This document provides a standard operating procedure for testing the susceptibility of adult mosquitoes to insecticides using the WHO bottle bioassay method. The procedure involves coating glass bottles with a known concentration of an insecticide dissolved in acetone or a solvent/surfactant mixture. Adult female mosquitoes are exposed to the insecticide inside the bottles for 1 hour, after which mortality is recorded at 24 or 72 hours post-exposure. The discriminating concentrations to be used for different insecticides and mosquito species are provided. Proper preparation, handling, and storage of stock solutions is described. Health and safety precautions for working with insecticides are also outlined.
