The document describes a protocol for shearing DNA molecules to an appropriate size for next-generation sequencing library preparation using the Bioruptor. The protocol shears genomic DNA or cDNA from Arabidopsis thaliana into fragments around 200-600 nucleotides in length. This is done by placing the DNA sample in the Bioruptor and running cycles of 30 seconds on and 2 minutes off for a total of 3 cycles. The sheared DNA fragments can then be directly used to make libraries for next-generation sequencing technologies such as ChIP-seq or RNA-seq.
