This document provides instructions for performing three experiments:
1) Estimating protein concentration using the Lowry method.
2) Isolating nucleic acids from E. coli and measuring concentration using a spectrophotometer.
3) Separating and visualizing DNA using agarose gel electrophoresis.
Reagents, equipment, and step-by-step procedures are described for each experiment. Calculations are shown for determining protein concentration from absorbance measurements.

SAMPLE SYNTHESIS FOR COMPOSITE • Sample preparation was done by Extraction of Cucamelon powder for 5 hours at 80 C • Extract was filter by simple filtration over night. • 0.04 Molar solution sample was prepared, first 70ml extract was taken in a beaker and 0.1 C-Tab was added in it, then 10ml solution of 0.4404 Zinc Acetate was added into extract. pH was maintained to 10. • The whole 80 ml solution was prepared by hydrothermal method in oven at 120 C for 2 hours. • Then after cooling the sampler it was centrifuge for 3 to 4 times, finally sample for Composite was prepared.
- 4. AGNO3 COMPOSITE For AgNO3 Composite 65ml extract was taken which was prepared by extraction for five hours at 80 C and filtered by simple filtration overnight. In 5ml DI water 0.0271792 AgNO3 was mixed. In 10ml DI water prepare sample was added. That solution was added in 5ml AgNo3 solution and this whole solution was added into extract drop by drop. pH was maintained to 10. 80ml solution was added into autoclave for 2 hours at 120 C.
- 5. AGNO3 DOPING 5% Doping of AgNO3(0.0015M) was prepared by adding 0.0203844 AgNO3 into 10ml DI water. 0.50046 Zinc Acetate was also added in AgNO3 solution. This solution was then added into 70ml extract which was prepared by 5 hour extraction at 80 C and filtered by simple filtration. pH was maintained to 10 and then whole solution was added into autoclave for 2 hours at 120 C.
- 6. COPPER ACETATE DOPING 5% Doping of Copper(II) Acetate(0.0015M) was prepared by adding 0.0217956 Copper Acetate into 10ml DI water. 0.50046 Zinc Acetate was also added in AgNO3 solution. This solution was then added into 70ml extract which was prepared by 5 hour extraction at 80 C and filtered by simple filtration. pH was maintained to 10 and then whole solution was added into autoclave for 2 hours at 120 C.
- 7. C0PPER ACETATE COMPOSITE For Copper Acetate Composite 65ml extract was taken which was prepared by extraction for five hours at 80 C and filtered by simple filtration overnight. In 5ml DI water 0.290608 Copper Acetate was mixed. In 10ml DI water prepare sample was added. That solution was added in 5ml Copper Acetate solution and this whole solution was added into extract drop by drop. pH was maintained to 10. 80ml solution was added into autoclave for 2 hours at 120 C.
