This document compares two analytical methods, photometric and HPLC-ELSD, for determining components in Tribulus terrestris, particularly the steroidal saponins protodioscin and protogracillin. The photometric method is non-selective and can give misleading results by responding to other compounds, while HPLC-ELSD is more selective but degradation can occur during extraction. Analysis of samples from different geographical origins showed wide phytochemical variation detectable by HPLC-ELSD but not the photometric method. HPLC-ELSD is recommended for accurate analysis of Tribulus terrestris products.
