9. Introduction to Drug Delivery in Cancer Therapy
• A new era of Targeted drug delivery and precision medicine using nanotechnology in Cancer therapy
• DNA structures are modified with functional groups to act as a DNA carriers, which can directly deliver
the drug into specified target cells.
• This application came into existence with the appearance of scaffolded DNA origami method.
• The conventional method has many drawbacks like low solubility, low stability, and cytotoxicity. Thus,
leads to inadequate efficiency of cancer therapy.
• This technique uses DNA nanostructures for delivering the drug directly into the cancer cells and
reduces severe side effects.
10. Nanoparticle Drug Delivery in Cancer Therapy
DOXORUBUCIN [DXO]
• Doxorubicin is a type of chemotherapy drug called an anthracycline.
• It slows or stops the growth of cancer cells by blocking an enzyme called topo isomerase.
• DOX can cause deaths of healthy cells too.
• Commonly used doxorubicin-containing regimens are AC,TAC, ABVD, BEACOPP, CHOP and FAC.

12. Main points to be discussed -
● How does Doxorubicin intercalate with DNA origami
● The procedure for building a drug carrier using DNA origami.
DNA Origami : Doxorubicin drug carrier.
13. Functional part of the structure
● Amino acid - not interacting with
the phosphates but is sitting in the
minor groove.
● b, c, and d rings constitute an
intercalative region.
● A ring interact through hydrogen
bonding with the double-helix.
● DOX is intercalated in the GC
sequences

into the triangle and tube structures through the hybridization of staple strands. Watson−Crick base pairs in the double helices
serve as docking sites for doxorubicin intercalation. (b) drug loading efficiencies: Efficiency of doxorubicin incorporation into

circumvention of drug resistance. Journal of the American Chemical Society, 134(32), pp.13396-13403.
15. The advantages
❖ Stable.
❖ Create complex shape.
❖ Very precise.
❖ Racognies by the bod.
Limitations
❖ Expensive.
❖ Sometimes can be killed by antibody.
