This document provides an introduction to genome-wide association studies (GWAS) and their role in discovering the genetic basis of complex traits and diseases. It explains that GWAS are a hypothesis-free approach to searching the entire human genome for genetic variants associated with a trait using common single nucleotide polymorphisms. Over 2,000 traits and diseases have been studied through GWAS, identifying over 15,000 genetic associations. The document traces the development of GWAS from early human genome sequencing and projects to characterize human genetic variation to the availability of high-throughput genotyping arrays that enabled widespread application of GWAS in disease research.

Quantile regression is an extension of linear regression that relates specific quantiles (percentiles) of the target variable to the predictor variables rather than just the mean. It makes fewer assumptions than ordinary least squares regression about the distribution of the target variable and is more robust to outliers. Quantile regression can provide a more complete picture of the relationship between variables by examining how predictors influence different parts of the conditional distribution.

MAPS2018 Keynote address on EY report: Life Sciences 4.0 – Securing value thr...EY
Summary: This keynote address presented by Pamela Spence, EY Global Life Sciences Leader (pspence2@uk.ey.com) at MAPS 2018 – the annual meeting for Medical Affairs Professional Society – discusses our latest life sciences report and the industry demands for a customer-focused, data driven approach to health care. We describe the accelerating pace of change as technological advances and the escalating expectations of payers, physicians and patient consumers are combining to disrupt the life sciences business model. Data and algorithms that maximize health outcomes based on individual needs and preferences are becoming the ultimate health care consumable. To create value now and in a future that we call Life Sciences 4.0, life sciences companies must build – or participate in – interoperable information systems that collect, combine and share data. For ten years or more, China has been a uniquely powerful engine of the global economy, regularly posting high single-figure or even double-digit annual increases in GDP. More recently, growth has slowed, prompting sharp falls in international commodity prices and casting a shadow over the near-term prospects for developed and emerging markets.
What will happen next? Pessimists struggle to see what China can do for an encore after what they say was an extraordinary, one-off period of catching up. Optimists believe that during the next 10 to 15 years, China has the potential to continue to outperform the rest of the world and to take its place as a full-fledged advanced economy (see summary infographic, “What’s next for China?”).

Generic drugs can be approved through an Abbreviated New Drug Application (ANDA) which relies on the safety and efficacy data of the branded drug. The ANDA process requires generic manufacturers to show bioequivalence to the branded drug through bioavailability and bioequivalence studies rather than completing full clinical trials. If bioequivalence is established, it demonstrates that the generic drug delivers the same amount of active ingredients into a patient's bloodstream in the same amount of time as the branded drug. The Hatch-Waxman Act established the modern ANDA approval process and aims to balance promoting generic drugs to reduce costs while also compensating branded manufacturers for regulatory time lost from patents.

The document discusses options for evolving the format of the National Drug Code (NDC) to improve interoperability in the US drug supply chain. It presents four options - making no changes (Option A), adding a future date (Option B), zero padding the 11-digit NDC (Option C), or adopting a harmonized 12-digit format (Option D). Option D may provide the greatest interoperability but also be the most disruptive. The NDC identifier could also be enhanced to include additional designations for controlled substances, hazardous materials, and unit dosing. Any changes require consideration of technical complexity and costs across the healthcare system.

Local Advanced Lung Cancer: Artificial Intelligence, Synergetics, Complex Sys...Oleg Kshivets
Overall life span (LS) was 1671.7±1721.6 days and cumulative 5YS reached 62.4%, 10 years – 50.4%, 20 years – 44.6%. 94 LCP lived more than 5 years without cancer (LS=2958.6±1723.6 days), 22 – more than 10 years (LS=5571±1841.8 days). 67 LCP died because of LC (LS=471.9±344 days). AT significantly improved 5YS (68% vs. 53.7%) (P=0.028 by log-rank test). Cox modeling displayed that 5YS of LCP significantly depended on: N0-N12, T3-4, blood cell circuit, cell ratio factors (ratio between cancer cells-CC and blood cells subpopulations), LC cell dynamics, recalcification time, heparin tolerance, prothrombin index, protein, AT, procedure type (P=0.000-0.031). Neural networks, genetic algorithm selection and bootstrap simulation revealed relationships between 5YS and N0-12 (rank=1), thrombocytes/CC (rank=2), segmented neutrophils/CC (3), eosinophils/CC (4), erythrocytes/CC (5), healthy cells/CC (6), lymphocytes/CC (7), stick neutrophils/CC (8), leucocytes/CC (9), monocytes/CC (10). Correct prediction of 5YS was 100% by neural networks computing (error=0.000; area under ROC curve=1.0).

We have a number of quality assurance processes in line with UNFPA policy for all the product categories. In 2011 in consultation with WHO, and various other international procurement agencies, UNFPA developed a QA policy for RH medicines. The criteria is we only procure hormonal contraceptives and uterotonics (oxytocin, misoprostol) from WHO prequalified sources or SRA approved sources or those that have undergone the WHO Expert Review Panel process and have been recommended for procurement. The criteria for other essential medicines stipulate a thorough review by a committee of experts focusing on all the safety, efficacy and quality parameters in the dossiers and the annexes of an interagency pharmaceutical product questionnaire which essentially requires clinical data as per drug technical files. UNFPA procures medical equipment, kits and surgical equipment. There are a set of technical requirements for manufacturers and the products which then go through a detailed technical evaluation for the specifications, performance parameters, quality certifications as well as other regulatory requirements. The evaluation also includes review/inspection and testing of samples. A critical component is the development of guidelines, SOPs to refine the processes and harmonize standards and procedures In an effort to harmonize and build better understanding among stakeholders and streamline processes, capacity building activities are ongoing with manufacturers, quality control laboratories, regulatory authorities and other partner agencies
The RHI consolidatesharmonized data from multiple sources, and organizesit in the 6 different web reports seen on the right-hand side. The websiteoffers a detailed report in whichyoucansee the specifics of a givenshipment, a report thatlists all shipmentsgoing to a given country and severalsummary reports. There are alsographing and mapping options.Theshipmenthistory and shipmentdetails reportsrequire registration, whichcanbeeasilydone online.
We continue to see improvement in 53mm Std Male Condom Wait Time Savings. This is due to refinement of Inventory Procedures which has reduced wait times to average of 2.5 weeks during 2012 (vs. Average of 3 weeks during 2011), plus increased lead times from Fresh Production (Average 2012: 17 weeks, Average 2011: 14 weeks).
