2. Small RNA expression showed the highest fold changes in response to nitrofen.
3. Bioinformatic analysis identified three important biological functions of microRNAs in lung development impacted by nitrofen: developmental induction, ventricular cardiac muscle tissue morphogenesis, and regulation of apoptotic signaling.

RET is an abbreviation for "rearranged during transfection", as the DNA sequence of this gene was originally found to be rearranged within a 3T3 fibroblast cell line following its transfection with DNA taken from human lymphoma cells. The human gene RET is localized to chromosome 10 (10q11.2) and contains 21 exons.
The natural alternative splicing of the RET gene results in the production of 3 different isoforms of the protein RET. RET51, RET43 and RET9 contain 51, 43 and 9 amino acids in their C-terminal tail respectively. The biological roles of isoforms RET51 and RET9 are the most well studied in-vivo as these are the most common isoforms in which RET occurs.
Common to each isoform is a domain structure. Each protein is divided into three domains: an N-terminal extracellular domain with four cadherin-like repeats and a cysteine-rich region, a hydrophobic transmembrane domain and a cytoplasmic tyrosine kinase domain, which is split by an insertion of 27 amino acids. Within the cytoplasmic tyrosine kinase domain, there are 16 tyrosines (Tyrs) in RET9 and 18 in RET51. Tyr1090 and Tyr1096 are present only in the RET51 isoform.
The extracellular domain of RET contains nine N-glycosylation sites. The fully glycosylated RET protein is reported to have a molecular weight of 170 kDa although it is not clear to which isoform this molecular weight relates.

Lipid-transfer proteins (LTPs) were initially discovered as cytosolic factors that facilitate lipid transport between membrane bilayers in vitro. Since then, many LTPs have been isolated from bacteria, plants, yeast, and mammals, and extensively studied in cell-free systems and intact cells. A major advance in the LTP field was associated with the discovery of intracellular membrane contact sites (MCSs), small cytosolic gaps between the endoplasmic reticulum (ER) and other cellular membranes, which accelerate lipid transfer by LTPs. As LTPs modulate the distribution of lipids within cellular membranes, and many lipid species function as second messengers in key signaling pathways that control cell survival, proliferation, and migration, LTPs have been implicated in cancer-associated signal transduction cascades. Increasing evidence suggests that LTPs play an important role in cancer progression and metastasis. This review by Sima Lev describes how different LTPs as well as MCSs can contribute to cell transformation and malignant phenotype, and discusses how “aberrant” MCSs are associated with tumorigenesis in human.
Dual role of NRF2 in Cancer
NRF2 plays a dual role in cancer - it can function as both a tumor suppressor and promote cancer progression. Under normal conditions, NRF2 activates antioxidant genes to protect against oxidative stress and carcinogens. However, in cancer cells NRF2 is often hyperactivated and allows cells to evade chemotherapy by increasing antioxidant defenses and drug efflux pumps. Brusatol is a potential inhibitor of the NRF2 pathway that could sensitize cancer cells to chemotherapy by reducing NRF2 levels and antioxidant gene expression. More research is still needed to better understand the context-specific roles of NRF2 and develop NRF2 inhibitors for safe clinical use in cancer treatment.

However, our knowledge of microarray data analysis methods helps to classify the genes associated with differential and undifferential endocrine lineage, ductal cell and exocrine regions determine neurogenesis and neuron specific signaling pathways. Using Meta analysis of statistical rank correlation algorithm to rank the genes based on gene signatures. The reveal predicts 154 38 genes that were consistently and significantly up regulated 247 62 were down regulated in Breast cancer. Using functional annotation of gene clusters reveals only 47 genes is presumably associated with neuronal development and cell differentiation. A number of studies have been carried out for the treatment of breast cancer which includes the targeted therapies and increased survival rates in women. Essential PI3K/mTOR signaling pathway activation is observed in most breast cancers. The cell growth and tumor development in this case involves phosphoinositide 3 kinase (PI3K)/ Akt/mammalian target of rapamycin (mTOR) pathway. It has been observed, through preclinical and clinical trials, that there are a number of other inhibitors of PI3K/Akt/mTOR pathway, which either alone or in combination with other agents can be used for treatment of cancer. Pre-clinical studies have confirmed that P13K, Akt and mTOR inhibitors achieve anticancer effects by targeting different levels of the PI3K/Akt/mTOR pathway. This chapter evaluates the role of mTOR along with some of its inhibitors and the PI3K/Akt/mTOR pathway in the pathogenesis and prevention of breast cancer.
