This document describes a methodology for isolating cimetidine, a histamine H2 receptor antagonist used to treat stomach ulcers, from human plasma using HPLC for quantitative analysis. The objectives are to understand cimetidine's mode of action and effects of overdose, and to develop an HPLC method for analyzing spiked plasma samples. The methodology uses HPLC with a C18 column, UV-Vis detector at 230nm, and mobile phase flow rate of 1.0 mL/min to separate and quantify cimetidine. Sample preparation involves preparing standard solutions and extracting cimetidine from plasma using solid phase, liquid-liquid, or protein precipitation extraction for HPLC analysis.
