TALEN kits are commercially available and use either ligation-independent or Golden Gate cloning methods to construct TALENs. The Golden Gate method uses type IIS restriction endonucleases to create unique DNA overhangs, allowing DNA fragments to be assembled in a single reaction without ligation. Vectors such as pZHY500 and pZHY501 are compatible with the Golden Gate method for expressing TALENs in yeast. TALENs are delivered into cells using electroporation, which applies an electric field to create pores in the cell membrane.
