Bizim çalışma amacımız bazı sentetik ve doğal sekonder metabolitlerin meme kanserinde antiproteazomal aktivitesinin taranmasıdır. Asteraceae familyasından olan Centaurea urvillei DC. subsp. Urvillei ’den izole edilen 14 madde ve N-(sübstitüe)-fenil-2-(açilamino)asetamid, propanamit ve sülfonamit grubu 14 sentetik bileşik çalışmada kullanılmıştır.
Çalışma sonucunda özellikle bitkisel metabolitlerde aktivite gözlenmiş ve kamferol ve apigenin’in meme kanserinde apoptozisi indüklediği tespit edilmiştir. Bu da bu bileşiklerin antiproteazomal aktivite yoluyla meme kanserinde umut verici ilaç aday molekülleri olabileceğini göstermiştir.
İleride yapılacak çalışmalar ile, söz konusu bileşiklerin antiproteazomal etkilerinin meme kanserine spesifik olup olmadığı, diğer kanserlerdeki etkinliği incelenerek tespit edilmeye çalışılacaktır. Tiyazolidindionlar ilk olarak 1970’li yılların sonuna doğru keşfedilmiştir. Çeşitli türevler
geliştirildikten sonra 3 molekül; troglitazon, rosiglitazon, pioglitazon klinik kullanım için onay
almıştır.
Piyasaya ilk çıkan tiyazolidindion troglitazon olmasına rağmen ender de olsa hepatik yetmezlik
ve ölüme ilerleyebilecek idiosinkratik karaciğer toksisitesi gelişimi ile ilişkisi bulunması üzerine
2000 yılında kullanımdan çekilmiştir.
Rosiglitazon ise tip 2 diyabetik hastalarda kalp krizi ve inme gibi kardiyovasküler olay riskini

Transjenik fare modelleri ile yapılan bir
çalışmada, PPARβ/δ VE PPARγ gen
ekspresyonunun eliminasyonu ile akciğer
kanserinin şiddetlenmesi arasında bir ilişki
bulunmuştur.
PPARγ aktivatörlerinin, hücrelerin
farklılaşma ve apoptozisinde rol oynadığı
anlaşılmıştır.
Diyabet tedavisinde tiyazolidindion kullanan
hastalarda akciğer kanseri gelişme riskinin,
başka bir tedavi alan hastalara oranla
anlamlı şekilde daha az olduğu
gösterilmiştir.

akciğer kanserinde hücre çoğalmasını
inhibe ederek etki göstermektedir.
Akciğer kanserinde hücre gelişiminde rol

uyarıldığı öne sürülmüstür. Gerçekten de
yakın zamanda yapılan bir çalışmada,
selektif PPARβ/δ agonisti olan GW501516

gösterilmiştir.
İnsanlardan elde edilen veriler; PPARγ’nın kolon kanserinde
baskılayıcı bir role sahip olduğunu ve bu reseptörün düşük
seviyelerdeki ekspresyonunun kolon kanseri riskini
arttırabileceğini göstermiştir.
Ancak hayvan modelleriyle yapılan bazı invivo çalışmalar

farelerde PPARγ aktivasyonunun pankreas

Bu bulguların tam aksine PPARγ
ekspresyonundaki artış hasta sağ kalımındaki
azalma ile ilişkili bulunmuş ve PPARγ’nın tümör
gelişimi ve ilerlemesinden sorumlu olduğu ortaya
konmuştur.
Türler arası PPARγ farklılıkları bu paradoksu
açıklamak için olası bir seçenek olabilir.
27. ÜRİNER SİSTEM
KANSERLERİ VE PPAR
PPARγ ekspresyonunun artışın, mesane kanserinin derecesini ve
aşamasını arttırdığı iddia edilmiştir.
PPARγ ve PPARα reseptörlerinin ikisine de afinite gösteren bazı
dual agonistlerle yapılan çalışmalarda; bu bileşiklerin fare ve
sıçanlarda mesane ve üretelyumda karsinojenik etkilerinin
bulunması, bu ilaçların klinik kullanımı ile ilgili güvenlik
sorunlarını arttırmıştır
Ancak yapılan başka bir çalışma PPARγ’nın normal üretelyumda
ekspresse edildiğini ve tümör progresyonunu azalmasıyla ilişkili
olduğunu iddia etmektedir.
Metilen yerine diindolmetan içeren yeni sınıf PPARγ agonistlerin
mesane kanserine karşı önceki kuşaklara kıyasla daha güçlü
antikanser etkiye sahip olduğu gösterilmiştir.

hattında, T0070907’nin agonistlere kıyasla daha
güçlü antiproliferatif etkiye sahip olduğu
gösterilmiştir.
Yemek borusu kanseri hücre hattında anti invaziv ve
antiproliferatif etkileri kanıtlanmıştır.
MM (Multiple Myeloma) hücre hattına diğer kanser
