Effect of time and proportion of leaf harvest on pest, forage and root yields...Agriculture Journal IJOEAR
Dearth of knowledge exists regarding the leaf harvest intensity and frequency thresholds that support optimum forage and fresh storage root yields in Sierra Leone. A study was carried out to assess the effects of leaf harvesting time and proportion on Cylas puncticollis infestation, growth and yield of sweet potato in the inland valley swamp and upland ecologies of Njala. Treatment combinations comprised of two varieties ("Kabia" and "Gbanie"), four leaf harvest regimes: 0, 30 60 and 90 days after planting (DAP); and four-leaf harvest intensities (0, 25, 50 and 100%). The experiment was laid out in a randomized complete block design (RCBD) with three replications. Data collected included Cylas puncticollis severity on vines and storage roots, root dimensions and numbers, fresh foliage and storage root yields. The results revealed that leaf harvesting twice at 25 and 50% contributed more to optimum forage and storage root yields and related attributes of sweet potatoes compared to other treatments. The present study suggests that good agronomic management of sweet potato that supports optimum forage and storage root yields should be selected to meet the dual purpose for which it is grown. These findings serve as good guide for incorporation of leaf harvesting time, proportion of leaf harvest in germplasm assessment and new population development objectives.
Organogenic Regeneration of an Elite Cultivar of Chinese Jujube (Zizyphus juj...Agriculture Journal IJOEAR
Abstract— An efficient and relatively simple regeneration system was developed for an elite cultivar of Chinese Jujube, a perennial tree, by culturing young twig segments as explants from 8-15 year old trees. The twig segments were disinfected by submerging them in 1% sodium hypochlorite (NaOCl) for 15 min with 3 min vacuum. Calli developed from both ends of the twig segments on half-strength MS medium supplemented with sucrose and BA or BA and NAA in combination. The frequency of shoot formation from calli was higher than 80% when the explants were placed on the half - strength MS medium supplemented with BA (2.581 μM) and NAA (2.685 μM). Roots were produced from adventitious buds for 90% of the regenerated shoots when they were placed on the MS medium supplemented with 4.920 μM IBA and 5.708 μM IAA. After transplanting to soil, 82% of the regenerated seedlings survived when they were covered with glass containers to maintain humidity. The results suggest that Chinese jujube can be reproduced and multiplied using organogenesis with the appropriate explant and culture medium.

Effect of Trichoderma Fortified Compost on Disease Suppression, Growth and Yi...IJEAB
Trichoderma species are commonly used as effective biological control agents against phytopathogens especially the soil-borne fungi while some isolates are able to ameliorate plant growth. In the present study, Trichoderma fortified compost with different substrates were evaluated to reduce the pre-emergence and post-emergence seedling mortality, diseases of stem and root of chickpea caused by several soil-borne fungal pathogens, including Fusarium oxysporum, Rhizoctonia solani and Sclerotium rolfsii at different growth stages in the field under natural epiphytotic conditions. Among the twenty isolates of T. harzianum, Co-7 showed the most effective antagonist against the test pathogens in dual culture method. In field experiment, subsequently it was used for inoculum preparation with colonized wheat grain and mixed with well-matured decomposed composting materials like, saw dust, cow dung, tea waste, water hyacinth and poultry manure. Trichoderma fortified compost with poultry manure was found significantly effective in reducing pre-emergence and post emergence seedling mortality, disease incidence and disease severity of chickpea in the field. Interestingly, all the treatments significantly increased but Trichoderma fortified compost with poultry manure was the best to boost seed yield and quality.
ABSTRACT- The present study was conducted to evaluate the effectiveness of thermotherapy to inactivate Potato leaf
roll virus (PLRV) from the potato tubers. For this purpose an experiment was carried out at Newly Developmental Farms
(NDF) of the University of Agriculture, Peshawar Pakistan. Potato tubers infected with PLRV were collected from
farmer’s fields. The potato tubers were than treated with hot water at average 370C for various intervals of time.
Afterwards these heat treated tubers were shifted to fields for sowing. In field condition minimum % incidence (16.66%)
of PLRV was observed from the treatments T3 (2 hours hot water treatment), T4 (2 ½ hours hot water treatment) and T5
(3 hours hot water treatment) respectively while in control 53.33 % incidence of PLRV was recorded. Therefore it can be
concluded that thermotherapy at 370C for 2 hours, 2 ½ hours and 3 hours in case of hot water treatment were effective in
fully or partially elimination of PLRV from potato tubers. Further combine effect of thermotherapy, confidor and neem
extract was evaluated against PLRV. It was observed that in T6 (hot water treatment for 2 ½ hours, insecticide and
biocide) % incidence of PLRV was 13.2% with maximum vegetative parameters such as % germination, height (cm),
tuber size (cm) and yield (kg) recorded followed by T4 (Confidor + 2½ hours hot water treatment) and T1 (2 ½ hours hot
water treatment) where % incidence of PLRV was 16.66% and 20% respectively. Moreover the treatment T2 (Confidor)
when applied individually was found to more effective against PLRV as compared to T3 (Neem extract) with % incidence
value 26.66% and 33.33% respectively.

ABSTRACT- The present study describes the hormonal regulation on morphogenesis in vitro in nodal segments of T. indica. The nodal explants, sterilized with 0.1% HgCl2, were cultured on Murashige and Skoog Medium enriched with various combinations and concentrations of plant hormones auxin and cytokinin to study the hormonal regulation on morphogenesis in vitro in T. indica. BAP at high concentration could not evoke any morphogenetic response in nodal explants. Calli formation at the basal part of nodal explants were noted on medium containing BAP (0.1 mg/L) and 2,4-D (5.0 mg/L). 0.1 mg/L BAP was found most effective in the shoot development of the T. indica. Rhizogenesis was observed on half-strength MS medium supplemented with 1.0 mg/l IAA and 0.1 mg/l NAA. The study may also be used mass-propagation and conservation of this medicinal plant species.

Current control strategy focuses on use of synthetic pesticides that consequently lead to decimation of natural enemies, development of insect resistance and resurgence and upset biodiversity. Botanical pesticides have been used widely in smallholder farmers but not much documented literature exists on efficacy of these products. A field trial was done to assess the efficacy of crude aqueous extracts of Tephrosia vogelii, Allium sativum and Solanum incanum in controlling Brevicoryne brassicae in Brassica napus production. The trial was laid in a randomized complete block design (RCBD) with five treatments replicated four times. The five treatments used in the experiment were T. vogelii, A. sativum, S. incanum, dimethoate and control. Wingless adult female aphids were inoculated three weeks after transplanting of seedlings. Spraying and data collection were done weekly for four weeks. Data was collected on aphid nymph and adult counts on the third leaf from the aerial plant part of randomly selected plants from each treatment for 24 hours after the application of treatments and total plant fresh weight per each treatment. There were significant differences (p<0.05)><0.05) on the yield of rape. It was concluded that T. vogelii, S. incanum and A. sativum aqueous crude extracts have some pesticidal effects on aphid in rape production.

Application of certain bacterial and fungal species for controlling Meloidogy...Premier Publishers
Bacillus subtilis (Bs) and B. megaterium (Bm), Pseudomonas fluorescens (Pf) as well as Trichoderma harziamum (Th), Trichoderma vierns (Tvr)and T. viride (Tv) were applied alone as well as in combination in pot experiment for controlling M. incognita in Cowpea. Results showed that Bs, Bp and Pf had nematicidal effects against M. incognita parameters viz., J2 in soil and roots, galls and egg-masses compared to untreated check. Bm alone had highly nematicidal activity against J2 in soil, while Bs was the best against J2 in roots, galls and egg-masses. Bs + Bm in combination was effective against M. incognita parameters. The tested species of Trichoderma also reduced the parameters of M. incognita.Th alone was highly effective in reducing the tested nematode parameters, followed by Tvr and Tv treatments. Tv + Tvr highly reduced the numbers of J2 in soil and roots, while Th+Tv+Tvr significantly reduced the galls and egg-masses numbers. All treatments also increased the growth parameters of cowpea viz., shoot length, fresh and dry shoot weights, and leaf numbers.

The tested species of Trichoderma also reduced the parameters of M. incognita.Th alone was highly effective in reducing the tested nematode parameters, followed by Tvr and Tv treatments. Tv + Tvr highly reduced the numbers of J2 in soil and roots, while Th+Tv+Tvr significantly reduced the galls and egg-masses numbers. All treatments also increased the growth parameters of cowpea viz., shoot length, fresh and dry shoot weights, and leaf numbers.

The study was conducted to test the activity of Pseudomonas fluorescens, Rhodotorula sp and fermented neem extract to protect potato plants against potato virusY disease development under field conditions. Infected potato tubers were soaked in P. fluorescens, Rhodotorula sp suspensions and in fermented neem extracts separately and sown in the field in completely randomized block design. The development of virus symptoms and the accumulation of virus in the plant based on Enzyme Linked Immunosorbent Assay (ELISA) were followed. The results obtained showed that the treatment of potato tubers with the three agents have significantly accelerated plant emergence, 5-6 days early than non treated ones, and improved plant growth, the plant dry weights ranged from 120-177 g/plant compared to 42 g/plant in non treated plants. The enhancement of plant growth was found associated with reduction in disease severity based on symptoms development and restriction of virus concentration as proved by ELISA absorbance of 405 nm, 0.14-0.23 compared with 2.50 in non treated plants. The results indicated that the use of bioagent to induce systemic resistance provide an efficient tool, as insecticide alternative to manage potato virus Y in potato. Red rot is a devastating disease in sugarcane caused by fungus, Colletotrichum falcatum. In this study, eighteen different botanicals were screened for identifying effective antifungal compound against C. falcatum. Among the plants screened, 15 per cent aqueous leaf extract of Psoralea corylifolia alone inhibited 100 per cent growth of both mycelium as well as spore germination under in vitro conditions. The extract did not exhibit any inhibitory effect to the beneficial microbes viz., Pseudomonas fluorescens, Bacillus megaterium and Gluconacetobacter diazotrophicus which are normally used in sugarcane. The effective plant extracts exhibiting 100 per cent antifungal activity was subjected to TLC, HPLC and GC-MS analysis to identify the bioactive antifungal compound. It revealed the presence of 7H-furo [3,2-G] (1) benzopyran-7-one as main bioactive compound which is thought to be the intermediate of antifungal compound, 8 – methoxypsoralen formed during biosynthesis.

A pot experiment was conducted to evaluate the nematicidal efficacy of a biocontrol agent, Pseudomonas flourescens for the management of root-knot nematode, Meloidogyneincognita on chickpea (Cicer arietinum L.) cv.‘Avarodhi’ under glasshouse conditions. All the treatments were found to significantly improve the growth and physiological parameters of chickpea and reduction in pathological parameters as compare to untreated inoculated control. The highest improvement was observed in those plants treated with P. flourescens alone. Concomitant and sequential inoculation of P. flourescens with M.incognita also showed significant improvement in growth parameters of chickpea. Least enhancement in growth parameters was observed in those plants inoculated with nematode alone. It may be due to the nematcidal behaviour of P. flourescens against root-knot nematode, M. incognita. Hence, it may be concluded that P. flourescens as biocontrol agent is better substitute against chemical nematicides for the sustainable management of M. incognita and reduce environmental hazards.
ABSTRACT- Phylloplane fungi presented on the leaf surface were screened and selected for the assessment of their
potential against Alternaria alternata causing leaf spot disease of Rauwolfia serpentina (Sarapgandha). Among the
antagonists Trichoderma harzianum ISO-2 showed minimum conidial germination. Effect of foliar spray of phylloplane
fungi on the lesion development was also studied. In mist chamber studies seven treatments comprising application of
antagonists in the presence of pathogen proved effective in causing per cent disease reduction by 30-50 % in comparison
with the control. In the field trials Penicillium sublateritium showed the maximum fresh and dry root weight. P. sublateritium
also showed significant percentage disease reduction in comparison to control.

The objective of this study was to examine the antiviral activity of native lactoferrin against Potato virus x, the most important virus that severely affects potato crop and productivity in Egypt, using tissue culture technique and spraying the plants in greenhouse by the aqueous solution of lactoferrin.
— Post-harvest diseases are a major problem for banana yield. Despite treatments with chemical fungicides, a persistence of diseases is noticed. This study aims at proposing a biological control method against banana post-harvest diseases by using banana rachis leachate. The effect of leachate has been tested in vitro on mycelial growth, conidial germination and in vivo on pathogenic fungi virulence. All leachate concentrations (5, 15 and 20%) tested showed antifungal activity on the tested fungi. However, the 20% concentration was more effective with complete inhibition of mycelial growth and conidial germination of all fungi. No symptoms of crown rot and anthracnose were observed after treatment of bananas with leachate. However, with azoxystrobin, the prevalence of crown rot and anthracnose was 60% and 30%, respectively. Banana rachis leachate recorded highly significant reduction of banana finger rot prevalence compared to azoxystrobin. Banana rachis leachate have strong antifungal properties that may be useful to control banana post-harvest disease as a safe alternative option to chemical fungicides Keywords— banana; post-harvest diseases; banana rachis leachate, antifungal activity.
Toxic Effects of some Pungent Smelling Seed Extracts and Powders on Sitophilu...Premier Publishers
Sitophilus zeamais is a notorious field-to-store pest of maize in the Tropic. The use of chemical insecticides, regardless of its adverse effects has been the major means of managing this pest. In this study, bio-insecticides of plant origin was used to control maize weevil in the store. The dry fruits of Capsicum frutescens, Piper guineense and Aframomum melegueta were extracted with ethanol, using Soxhlet apparatus. The extracts and powders were evaluated on S. zeamais Motschulsky for mortality, oviposition and adult emergence. The long term storage of the treated maize was also investigated. The results obtained shows that all extracts and powders from C. frutescens and P. guineense caused 100 % mortality by day 12 of exposure with all extract concentrations and doses of powder applied. Powders of A. melegueta was able to achieve 100 % mortality only by day 12 of exposure. The extracts also suppressed oviposition and subsequent adult emergence. Extracts and powders of A. melegueta and P. guineense completely prevented infestation and hence damage of the treated maize grains for a period of three months. The result obtained suggests that extracts and powders of the test plants most especially that of P. guineense and C. frutescens can be utilized in protecting stored maize grains from infestation by S. zeamais.

Abstract: A total of nine cucumbers, each of Garden egg and pawpaw samples were collected from Wurukum, High level and Wadata markets and cultured on appriopate agar, for colony count and isolation of bacteria according to their cultural and biochemical characteristics. The results revealed that garden egg from High Level Market had the highest bacterial count (1.9x105cfu/g) and the least was pawpaw from High Level Market (1.1 x 105 cfu/g). However, it was not statistically significant. The bacteria isolated were; Propianol bacteria (23.3%), Escherichia coli (16.6%), Staphylococcus aureus (36.7%), Bacillus spp (10.0%) and Corynebacteria (13.3%). The fungal isolates were Aspergillus flavus (10%), Aspergillus fumigatus (20%), Aspergillus nidulus (10%), Aspergillus terreus (20%) and mucor (40%). The result of this study shows fruits sold in Wurukum, High Level Market and Wadata Market are contaminated and may cause harm to consumers, so measures such as proper handling should be taken to control the contamination of these fruits.

Abstract:
Efficacy of bioagent (Paecilomyces lilacinus) and the bioproduct (Radiant) in various combinations was
assessed on the reproduction of Meloidogyne incognita on eggplant. The influence of P. lilacinus and
Radiant was determined on egg hatching and second stage juvenile (J2) mortality under in vitro conditions.
The concentrations of 1% and 100% of Radiant and P. lilacinus respectively both alone and in combined
application caused significant mortality and reduction in egg hatching at all time intervals. The interaction
of P. lilacinus and Radiant was determined individually, concomitantly, and sequentially on reproduction
of M. incognita on eggplant under greenhouse. The reproduction of M. incognita was significantly reduced
in the concomitant treatment consisting of both P. lilacinus and Radiant followed by sequential and
individual treatment of Radiant and the plant growth parameters incresed significantly. Our findings
suggest that P. lilacinus and Radiant have the ability to regulate nematode population and may serve as
nematicides.

1. Introduction
Eggplant (Solanum melongena L.) is an important cheap summer vegetable with medicinal properties as
well as containing water, protein and vitamin A (Tindall 1978). In Pakistan, it is cultivated on an area of
8673 ha having annual production of 88148 tonnes (FAO 2009). The average harvested yield is very low
due to various pests such as insects, fungi, bacteria, viruses and nematodes. In Pakistan, root-knot
nematode, M. incognita caused severe damage to eggplant and other vegetables and their host range include
more than 3000 plant species (Anwar et al. 2009). High temperature is suitable for endurance and
reproduction of nematodes. Among the root knot nematodes, M. incognita and M. javanica are of wide
spread and economically most important (Anwar & McKenry 2010).
Nematode control is far more complex than any other kind of pathogens because nematodes mainly attack
under ground parts of plants (Sikora & Fernandez 2005). A range of management strategies studied,
including crop rotation, soil amendments and nematicides that could be collectively used to enhance the
activity of naturally occurring biological control methods (Sikora 1992). Even though chemical nematicides
have a great promise for the control of nematodes but restricted due to health hazards involved. Different
practices are used in the integrated pest management (IPM) but the biological control would be the most
enviable. Culture filtrates from various fungi such as species of Paecilomyces, Verticillium, Fusarium,
Aspergillus, Trichoderma, Myrothecium and Penicillium were tested against juvenile mortality and on egg
inhibition of nematodes. These filtrates proved to be toxic due to the release of antibiotics or enzymes
(Dorcas et al. 2010). The potential of B. thuringiensis var. Berliner, Saccharopolyspora spinosa, Fusarium
spp. and Trichoderma spp. against G. rostochiensis on potato was evaluated. The plant growth and yield
was improved by all the treatments. Significant inhibitory effect was caused by bioagents on the
reproduction of the nematode (Trifonova 2010).

Keeping in view the importance of biological control of root knot nematodes, it was planned to manage M.
incognita by the interaction of P. lilacinus and Radiant. The objective of the present study was to assess the
effectiveness of P. lilacinus and the Radiant (active ingredient; Spinetoram; 11.7%) for the management of
M. incognita on eggplant. Radiant was not yet used for the management of root-knot nematode and it was
also integrated for the first time with P. lilacinus for the control of M. incognita.
2. Materials and Methods
2.1 Nematode inoculum
Seeds of tomato cv. Money maker were planted in soil sterilized with formalin. Transplanting of seedlings
was done after three weeks in earthen pots. Seedlings were inoculated with 5000 J2s per pot by making
holes around the plant rhizosphere.
2.2 Fungal inoculum
Pure culture of Paecilomyces lilacinus was obtained from the culture collection of Arid Agriculture
University, Rawalpindi. One week old cultures of bioagent maintained on PDA slants at 28 ± 1 were used
for the present study. Culture filtrates of P. lilacinus were obtained by growing one week old culture of P.
lilacinus on Potato Dextrose Broth (in 250 ml flasks containing 100 ml broth media). Flasks were
autoclaved at 121 OC and 15 psi for 15 minutes. Each flask was inoculated with three scoops of 5mm
diameter from pure culture containing petri dishes and incubated at 25OC for 15 days. After incubation the
cultures were filtered through Whatman filter paper No.1 to remove the mycelial mats. These filtrates thus
obtained were considered as standard (100% concentration).
2.3 Influence of P. lilacinus and Radiant on juvenile mortality and egg hatching of M. incognita
Radiant (Spinetoram; 11.7%) was obtained from the local market to check its efficacy against M. incognita.
Different concentrations viz. 1%, 0.75%, 0.50% and 0.25% were prepared by adding requisite amount of
sterilized distilled water. Different concentrations of culture filtrates of P. lilacinus viz. 75%, 50% and 25%
were prepared from standard (100% concentration) by adding requisite amount of sterilized distilled water.
The population of M. incognita maintained on the roots of tomato cv. Money maker in the glass house of
Department of Plant Pathology, University of Agriculture, Faisalabad was used. Eggs were isolated from
egg masses and nematodes were extracted (Hussey & Barker 1973). The effect of different concentrations
of Radiant and culture filtrates of P. lilacinus were investigated on juvenile mortality and egg hatching of
M. incognita. Three ml of each concentration of Radiant and P. lilacinus were pipetted into Petri dishes.
Fifty freshly hatched Juveniles of M. incognita in 0.5ml distilled water were added to each Petri dish. Fifty
eggs of M. incognita were picked and placed into each dish and kept at 280C. Eggs and juveniles placed in
distilled water served as control. Each treatment was replicated five times. Data on juvenile mortality and
egg hatching was recorded after 24, 48, 72 hours.
2.4 Interaction of P. lilacinus and Radiant on reproduction of M. incognita on egg plant individually,
concomitantly and sequentially in green house
Three weeks old eggplant seedlings were transplanted into 20 cm diameter earthen pots containing formalin
sterilized soil. Concentrations 1% and 100% of Radiant and P. lilacinus respectively were used because
these concentrations caused maximum juvenile mortality and reduction in egg hatching during in vitro
investigation. There were six treatments and five replications for each treatment.
2.4.1 Treatments (T)
T1 =P. lilacinus, T2 = Radiant, T3 = P. lilacinus + Radiant, T4 = P. lilacinus then Radiant, T5 =
Radiant then P. lilacinus, T6 =Check (Nematode).
Each pot received 20 ml of 100% concentration of P. lilacinus containing 1x 107 spores/ml and 20 ml of
1% concentration of Radiant except check plants according to the treatment scheme individually,
concomitantly and sequentially. A period of one week was followed in the sequential treatments. Freshly
hatched 1000 J2s of M. incognita per pot were used as inoculum by making 3-4 holes (3 cm deep) around

each plant with the help of pointed wood. After inoculation pots were kept in greenhouse (25-300C) in
completely randomized design. Plants were allowed to grow for 60 days. The data was recorded on number
of galls, number of egg masses per root system by staining the roots with Phloxine B solution for 5 minutes
(Holbrook et al. 1983), eggs per egg mass, number of females by staining the roots with acid fuchsin-
lactophenol, number of J2/100 cm3of soil and number of J2/g of root were determined by processing the soil
with the help of Cobb’s sieving and decanting method along with Baermann’s funnel technique (Southey,
1986). The reproduction factor (Rf) was calculated by dividing the final population (pf) by initial
population (pi), root weight (g), root length (cm), shoot weight (g), shoot length (cm) and number of leaves.
Experiment was repeated to confirm the results and thus the data was subjected to analysis of variance and
the significant difference in treatment means were separated by using Duncan Multiple Range Test at ( P=
0.05) and Bartlett's test at ( P= 0.05).
3. Results
3.1 Influence of P. lilacinus and Radiant on Juvenile mortality and egg hatching of M. incognita
Mortality of juveniles of M. incognita was affected by concentrations and exposure time in all treatments
(Table 1). The J2s treated with Radiant and combine application of Radiant and P. lilacinus had significant
more mortality as compared to P. lilacinus alone at all time intervals. The combined application of Radiant
and P. lilacinus showed the maximum mortality at all concentrations after all time intervals. Egg hatching
of M. incognita was inversely proportional to the concentrations of Radiant and culture filtrates of P.
lilacinus. Radiant at 1% concentration had significant (P = 0.05) more effect on mortality (50.0) after 72 h
from its 0.5% (48.2) and 0.25% (40.2) concentrations. Paecilomyces lilacinus at 100% concentration
caused significant greater mortality (40.2) as compared to all other concentrations at all time intervals. At
75% concentrations P. lilacinus caused significantly high mortality (31.6) from its 50% concentration
(24.4) after 72 hours of exposure. The combined application of Radiant and P. lilacinus (1 + 100%) showed
significant results of J2 mortality (50.0) as compared to the alone application of Radiant and P. lilacinus at
all time intervals. At 0.75+75% concentration results (50.0) were not significantly different from 1+100%
concentration (50.0) after 72 h of exposure but different from its 0.5+50% (48.4) and 0.25+25% (40.2)
concentrations.
Egg hatching of M. incognita was significantly reduced with the duration of exposure (Table 2). Egg
hatching was significantly decreased in Radiant and in combined application of Radiant and P. lilacinus
than treated with P. lilacinus alone at all time intervals. The combined application of Radiant and P.
lilacinus exhibited the maximum reduction in egg hatching of M. incognita at all concentrations after all
time intervals. At 1% concentration, Radiant had significant reduction (14.6) in hatching over all other
concentrations at all time intervals. Hatching of M. incognita was significantly (P = 0.05) reduced in 0.75%
(23.8), 0.5 % (31.0) and 0.25% (39.6) concentrations of Radiant after 72 h of exposure as compared to
control (49.00). Paecilomyces lilacinus at 100% concentration caused significant reduction (17.80) in
hatching over all other concentrations at all time intervals. At 75% concentration P. lilacinus caused
significant reduction (26.2) in hatching from its 50% (31.8) and 25% (41.4) concentrations at all time
intervals. Radiant in combination with P. lilacinus showed more significant reduction in egg hatching than
in alone application at all concentrations after all time intervals. The combined application at of Radiant
and P. lilacinus at 1+100% concentration caused significantly more reduction (10.4) in egg hatching from
all other concentrations at all time intervals. Egg hatching was also significantly reduced at 0.75+75%
(20.8), 0.5+50% (29.4) and 0.25+25% (38.2) concentrations as compared to control (49.0) after 72 h of
exposure.
3.2 Interaction of P. lilacinus and Radiant on reproduction of M. incognita on egg plant individually,
concomitantly and sequentially in green house
The interaction of P. lilacinus and Radiant on plant growth parameters and on reproduction of M. incognita
on eggplant was studied individually, concomitantly and sequentially in greenhouse. The plant growth
parameters varied significantly, there was significant (P= 0.05) increase in root length, shoot weight, shoot

length, number of leaves and decrease in root weight of eggplant was observed in concomitant treatment
consisting both bioagent and bioproduct (Table 3). An increase in growth parameters of eggplant treated
with P. lilacinus alone, followed by Radiant and sequential treatments of Radiant and P. lilacinus was
observed. The results revealed that root weight was maximum in the check plants treated with M. incognita
only due to the presence of galls on the roots. The reproduction of M. incognita was significantly reduced
in the concomitant treatment consisting of both P. lilacinus and Radiant followed by sequential and
individual treatment of Radiant (Table 4). Maximum reduction in number of galls (15.0), number of egg
masses (4.2) counted by staing the roots with phloxine, number of eggs per egg mass (75.4), number of
females (16.2), J2/100 cm3of soil (75.8), J2/g of root system (87.8) was observed in concomitant treatment.
There was significant reduction in the reproduction parameters of M. incognita in plants treated with
Radiant in individual treatment. Reproduction of M. incognita was also reduced in the sequential treatments
of Radiant and P. lilacinus followed by the individual treatment of P. lilacinus. Maximum reproduction of
M. incognita was observed on the roots of check plants. Reproduction factor was (0.6) in concomitant
treatment, (0.8, 0.8) in sequential treatments and (2.2, 0.8) respectively in individuals treatments of P.
lilacinus and Radiant as compared to check (13.4).
4. Discussion
Root- knot nematodes are the most prevalent and very important group of plant parasitic nematodes
occurring all over the world but found more the in areas with warm climate. Root knot nematodes reduce
the market value of infected plants by depriving their nutrients. Losses are profound at seedling stage and
results in devastation of the crop. Root knot nematodes are difficult to control because of their high
reproductive potential. Biological management of nematodes has become one of the most feasible
alternatives to the nematicides. Different practices are used in the integrated pest management but the
biological control would be the most enviable because most of the successful nematicides have been
restricted in agriculture due to health hazards involved (Veremis & Roberts 1996).
The present investigation revealed that Radiant and the culture filtrates P. lilacinus possessed the
nematicidal activity. Nematicidal effects of culture filtrates of various fungi on egg hatching and J2
mortality of root knot nematodes have been investigated by many workers (Cayrol et al. 1989; Hallmann &
Sikora 1996; Zaki & Maqbool 1991). In our study, the effectiveness of P. lilacinus and Radiant was
assessed on J2 mortality of M. incognita under in vitro conditions. The findings suggested that J2 mortality
of M. incognita was directly propotional to concentratins and duration of exposure. Our findings are in
conformity with the previous studies (Ahmad et al. 2010; Ayatollahy & Fatemy 2010; Goswami & Mittal
2002; Satyandra & Mathur 2010). Another in vitro experiment was conducted to assess the nematicidal
effects of P. lilacinus and Radiant on egg hatching of M. incognita. Egg parasitization of M. incognita
increased with the increase in concentration of Radiant and P. lilacinus and also with duration of exposure.
Maximum parasitization was assessed after 72 h. Antagonist fungi possessed larvicidal and ovipocidal
properties against root knot nematode. Most of the antagonistic fungi are egg parasitizing but a few culture
filtrates of these fungi produced the antibiotics, glioviren and gliotoxin, (Lumsden et al. 1992; Howell et al.
1993). The pathogenesity of P. lilacinus on the eggs of the Meloidogyne spp. was tested (Bonants et al.
1995). The enzyme in culture filtrates entirely bound with the nematode eggs. Immature eggs were more
susceptible to protease treatments, while the mature eggs have the resistance (Mukhtar et al. 2003; Siddique
& Haque 2000; Ashoub et al. 2009).
Our results confirmed that integration of P. lilacinus and Radiant reduced the population of M. incognita
and also improved the plant growth parameters. Filtrates of P. lilacinus strains related to their capability to
produce leucinostatins. It was also observed that root weight was directly propotional to the number of galls
which were maximum in the check plants. The growth parameters were improved by biological control
agent P. lilacinus whereas it also reduced M. javanica reproduction on simultaneous and sequential
inoculation of P. lilacinus with M. javanica (Ganaie & Khan 2010). The efficacy of Serratia 214
marcescens, three different T. harzianum isolates and carbofuran against M. incognita was 215 evaluated in
the glasshouse and the results showed that chemical nematicide could be replaced 216 to some extent by

microbial antagonists viz. Serratia marcescens and T. harzianum isolates as 217 they are environmentally
safe (Mahfouz et al. 2010). The integration gave better results than the alone application that is also proved
from our results which are in conformity with (Anjos et al. 2010; Park et al. 2004; Rao et al. 1998; Sharma
& Pandey 2009). It was assessed from the present investigation that plant growth parameters were
enhanced significantly and also the reproduction of M. incognita was decreased due to the application
Radiant and P. lilacinus. Our results revealed that the bioagent; P. lilacinus and the bioproduct; Radiant
have the potential to regulate the population of root-knot nematode, M. incognita by reducing egg hatching
and killing the J2s and if applied in a nematode infested field before planting.
