Diversity of O Antigens within the Genus Cronobacter - Martina
This study analyzed the diversity of O antigens in the bacterial genus Cronobacter by testing 82 strains representing all Cronobacter species. Restriction fragment length polymorphism analysis of the O-antigen gene cluster identified 11 previously reported and 6 new serotypes. Whole genome sequencing of reference strains confirmed the new serotypes and showed some existing PCR probes did not correctly identify genomic variations. Analysis of lipopolysaccharide phenotypes also differentiated 24 total serotypes among Cronobacter strains. Certain serotypes including C. sakazakii O2, O1, and O4 and C. turicensis O1 were found to predominately cause clinical infections. This work provides an updated systematic classification of Cronobacter serotypes.

This document describes the development of PCR primers to rapidly identify the anaerobic bacterium Robinsoniella peoriensis. R. peoriensis has been isolated from a variety of mammalian and human infections. The study developed two PCR primer sets targeting the 16S rRNA gene of R. peoriensis. Testing on 15 R. peoriensis strains and 4 closely related bacterial species showed one primer set accurately amplified only R. peoriensis DNA. The primer set could identify R. peoriensis from both bacterial cultures and boiled whole cells, demonstrating its potential for rapid identification in clinical and environmental laboratories.

Analysis of 31 other C. coli and C. jejuni strains found that 69% contained a transcribed spacer of 147 bp or 37 bp, also producing fragmented 23S rRNA. Phylogenetic analysis placed Campylobacter in the epsilon subdivision of Proteobacteria based on signatures in the 23S rRNA at positions 270 and 1850.
Cancer and chicken
Peyton Rous discovered the first retrovirus (Rous Sarcoma Virus or RSV) in 1911 that caused tumors in chickens. Studies in the 1950s showed that RSV could transform normal chicken cells into cancer-like cells. In the 1970s, scientists discovered that RSV contained a viral oncogene called v-Src that was derived from the chicken cell's proto-oncogene c-Src. This showed that proto-oncogenes exist in cells and can be activated to become oncogenes, contributing to cancer without a virus. Many human cancers involve the activation of cellular proto-oncogenes like c-Src.
CORONAVIRUS AKA SARS PATENT

Oncogenic viruses
Oncogenic viruses are viruses that can cause cancer. They do this by inducing genetic changes that alter the expression or function of proteins involved in controlling cell growth and division. There are RNA viruses and DNA viruses that are oncogenic. Retroviruses are a type of RNA virus that contain the enzyme reverse transcriptase and can integrate near proto-oncogenes, activating their expression. DNA viruses like papillomavirus and polyomavirus can contain oncogenes that encode transforming proteins to stimulate tumor formation. Many viruses are associated with specific types of cancer in animals and humans.

Candidemia has been identified as a public health problem in HIV-infected patients. The evaluation of CD4 count, transaminases and blood glucose, are being used as a means to monitor the health of HIV-infected patients, without excluding the diagnosis of candidemia and other opportunistic infections. In order to contribute in improving the care of HIV-infected patients attending Dschang District Hospital and later on, in other hospitals in Cameroon, we conducted from June to September 2014 a cross-sectional study, with general objective; to determine the association between candidemia and selected biochemical and haematological parameter changes in HIV-infected patients, as a possible indicator in monitoring HIV disease progression.
To do this, blood samples were collected from HIV-infected patients assigned to the UPEC of Dschang District Hospital for follow up, and haemogram report, CD4 counts, ALAT level, ASAT level, and glucose level in blood were evaluated by cytometric and spectrophotometric assays. Candida species were isolated from some blood samples, and then identified using CHROMagar Candida culture medium. The broth microdilution method was afterwards used to test the susceptibility of the fungal isolates vis-a-vis three conventional antifungal agents.
Mycological analysis of blood samples showed that eight (08) patients had candidemia, a prevalence of 6.11%. Eight (08) isolates were obtained from these eight (08) candidemic HIV-infected patients; this consisted of 4(50%) Candida albicans, 3(37.5%) Candida parapsilosis and 1(12.5%) Candida glabrata. All these isolates were resistant (MICs ranged from 2 to >256 µg/mL) to the antifungals used, that is, ketoconazole, amphotericin B and nystatin.
A significant correlation was found between candidemia and white blood cell count, with a correlation coefficient of r = 0.240 (p < 0.05). Based on the results obtained, the systematic diagnosis of candidemia should be performed in patients infected with HIV in Cameroon in order to improve on their care.

This document describes a study that sequenced the 23S rRNA gene of Campylobacter coli VC167 and analyzed its sequence and structure. It found that the gene contained a 147 bp transcribed spacer that resulted in fragmented 23S rRNA. Analysis of 31 other C. coli and C. jejuni strains found that 69% contained a transcribed spacer of 147 bp or 37 bp, also producing fragmented 23S rRNA. Phylogenetic analysis placed Campylobacter in the epsilon subdivision of Proteobacteria based on signatures in the 23S rRNA at positions 270 and 1850.
Cancer and chicken
Peyton Rous discovered the first retrovirus (Rous Sarcoma Virus or RSV) in 1911 that caused tumors in chickens. Studies in the 1950s showed that RSV could transform normal chicken cells into cancer-like cells. In the 1970s, scientists discovered that RSV contained a viral oncogene called v-Src that was derived from the chicken cell's proto-oncogene c-Src. This showed that proto-oncogenes exist in cells and can be activated to become oncogenes, contributing to cancer without a virus. Many human cancers involve the activation of cellular proto-oncogenes like c-Src.
CORONAVIRUS AKA SARS PATENT
This patent document describes the isolation and characterization of a novel human coronavirus (SARS-CoV) that is the causative agent of severe acute respiratory syndrome (SARS). It provides the nucleic acid sequence of the SARS-CoV genome and the amino acid sequences of its open reading frames. Methods are described for using these molecules to detect SARS-CoV and detect infections. Immune stimulatory compositions are also provided, along with methods for their use.

Oncogenic viruses
Oncogenic viruses are viruses that can cause cancer. They do this by inducing genetic changes that alter the expression or function of proteins involved in controlling cell growth and division. There are RNA viruses and DNA viruses that are oncogenic. Retroviruses are a type of RNA virus that contain the enzyme reverse transcriptase and can integrate near proto-oncogenes, activating their expression. DNA viruses like papillomavirus and polyomavirus can contain oncogenes that encode transforming proteins to stimulate tumor formation. Many viruses are associated with specific types of cancer in animals and humans.
Candidemia in HIV-positive patients in Dschang District Hospital (West Region...
Candidemia has been identified as a public health problem in HIV-infected patients. The evaluation of CD4 count, transaminases and blood glucose, are being used as a means to monitor the health of HIV-infected patients, without excluding the diagnosis of candidemia and other opportunistic infections. In order to contribute in improving the care of HIV-infected patients attending Dschang District Hospital and later on, in other hospitals in Cameroon, we conducted from June to September 2014 a cross-sectional study, with general objective; to determine the association between candidemia and selected biochemical and haematological parameter changes in HIV-infected patients, as a possible indicator in monitoring HIV disease progression.
To do this, blood samples were collected from HIV-infected patients assigned to the UPEC of Dschang District Hospital for follow up, and haemogram report, CD4 counts, ALAT level, ASAT level, and glucose level in blood were evaluated by cytometric and spectrophotometric assays. Candida species were isolated from some blood samples, and then identified using CHROMagar Candida culture medium. The broth microdilution method was afterwards used to test the susceptibility of the fungal isolates vis-a-vis three conventional antifungal agents.
Mycological analysis of blood samples showed that eight (08) patients had candidemia, a prevalence of 6.11%. Eight (08) isolates were obtained from these eight (08) candidemic HIV-infected patients; this consisted of 4(50%) Candida albicans, 3(37.5%) Candida parapsilosis and 1(12.5%) Candida glabrata. All these isolates were resistant (MICs ranged from 2 to >256 µg/mL) to the antifungals used, that is, ketoconazole, amphotericin B and nystatin.
A significant correlation was found between candidemia and white blood cell count, with a correlation coefficient of r = 0.240 (p < 0.05). Based on the results obtained, the systematic diagnosis of candidemia should be performed in patients infected with HIV in Cameroon in order to improve on their care.

It provides simple and intuitive integrated access to cancer genomics data, including copy number, mutation, mRNA and microRNA expression, methylation and protein and phosphoprotein data, on more than 5,000 tumor samples from 20 cancer studies (including 16 TCGA cancer types).
During the past year, we have added network visualization and analysis features to
the cBio Portal. These new features enable researchers to analyze genomic alterations in the context of known biological pathways and interaction networks, and to more easily mine data generated by the TCGA. A network of interest is derived from the Pathway Commons project, based on the query genes specified by the user. Multidimensional genomic data are overlaid onto each node of the network, highlighting the frequency of somatic mutation and copy number alteration (and optionally mRNA up/down-regulation). Users can manage the complexity of the network by filtering by total alteration frequency of genes or by type and source of the interactions. This provides an effective means of managing network complexity, while automatically highlighting those genes most directly relevant to the cancer type in question. In addition, drugs and drug target data can optionally be shown in relation to the network of interest. In this talk, we would like to illustrate the main network analysis features using data from the TCGA project. We will also discuss our future plans for the network view.

This document summarizes efforts by the Genome in a Bottle Consortium to develop an integrated set of genotype calls for the NA12878 reference sample using data from 14 sequencing datasets. They identify characteristics of bias and errors in the different datasets to arbitrate between discordant variant calls. The resulting high-confidence SNP and indel calls are being used as a benchmark to evaluate the performance of sequencing and analysis methods. The integrated calls appear to have higher sensitivity than previous microarray-based assessments. The consortium is exploring expanding this approach to structural variants and using pedigree relationships.

ESR spectroscopy is the most sophisticated way to investigate the quality of the food and the free radicals induced during the processing of the food. ESR spin trapping technique is useful for the detection of highly unstable radicals in the food. The antioxidant capability of liquid food and beverages in mainly performed by spin trapping technique.

Nucleophilic addition is the most important reaction of carbonyls. Not just aldehydes and ketones, but also carboxylic acid derivatives in general.
Carbonyls undergo addition reactions with a large range of nucleophiles.
Comparing the relative basicity of the nucleophile and the product is extremely helpful in determining how reversible the addition reaction is. Reactions with Grignards and hydrides are irreversible. Reactions with weak bases like halides and carboxylates generally don’t happen.
Electronic effects (inductive effects, electron donation) have a large impact on reactivity.
Large groups adjacent to the carbonyl will slow the rate of reaction.
Neutral nucleophiles can also add to carbonyls, although their additions are generally slower and more reversible. Acid catalysis is sometimes employed to increase the rate of addition.

Although Artemia has been known to man for centuries, its use as a food for the culture of larval organisms apparently began only in the 1930s, when several investigators found that it made an excellent food for newly hatched fish larvae (Litvinenko et al., 2023). As aquaculture developed in the 1960s and ‘70s, the use of Artemia also became more widespread, due both to its convenience and to its nutritional value for larval organisms (Arenas-Pardo et al., 2024). The fact that Artemia dormant cysts can be stored for long periods in cans, and then used as an off-the-shelf food requiring only 24 h of incubation makes them the most convenient, least labor-intensive, live food available for aquaculture (Sorgeloos & Roubach, 2021). The nutritional value of Artemia, especially for marine organisms, is not constant, but varies both geographically and temporally. During the last decade, however, both the causes of Artemia nutritional variability and methods to improve poorquality Artemia have been identified (Loufi et al., 2024).
Brine shrimp (Artemia spp.) are used in marine aquaculture worldwide. Annually, more than 2,000 metric tons of dry cysts are used for cultivation of fish, crustacean, and shellfish larva. Brine shrimp are important to aquaculture because newly hatched brine shrimp nauplii (larvae) provide a food source for many fish fry (Mozanzadeh et al., 2021). Culture and harvesting of brine shrimp eggs represents another aspect of the aquaculture industry. Nauplii and metanauplii of Artemia, commonly known as brine shrimp, play a crucial role in aquaculture due to their nutritional value and suitability as live feed for many aquatic species, particularly in larval stages (Sorgeloos & Roubach, 2021).

When I was asked to give a companion lecture in support of ‘The Philosophy of Science’ (https://shorturl.at/4pUXz) I decided not to walk through the detail of the many methodologies in order of use. Instead, I chose to employ a long standing, and ongoing, scientific development as an exemplar. And so, I chose the ever evolving story of Thermodynamics as a scientific investigation at its best.
Conducted over a period of >200 years, Thermodynamics R&D, and application, benefitted from the highest levels of professionalism, collaboration, and technical thoroughness. New layers of application, methodology, and practice were made possible by the progressive advance of technology. In turn, this has seen measurement and modelling accuracy continually improved at a micro and macro level.
Perhaps most importantly, Thermodynamics rapidly became a primary tool in the advance of applied science/engineering/technology, spanning micro-tech, to aerospace and cosmology. I can think of no better a story to illustrate the breadth of scientific methodologies and applications at their best.
