INTRODUCTION:-
• The HIV TRI DOT kit consist of three different regions.
(I). HIV-1 REGION:-
This region consist of precauted HIV-1 virus antigen such as gp41 & gp120 on
immuno filtration membrane.
(II). HIV-2 REGION:-
This region consist of precauted HIV-2 virus antigen such as gp36 on immuno
filtration membrane.
(III). CONTROL REGION:-
This region consist of precauted anti-mouse antibodie on immuno filtration
membrane.
(IV). CONJUGATE SYSTEM:-
. In HIV tri dot test, conjugate system is used known as a protein ‘A’ conjugate.
. This system is consist of anti HIV antibody with conjugate.
PRINCIPLE:-
• AT HIV-1 REGION:-
• If the patient suffer from AIDS by HIV-1 virus, the patient will have
HIV antibodies(gp120 & gp41) in the serum sample.
• After application of sample, these HIV antibodies will bind with
precauted antigens in HIV-1 region.
• This binding lead to the formation of Ag-ab complex.
• The conjugate system consist of anti-Hiv antibody with conjugate
after application of conjugate system this will bind with perform Ag-
ab complex.
• This binding activate the conjugate system and produce colour.
• Production of colour dot indicate the positive result for the presence
of HIV antibody against Hiv-1 virus in the patient sample.
AT ‘C’ REGION:-
• The conjugate system consist of anti HIV antibody which act as a
mouse antibody.
• This mouse antibody act as antigen for anti mouse antibody which is
already precauted on ‘C‘ region.
• The binding between mouse antibody & anti mouse antibody
activate the conjugate system & produce a colour.
• Production of colour dot at ‘C‘ region indicate that the procedure is
correct.
PROCEDURE
• Take a HIV tri dot kit.
• Open a foil pouch.
• Lable the kit with patient name & patient id.
• Add three drops of buffer solution.
• Add one drop of patient sample.
• Add five drops of buffer solution.
• Add two drops protein ‘A‘ conjugate system.
• Add five drops of buffer solution.
• Wait for the result.
INTERPRETATION
• POSITIVE RESULT:-
• +ve result for Hiv-1 virus. +ve result for Hiv-2 virus

• NEGATIVE RESULT:-

    AT ‘C’ REGION:- •The conjugate system consist of anti HIV antibody which act as a mouse antibody. • This mouse antibody act as antigen for anti mouse antibody which is already precauted on ‘C‘ region. • The binding between mouse antibody & anti mouse antibody activate the conjugate system & produce a colour. • Production of colour dot at ‘C‘ region indicate that the procedure is correct.

    PROCEDURE • Take aHIV tri dot kit. • Open a foil pouch. • Lable the kit with patient name & patient id. • Add three drops of buffer solution. • Add one drop of patient sample. • Add five drops of buffer solution. • Add two drops protein ‘A‘ conjugate system. • Add five drops of buffer solution. • Wait for the result.
