INTRODUCTION
Plant tissue culture (PTC) is a form of asexual propagation
of plants under laboratory conditions.
Plant tissue culture is a technique with which plant cells,
tissues or organs are grown on artificial nutrient medium,
either static or liquid, under aseptic and controlled
conditions.
3
HISTORY
1902 The idea of the totipotency of plant cell was given by
Haberlandt
1937 White first time established successful root culture of
tomato
1941 Vanoverbeek used coconut milk for growth and
development of young Datura embryos
1957 Skoog and Miller demonstrated the role of auxin and
cytokinin on root and shoot formation in tobacco – tissue
1962 Murashige and Skoog introduced the medium for tobacco
culture
1987 Isolation of Bt. gene form bacterium Bacillus thuringiensis
4
PRINCIPLE
PTC depends upon
1. Totipotency :- It is the ability of plant cells to
regenerate into a whole plant
2. Plasticity :- It is the ability of plants to alter their
metabolism, growth and development to best suit
their environment
5
TYPES OF PTC
Plant tissue culture
Embryo culture
Seed culture
Meristem culture
Cell culture Protoplast culture
Callus culture
Pollen culture
Organ culture
6
7
8
Protoplast Fusion
Application of PTC
Meristem culture
1. Production of virus free germplasm.
2. Mass production of desirable genotypes.
9
Anther culture/Pollen culture
1. Production of haploid plant
2. Production of useful gametoclonal variation.
10
Other Applications
1. Micropropagation can also be used to conserve rare or
endangered plant species
2. Micropropagation using meristem and shoot culture to
produce large numbers of identical individuals.
3. Large-scale growth of plant cells in liquid culture in
bioreactors for production of valuable compounds
4. To cross distantly related species by protoplast fusion and
regeneration of the novel hybrid
5. Production of identical sterile hybrid species can be
obtained.
