experiment was laid out
in a randomized complete block design with four replicates. The results of analysis of variance carried out on
early morphological characters of C. argentea L. at 3, 4, and 5weeks after sowing showed significant
(p<0.05 /><0.01) effects except for number of leaves per plant and leaf width at 3 and 5 weeks after sowing,
respectively. The replicates in blocks produced varying observable effects on the genotypes while genotype x
replicate showed significant variation on morpho-agronomic and yield traits except number of days to flowering
at 50 days and fruit length at maturity. Also, from the result of the mean separation, it is shown that
NG/MAY/09/015 performed the best for plant height at flowering, leaf length at flowering, leaf width at
flowering, and root biomass. NG/SA/07/213 produced the highest mean values of number of flowers per plant,
leaf biomass and pod weight at maturity. The highest values of number of primary branches and fruit length at
maturity (FLM) were observed for NG/TO/MAY/09/015, while NG/AO/MAY/09/015 had the highest for pod
weight at maturity. The result of principal component axis also showed that Prin 1 accounted for highest Eigen
Vector of 38.62% from the total variation. NG/MAY/09/015 (R2) genotype produced the highest Eigen Vector
of 6.705 from Prin 1. The correlation result showed that plant height had a significant positive association with
seed weight at maturity, pod weight at maturity, number of primary branches and fruit length at maturity, while
similar association existed between leaf biomass, number of primary branches and pod weight at maturity, as
well as between plant height at flowering and pod weight at maturity. Again, the number of primary branches is
also positive and significantly correlated with plant height, root biomass and leaf length. Furthermore, the
results of dendrogram and minimum spanning tree revealed variations in genetic relatedness and distance,

Thematic analysis in qualitative research is a time-consuming and systematic task, typically done using teams. Team members must ground their activities on common understandings of the major concepts underlying the thematic analysis, and define criteria for its development. However, conceptual misunderstandings, equivocations, and lack of adherence to criteria are challenges to the quality and speed of this process. Given the distributed and uncertain nature of this process, we wondered if the tasks in thematic analysis could be supported by readily available artificial intelligence chatbots. Our early efforts point to potential benefits: not just saving time in the coding process but better adherence to criteria and grounding, by increasing triangulation between humans and artificial intelligence. This tutorial will provide a description and demonstration of the process we followed, as two academic researchers, to develop a custom ChatGPT to assist with qualitative coding in the thematic data analysis process of immersive learning accounts in a survey of the academic literature: QUAL-E Immersive Learning Thematic Analysis Helper. In the hands-on time, participants will try out QUAL-E and develop their ideas for their own qualitative coding ChatGPT. Participants that have the paid ChatGPT Plus subscription can create a draft of their assistants. The organizers will provide course materials and slide deck that participants will be able to utilize to continue development of their custom GPT. The paid subscription to ChatGPT Plus is not required to participate in this workshop, just for trying out personal GPTs during it.

We will metaverse into the essence of immersive learning, into its three dimensions and conceptual models. Challenging the perception of learning as knowledge transfer, we introduce a 'Uses, Practices & Strategies' model operationalized by the 'Immersive Learning Brain' and ‘Immersion Cube’ frameworks. This approach offers a comprehensive guide through the intricacies of immersive educational experiences and spotlighting research frontiers, along the immersion dimensions of system, narrative, and agency. Our discourse extends to stakeholders beyond the academic sphere, addressing the interests of technologists, instructional designers, and policymakers. We span various contexts, from formal education to organizational transformation to the new horizon of an AI-pervasive society. This keynote aims to unite the iLRN community in a collaborative journey towards a future where immersive learning research and practice coalesce, paving the way for innovative educational research and practice landscapes.

Anti-Universe And Emergent Gravity and the Dark Universe
Recent theoretical progress indicates that spacetime and gravity emerge together from the entanglement structure of an underlying microscopic theory. These ideas are best understood in Anti-de Sitter space, where they rely on the area law for entanglement entropy. The extension to de Sitter space requires taking into account the entropy and temperature associated with the cosmological horizon. Using insights from string theory, black hole physics and quantum information theory we argue that the positive dark energy leads to a thermal volume law contribution to the entropy that overtakes the area law precisely at the cosmological horizon. Due to the competition between area and volume law entanglement the microscopic de Sitter states do not thermalise at sub-Hubble scales: they exhibit memory effects in the form of an entropy displacement caused by matter. The emergent laws of gravity contain an additional ‘dark’ gravitational force describing the ‘elastic’ response due to the entropy displacement. We derive an estimate of the strength of this extra force in terms of the baryonic mass, Newton’s constant and the Hubble acceleration scale a0 = cH0, and provide evidence for the fact that this additional ‘dark gravity force’ explains the observed phenomena in galaxies and clusters currently attributed to dark matter.

What is DNA Barcoding? ● DNA barcodes allow non-experts to objectively identify different species regardless of what the specimen has been exposed to or where it came from ● A species’ DNA is uniquely different to them ● DNA barcoding turns DNA into an easily translatable, unique pattern of bars, such as seen in a consumer product’s universal product code (UPC)
- 3. Goals ● DNA Barcoding has never been performed on the flora living on the island of Antigua ● This project hopes to initiate conservation efforts for this island, as well as surrounding Caribbean islands ● To be able to connect Antiguan flora to Barcoding projects around the globe
- 4. Acquiring the Data ● Location: Lookout Trail, Galleon Beach
- 5. Plant-crushing method (aka DNA extraction and PCR) 1. Get plant 2. Extract DNA 3. Run a gel electrophoresis 4. Sequence DNA 5. Upload DNA sequences to DNA subway and Blast websites

Data analysis and input ● Upload DNA into DNA subway, a DNA sequence annotator and analysis website ● Upload the resulting sequence to Bold- Systems SPD website, an integrated workbench supporting the assembly, analysis and publication of DNA barcode data by students. ● Enter Barcode data into BLAST website to identify regions of similarity between documented sequences and the unknown sequence and calculates the statistical significance
- 7. Findings ● Several of our collected samples produced a positive identification of known species when entered into the BLAST nucleotide analysis software ● When positive identifications came up, the unknown species was identified, documented and further researched

Cool plant Specimen Jasminum fluminense (Vell.) “Common Jasmine” medicinal plant used by Shangaan people in villages under Jongilanga tribal council, South Africa as a STI medication [7] Talinum paniculatum (Jacq. Gaertn.) “Fameflower" a source of pharmaceutical efficacy similar to ginseng, and a widely distributed and planted edible vegetable [1, 5] Rivina humilis L. “Bloodberry” the berries are safe to consume, rich in nutrient content and exhibit efficient biological activity [4]
- 9. Conclusions ● Several of the species we collected were able to be sequenced, identified and recorded in the Barcode Database ● The efforts of this research will lead to further evaluation of the floral species present on the island ● In addition to further experimentation, island conservation efforts will increase due to increased knowledge on what species of plants live on the island
- 10. If we were to repeat this experiment ● We would want to ensure we received enough of the plant sample to ensure a successful DNA extraction ● We would want to repeat the PCR process and electrophoresis process to ensure the best DNA samples of each species get sent for analyzation ● We would want to further analyze the surveyed location for increased awareness of what potentially could be found Vachellia farnesiana
