detecting DNA transfer events as they occur, enabling detection of transient transfer events.
The recording system captures spacers from HGT events stably into genomic arrays that can be
used to identify mobile elements beyond current methodologies. The ability to detect, in real-
time, transient transfer events and those occurring at low frequencies enables an in-depth
characterization of mobile DNA in complex microbiomes.
As DNA transfer is confined by recipient range we are only capable of recording DNA that can
enter the E. coli BL21 recording strain. When using other E. coli strains as donor we showed that
transfers can be resolved down to the individual plasmids from donors that can carry up to 6
putatively mobile plasmids. We find that the different plasmids varied in transfer efficiencies
likely reflecting differences in intrinsic plasmid transfer efficiency, donor transfer efficiency,
recipient receiving efficiency, or a combination of these factors. Even though the observed
mobile elements were all classified as plasmids, we still expect that phages are an important
contributor to HGT. However, as illustrated by the recording of phage M13, our system is not
optimal for detecting invading phages as CRISPR spacer adaptation requires active growth of the
recording cell19, which is often impaired upon phage infection. In addition, infection with
phages can lead to cell lysis and subsequent loss of recording cells from the population.
Furthermore, it is not clear what the expected concentration of E. coli targeting phages is in
human fecal samples. Previous studies indicate that plaque forming units (pfu) from human feces
on E. coli is in the range of 101107 per gram, with a median concentration of ~103 per
gram42,43. Considering that the concentration of E. coli in human feces is in the range of
107109per gram44, interactions between phages and the recording strain might be substantially
less common than interactions between commensal E. coli and the recording strain. Washing of
the fecal sample (i.e., to remove antibiotics and other factors that can inhibit the recording strain)
likely also contributed to the loss of phage particles. Lastly, as E. coli is not naturally competent,
we are not able to detect naked environmental DNA. Consequently, our recording strain is best
suited for detecting plasmid transfers.
When our approach is applied to clinical fecal specimens, we were able to identify active DNA
transfer in 22% of the samples (6 out of 27). It is difficult to evaluate whether the frequency of
observed HGT in the fecal samples (22%) is limited by the methodology or whethe.

he wished they would state them. The sum with which he should think of filling up the blank
would not be such as to materially affect our finances.
2. [Mr. Macon:] The sufferings of the people of Savannah were doubtless very great; no one
could help feeling for them. But he wished gentlemen to put their finger upon that part of the
Constitution which gave that House power to afford them relief. Many other towns had suffered
very considerably by fire. He believed he knew one that had suffered more than Savannah in
proportion to its size: he alluded to Lexington in Virginia, as every house in the place was burnt.
If the United States were to become underwriters to the whole Union, where must the line be
drawn when their assistance might be claimed? Was it when three-fourths or four-fifths of a
town was destroyed, or what other proportion? Insurance offices were the proper securities
against fire. If the Government were to come forward in one instance, it must come forward in
all, since every sufferer's claim stood upon the same footing. The sum which had been given to
the sufferers at St. Domingo was to be charged to the French Republic, and that given to Count
de Grasse's daughters was in consideration of their father's services. But New York had as great
right to come forward and expect relief as Savannah. He felt for the sufferers in all these cases,
but he felt as tenderly for the Constitution; he had examined it, and it did not authorize any such
grant. He should, therefore, be very unwilling to act contrary to it.
3. Mr. Rutherford said, he felt a great deal of force on what gentlemen had said. There were two
circumstances which were perfectly conclusive in his mind. He saw it our duty to grant relief
from humanity and from policy. Savannah was a city of a minor, helpless State; it was a very
young State, yet it was a part of the Union, and as such, was as much entitled to protect.

Post-Lab Assessment Ruffed lemurs found in the eastern rainforests of Madagascar typically live arboreally, in the crowns of large trees, with most of their time spent 15 to 25 meters (50-80 feet) above the forest floor. Their diet comprises mainly of fruit, flowers, and young leaves accessible year round. Prized for their meat and fur, ruffed lemurs have been hunted nearly to extinction. Many residents of Madagascar find a large portion of the protein in their diet from animals hunted in the rainforest. Called bushmeat, this form of protein can be a valuable dietary resource to people without access to other food. However, many wild animals carry diseases that can be transmitted through their blood to humans who are processing the meat for consumption. For example, the Ebola epidemic of 2014 likely began with the transmission of the virus from a fruit bat to humans in Guinea. In addition to hunting, ruffed lemurs also suffer from habitat loss due to deforestation, climate changes, and urban development. Because of these threats, the ruffed lemur became critically endangered in 2008 meaning the species faces a very high risk of extinction. Fortunately ruffed lemurs reproduce easily in captivity so they make an excellent species for reintroduction into the wild. Recently conservation biologists noticed that a conspiracy, or group, of lemurs in a Madagascar wildlife preserve were behaving oddly. Several lemurs were seen on the ground acting lethargic. The biologists sedated three lemurs to run some tests and see if they could understand this strange new behavior. All three of the lemurs had elevated body temperatures and appeared to be dehydrated and underweight as if they had not been eating regularly despite the availability of food in the tree tops of the preserve.
