1. Kool Aid Dye Separation and Analysis Pre Lab Information

2. Purpose: 1. Separate a mixture of dyes using column chromatography. 2. Analyze the separated samples for purity using spectrometry.

4. Mobile Phase – the phase that moves.

5. Analyte – the substance being separated.

7. Separation is based on: 1. The size of the particles. a. Larger particles come through slower. b. Smaller particles come through faster. 2. The dye’s attraction to the column. a. More attracted = comes through slower. b. Less attracted = comes through faster.

9. We use distilled water as our reference. This tells the instrument what zero absorbance (or 100% transmittance looks like).

11. Not pure = one larger peak for the color you see and another smaller peak at the color you don’t see (also following complimentary color rules). Peak height varies directly with concentration. More concentrated = higher peak.
