1) AAS is a technique used to determine the concentration of chemical elements in samples by measuring light absorption by free atoms. It can analyze over 62 elements and is commonly used in pharmaceutical, food, and environmental applications.
2) The basic components of an AAS instrument are a hollow cathode lamp, monochromator, atomizer, detector, and nebulizer. Samples are atomized in a flame or graphite furnace then irradiated to cause absorption of specific wavelengths that are measured.
3) AAS is based on the principle that free atoms generated from a sample can absorb radiation at specific frequencies, allowing quantification of elemental
This document discusses headspace analysis, which analyzes the gas above a sample in a chromatography vial. Headspace analysis is suited for very volatile analytes in solid or liquid samples, such as residual solvents in pharmaceuticals, flavors in beverages, and alcohol in blood or urine. It has advantages like minimal sample preparation and easy automation. Key factors that affect headspace analysis efficiency are the partition coefficient (K value) and phase ratio (b value), both of which can be optimized based on temperature, salts added, sample volume, and vial design. A typical headspace analyzer automatically analyzes up to 10 vials using a motorized needle and can be connected to a GC instrument. Headspace analysis is commonly used

Atomic Absorption spectrometer is an instrument used for quantitative analysis of most of the metals in nano grams. This is highly sensitive technique used for analysis.
Gas chromatography is a technique used to separate and analyze volatile compounds. It works by injecting a sample into a column through which an inert gas flows, carrying the separated components out at different rates depending on their interactions with the stationary phase coating the column. The separated components are detected to produce a chromatogram showing peaks that can be analyzed to determine the identity and quantity of each component in the original sample.
Bioanalysis of drugs from biological samples involves three key steps: sample collection, sample preparation, and detection methods. Common biological samples include blood, plasma, serum, urine, and tissues. Sample preparation techniques aim to remove interfering matrix components, concentrate the analyte, and prepare the sample for detection. Common techniques include protein precipitation, liquid-liquid extraction, and solid phase extraction. The goal is to isolate and concentrate the drug or metabolite for accurate quantitative analysis.

Thin layer chromatography is a technique used to separate mixtures into their components. It involves a stationary phase, such as silica gel, and a mobile phase, such as a solvent. A sample is applied to the plate and the mobile phase moves up the plate, separating the components by their interaction with the stationary phase. The separated components are then detected and quantified. TLC is useful for analyzing mixtures and is simpler and less expensive than other chromatography methods.
Capillary electrophoresis principles and applications Indira Shastry
Capillary electrophoresis is a technique used to separate charged molecules like proteins, nucleic acids, and other small molecules based on their electrophoretic mobility. It has several advantages over traditional gel electrophoresis methods like faster separation, higher resolution, and requiring only small sample volumes. In capillary electrophoresis, samples are injected into a thin, fused silica capillary tube and separated under the influence of an applied electric field based on differences in their charge and size. Key applications of capillary electrophoresis include hemoglobin electrophoresis for detecting abnormal hemoglobins, serum protein electrophoresis, and DNA sequencing. It provides a rapid, automated, and high-resolution method for analyzing biomolecules.
This document provides an overview of electrophoresis techniques presented by Miss Sayanti Sau. It discusses the basic principles of electrophoresis and defines different types including zone electrophoresis techniques like paper, gel, thin layer, and cellulose acetate electrophoresis. It also covers moving boundary electrophoresis techniques such as capillary electrophoresis, isotachophoresis, and isoelectric focusing. Details are provided on gel electrophoresis methods including agarose, polyacrylamide, and SDS-PAGE. Applications and advantages of various electrophoresis techniques are highlighted.
Atomic Absorption Spectroscopy uses the principle that free atoms generated from a sample can absorb radiation at specific frequencies, allowing the technique to quantify the concentration of various metals and metalloids present. The sample is atomized using a flame or graphite furnace and exposed to light from a hollow cathode lamp, with absorption measured to generate calibration curves and determine unknown concentrations. This document discusses the classification and control of elemental impurities in pharmaceutical products. It divides elements into three classes based on their toxicity and likelihood of occurrence in drugs. Class 1 elements are highly toxic and should be evaluated across all potential sources. Class 2 elements are route-dependent toxins divided into 2A and 2B based on probability of occurrence. Class 3 elements have relatively low oral toxicity but may warrant consideration for inhalation and injectable routes. The document also describes establishing permitted daily exposure limits, applying a risk-based approach to control impurities, and maintaining an overall control strategy throughout a product's lifecycle.
Gas chromatography is a technique used to separate and analyze compounds that can be vaporized. It works by partitioning samples between a gas mobile phase and liquid stationary phase in a column. The separation depends on how the compounds partition between the phases. Key components of a GC system include the carrier gas, sample introduction system, column for separation, and detection system such as FID or TCD. Common applications include analysis of aromatics, hydrocarbons, flavors, and other volatile organic compounds.
