2. INTRODUCTION
BREAST CANCER
A disease in which cells in the breast grow out of control. It's most commonly
diagnosed in women.
It can start in different parts of the breast, such as the lobules, ducts, nipple, stroma,
blood vessels and lymph vessel.
Most Common Symptoms:
- Change in size in one or both breasts
- Lump or swelling in armpits
- Dimpling of the skin
- Rash on or around nipple
3. INTRODUCTION
NAA25
NAA25 is a protein coding gene that appears to be very prevalent in breast cancer.
NAA25-deficient cells:
-> increases cell apoptosis
-> delays G2/M phase
-> decreases cell clone formation
4. OBJECTIVE
To see the relationship between NAA25
gene and breast cancer by comparing the
gene expression between normal tissue
and breast cancer tissue
-> Specifically, to see effects that the gene
has on breast cancer cells when it’s
altered.

WESTERN BLOT
A laboratory technique that is used
when wanting to detect a specific
protein in a blood or tissue sample.
To separate the sample’s proteins, it
involves the use of gel
electrophoresis
COLONY FORMATION ASSAY
An in vitro cell survival analysis
based on the ability of a single cell
to grow into a colony.
It involves steps such as treatment,
plating and staining.
6. METHODS
APOPTOSIS ASSAY
Detects and quantifies how many
cells have undergone apoptosis
(programmed cell death).
RT-qPCR
Monitors the amplification of a specific
DNA molecule during PCR (polymerase
chain reaction).
7. RESULTS
Usage of Western-blot and RT-qPCR to analyze
NAA25 expression in breast cancer tissues
and para-carcinoma tissues
● NAA25 was highly expressed in breast
cancer tissues.
● Thicker Line = More Expression
8. RESULTS
A
NAA25 gene expression in breast cancer
cell line T47D was greater than in
MCF10A.
B
After the knockdown of NAA25 (in T47D
cell line), the mRNA and protein
expression of the gene was diminished .

lncHSAT164, was highly expressed in breast
cancer, and it is shown that when
overexpressed, it could promote colony
formation and when down-expressed, it could
promote cell apoptosis and regulate cell cycle.

DISCUSSION
10. CONCLUSION
1
First, molecular biology gives the medical field different methods and techniques to
learn and see how different diseases are created, as well as how they progress.
Without these tools, we, as doctors, wouldn’t be able to come up with effective
vaccines or treatments to stop these illnesses.
2
In addition, investigation in the medical field is heavily based on
molecular biology, since it’s important to know how cells
function, which cells are in charge of specific reactions, and what
happens when a cell has an error in their genetic code. This helps
us understand how diseases work.
