This document provides instructions for a squash method to prepare plant or animal tissue samples. The method involves fixing tissue samples in a Carnoy or fixative solution of 3 parts ethanol to 1 part acetic acid, washing the samples, staining them with acetocarmine, destaining them briefly in acid alcohol, and squashing them under a cover slip to view under a microscope. The squash method allows for thin sections of tissue to be viewed microscopically while maintaining the integrity of cellular structures.
