Proteins are responsible for things like enzymes, antibodies, structure, transport, storage, and
messaging. In order for proteins to function properly and provide these functions in the body,
deoxyribonucleic acid is (DNA) required. DNA provides ribonucleic acid (RNA) that is translated into
a certain protein. These concepts lay the foundation for labs 5 through 7.
In lab 5, a series of steps were taken that allowed us to obtain purified genomic DNA. Polymerase
chain reaction (PCR) was utilized in order to acquire the specific gene that encodes for the protein
adhP. PCR works by using different temperatures and properties of DNA replication (i.e. DNA

DNA is a negative molecule so it will move towards our anode (positive end) as the small fragments
will move faster and the large ones slower. With a DNA ladder, we could compare the size of the gene
we needed to the marker with a known size. TOPO cloning along with transformation were then
carried out. With help from DNA topoisomerase, TOPO cloning uses our gene from the PCR reaction
and places it within our vector (pET101/D TOPO) via directional cloning. Directional cloning allows

During the month of November, I became our school s acting testing coordinator. Our testing
coordinator was in an accident and went out on medical leave. She was suddenly out for an
undetermined amount of time and with a severe concussion was unable to remember what she
completed and what she did not. During her absence, there were two assessments and a survey that
needed to me completed. These assessments included SAT 10, FFT (Teacher Survey) and MAP R.
This was a very demanding experience. The first thing that needed to be completed was the FFT
Teacher Survey. This is important to all teachers on cycle using the Framework for Teaching. The
Survey is a vital component to a teachers overall effectiveness. If left incomplete, the teacher receives
a score of zero for that portion. When I became the acting testing coordinator, only two weeks were
left in the window and no surveys were completed. Within two weeks, I needed to complete 167
surveys for 7 teachers. Unfortunately, the teachers could not assist in this endeavor. I tried to create a
schedule were I was able to conduct all 167 surveys. This was impossible if I was still going to meet
with my intervention students. Therefore, I rounded up a team of individuals that had opportunity and
the willingness to assist. This team consisted of the ESOL teacher and the Guidance Counselor. The

change but rather a failure of building a relationship between the management team and the team
members of the organisation. Thus it is the management team responsible as well for the arising of
resistance to change as it is an active participation of the process.
A change is the process of changing the old way or usual way of doing thing and implements a new
way or different path of doing thing and thus change could create fear of the unknown for the
participants (Mintzberg Waters, 1985)6. In order to make change happen, it is important for both
parties, the change recipient and change agent to actively seeking information, feedback, goal and
objective of change. The key word here would be Communication . By being honest to the recipient,
letting them known of the expectation from the change and the beneficial likeliness that change would
bring to personal gain and to the organisation, this would encourage the participant to adopt change
process (Kim Rousseau, 2006)7.

Frankenstein and Gothic Literature To define gothic literature, someone might state that this genre
includes mystery and horror in writing with characters fighting for power over one another. The
stories written in this way typically include strange and mysterious locations that characters find
themselves in as well as horrific situations. The plot might include threatening and dangerous
circumstances that evoke fear out of the readers themselves. After comparing a definition of gothic
literature to the story Frankenstein, the plot aligns perfectly with the explanation.

Ingolstadt represents the unknown in certain ways. For example, it is the place that doctor
Frankenstein spends most his young adulthood studying and creating the creature. This place is where
the most horrific situations begin including the abandonment of his family, search for knowledge, and
creation of the most dangerous human like being. In contrast, Geneva represents some sort of safe
haven for doctor Frankenstein. In the beginning of the novel, this location is where he grew up and
had a fairly normal childhood. In contrast, it is where he flees to forget about his blunder and removes
himself from the depressing situation. Inevitably, Geneva transforms into a dark place after the murder
of Frankenstein s youngest brother. This once again shows the mystery side of the story, and readers
could only wonder if the ugly creature had anything to do with it. Likewise, the forest the organism
spends his time in after his abandonment shows similar characteristics of mystery, but differs by
adding the aspect of loneliness. The creature could never understand why the majority of people were
afraid of him and shamed him. Therefore, he decides to reside in a cold and lonely forest, living in a
small hut. Eventually, Frankenstein finds his creature in these woods and they meet again and discuss
the life he has lived, observing the family that lives by him, learning and developing language and
social skills, and trying to survive. Here, doctor Frankenstein begins to truly understand what he had
created and he realizes that this creature only longed for love and to be loved after his struggle to fit

In the poem, Lynching Song , a black man is being hanged by a group of white men. And although he
might have died, he states that the whit men are the ones who are really dead. The reader captures the
true meaning of the poem through the formal features that are being presented. In the five stanza
poem, there are four lines in the first two stanzas; three in the next two, and the last stanza only has
one line. Not all the lines are the same beat either, in the first stanza the first two lines have four beats
and the next two lines have five beats. In the second stanza, the first line has three beats and the
