HEAD SPACE SAMPLING
 Volatile Organic compounds (VOCs) are organic
chemicals that easily become gaseous at room
temperature.
 Some are known as Cancer- causing chemicals in
human or global warming.
 So, it is essentially required to analyze VOCs in various
applications.
What is a Headspace Auto sampler?
 Headspace Autosampler is one of sample preparation systems for Gas
chromatography that extracts volatile organic compounds out of liquid
or solid samples to inject them into Gas chromatography or GC/MS
automatically.
TYPES OF HEAD SPACE SAMPLING:
 It is of two types:
1. Static Headspace sampling
2. Dynamic Headspace sampling
STATIC HEADSPACE SAMPLING
 In static headspace sampling no trap is needed.
DYNAMIC HEADSPACE SAMPLING
 In dynamic headspace sampling trap is needed for
concentration.
• Dynamic Headspace (DHS) is an extraction technique used to
concentrate analytes from a solid or liquid sample. The sample is
heated and agitated in a sealed vial and the headspace above
the sample is purged onto a solid sorbent tube. The solid
sorbent tube is then thermally desorbed (to evaporate the
analyte) into the gas chromatograph. DHS is a non-equilibrium
technique, as the analytes in the headspace are constantly swept
onto the tube during extraction. The lack of a stationary phase
in this technique results in non-discriminatory collection of
analytes from the sample. Low levels of detection can be
achieved as exhaustive extraction conditions can be developed.
APPLICATION OF HEADSPACE AUTOSAMPLER
 PHARMACEUTICAL APPLICATION:
 Analysis of organic volatile impurities in pharmaceutical
products.
 Blood Alcohol Analysis.
 Water insoluble samples including Dimethylacetamide
and 1,3-Dimethyl-2-Imidazolidinone.
ENVIRONMENTAL APPLICATION:
 Analysis of volatile organic compound in drinking
water.
 Analysis of volatile organic compounds in various
solid waste matrices.
 Formaldehyde analysis in drinking water.
 Determination of MTBE (Methyl tert-butyl ether), TBA
( tert-butyl alcohol) and BTEX ( Benzene, toluene,
ethylbenzene, xylene) in soil.
CHEMICAL APPLICATION:
 Analysis of purity of petroleum product.
 Analysis of oxygenated volatile organic
compounds in Refinery Effluents.
FOOD APPLICATION:
 Analysis of flavors components.
 It is a standard test method for determining
residual solvent in packaging materials.
