N-Heterocyclic carbene promoted
decarboxylation of lignin-derived aromatic acids
Background
• Lignin remains very difficult to efficiently convert into biofuels
and bioproducts
• N-heterocyclic carbenes (NHCs) are an intriguing class of
catalysts for lignin decarboxylation due to mild conditions
and high product selectivity through the strong
nucleophilicity and basicity of NHCs
• Certain ionic liquids have the potential to be effective NHCs
Approach
• Used,the ionic liquid (1-ethyl-3-methylimidazolium acetate,
[C2C1Im][OAc]) as an air- and moisture-stable carbene
precursor for the decarboxylation of cinnamic acid
derivatives of lignin
• NHC generated by the deprotonation of [C2C1Im][OAc]
Outcomes and Impacts
• NMR experiments indicate a “clean” decarboxylation
reaction occurs, with no degradation or polymerization of
final products observed
• Decarboxylation generated quantitative yields of 2-methoxy-
4-vinylphenol from the decarboxylation of ferulic acid (and
other substrates with similar functionality)
• Excellent recyclability of [C2C1Im][OAc] demonstrated using
a biphasic reaction system with ethyl acetate as the organic
phase

Background
• Drought stress is a major obstacle to crop productivity, and the severity and
frequency of drought are expected to increase in the coming century.
• Certain root-associated bacteria have been shown to mitigate the negative
effects of drought stress on plant growth, and manipulation of the crop
microbiome is an emerging strategy for overcoming drought stress in
agricultural systems, yet the effect of drought on the development of the root
microbiome is poorly understood.
Approach
• Two varieties of sorghum were grown in the field in California and sampled
weekly during the growing season.
• Preflowering drought, postflowering drought and irrigated control treatments
were compared.
• 16S rRNA amplicon and metatranscriptome sequencing, as well as root
metabolomics, was used to assess the response of the microbiome
composition to drought.
Outcomes and Impacts
• Drought delays the development of the early sorghum root microbiome and
causes increased abundance and activity of monoderm bacteria, which lack
an outer cell membrane and contain thick cell walls.
• Altered plant metabolism and increased activity of bacterial ABC transporter
genes are correlated with shifts in community composition.
• Plants accumulate several metabolites in response to drought, e.g. with
glycerol-3-phosphate and proline showing 3000-20000 fold increase.
• Inoculation experiments with monoderm isolates indicate that increased
colonization of the root during drought can positively impact plant growth.
• Collectively, the results demonstrate the role that drought plays in
restructuring the root microbiome and highlight the importance of temporal
sampling when studying plant-associated microbiomes.

N-Heterocyclic carbene promoted decarboxylation of lignin-derived aromatic acids Background • Lignin remains very difficult to efficiently convert into biofuels and bioproducts • N-heterocyclic carbenes (NHCs) are an intriguing class of catalysts for lignin decarboxylation due to mild conditions and high product selectivity through the strong nucleophilicity and basicity of NHCs • Certain ionic liquids have the potential to be effective NHCs Approach • Used,the ionic liquid (1-ethyl-3-methylimidazolium acetate, [C2C1Im][OAc]) as an air- and moisture-stable carbene precursor for the decarboxylation of cinnamic acid derivatives of lignin • NHC generated by the deprotonation of [C2C1Im][OAc] Outcomes and Impacts • NMR experiments indicate a “clean” decarboxylation reaction occurs, with no degradation or polymerization of final products observed • Decarboxylation generated quantitative yields of 2-methoxy- 4-vinylphenol from the decarboxylation of ferulic acid (and other substrates with similar functionality) • Excellent recyclability of [C2C1Im][OAc] demonstrated using a biphasic reaction system with ethyl acetate as the organic phase Liu et al. Drought delays development of the sorghum root microbiome and enriches for monoderm bacteria Background • Drought stress is a major obstacle to crop productivity, and the severity and frequency of drought are expected to increase in the coming century. • Certain root-associated bacteria have been shown to mitigate the negative effects of drought stress on plant growth, and manipulation of the crop microbiome is an emerging strategy for overcoming drought stress in agricultural systems, yet the effect of drought on the development of the root microbiome is poorly understood. Approach • Two varieties of sorghum were grown in the field in California and sampled weekly during the growing season. • Preflowering drought, postflowering drought and irrigated control treatments were compared. • 16S rRNA amplicon and metatranscriptome sequencing, as well as root metabolomics, was used to assess the response of the microbiome composition to drought. Outcomes and Impacts • Drought delays the development of the early sorghum root microbiome and causes increased abundance and activity of monoderm bacteria, which lack an outer cell membrane and contain thick cell walls. • Altered plant metabolism and increased activity of bacterial ABC transporter genes are correlated with shifts in community composition. • Plants accumulate several metabolites in response to drought, e.g. with glycerol-3-phosphate and proline showing 3000-20000 fold increase. • Inoculation experiments with monoderm isolates indicate that increased colonization of the root during drought can positively impact plant growth. • Collectively, the results demonstrate the role that drought plays in restructuring the root microbiome and highlight the importance of temporal sampling when studying plant-associated microbiomes.
