Restriction enzymes cut DNA at certain sites to create multiple DNA fragments.
Restriction enzyme HindIII has known DNA fragment lengths and recognition sites
when digesting lambda DNA, while the lambda DNA recognition site for restriction
enzyme XhoI is unknown. The goal of this study is to determine the lambda
recognition site of XhoI by comparing a HindIII digest and a HindIII and XhoI
double digest on an electrophoresis gel. The HindIII digest had a band at 9.4 kb, but
this band was not visible in the double digest, therefore we concluded the recognition

XhoI is a type I restriction enzyme, so the location of the recognition site on
lambda DNA is unknown but it is known that it cleaves at CTCGAG sites (XhoI

The goal of this study is to determine the recognition site of XhoI on lambda DNA

To begin the process to determine the XhoI recognition site in the lamda DNA
fragment we first prepared 4 tubes of solutions containing 10X Optizyme reaction
buffer, sterile water, lambda DNA (0.3 ug/ul), XhoI (10u/ul, 3000u), and HindIII
(10u/ul, 7500ul). Tube 1 contained 2ul 10X Optizyme, 16ul sterile water, and 2ul

agenda As Golding s Lord of the Flies develops, power does also. When a group of
boys are stranded on an island, the boys power shifts dramatically. The boys fight for
power and savagery increases as the time on the island increases. The island was split
into groups due to Jacks fiend for power, this was the cause of all evil on the island.
Power on the island causes murder and great destruction. The conch is a symbol
for power and order, and Ralph controls the privilege of holding the power. When
Jack s character is introduced in chapter 2 he is already fighting for the power of
the conch with Ralph. The conches role was very significant in early chapters to
keep the boys on the same page but the power of the conch was quickly lost to
Jacks uprising. Jack s power takes over him and turns him into a savage creating
destruction to the whole island. They painted their faces like savages (Golding

On September 15 I went to La Bienvenida that was held on campus, in the Peace
Garden. Before this year, I have never heard of Hispanic heritage month and no one
meant it in my high school. The only time I heard about a group of being recognized
in a month is African Americans. This is commonly known as black history month,
or also known as African American history month, is widely recognized and
acknowledged. This is taken in February to remember important people and event

Have you ever watched a cartoon or a show or even a movie and you see yourself
in or through that character ? I recently found myself watching the movie Aladdin
with my little cousin. I saw how Jasmine stood up for the poor kids in the village

we connected in some way and how I am a lot like her. There are other characters
who I am also like in other ways. The three fictional characters that most represent my
personality are Donald Duckfor his short temper, Bubbles from The Powerpuff Girls
for her sweetness, and Jasmine from Aladdin for her way of standing up for what s
right and speaking her mind no matter who she is against. Although Donald is a
duck and a guy, we do have the common aspect of losing our temper after being
pushed to a certain point. In an episode on TV called Donald Duck cured duck,
Donald Duck loses his temper because he can t open the window and he starts
breaking plates and he even breaks through walls. Daisy Duckthen tells him she won
t go out on a date with him until he cures his temper. Donald Ducks temper makes
his friends not want to be around him. Although his friends tend to have patience
for him, in this episode Daisy Duck didn t and she refused to go out with him because
of his moodiness. Since Donald is very caring and doesn t want to lose his friends he
does what he can to keep his temper under control. Like Donald Duck, I do control

In both Letters from a Birmingham Jail and Antigone , Civil disobedience
becomes the answer to conflicts both fictional and in reality. Antigone and Martin
Luther King, Jr. are both icons for defying certain laws and people in charge in
attempts to fight for what they believe in. Although they both technically broke the
law to support a cause, Antigone and King had different approaches in doing so
and different goals. Antigone openly defied her leader in a direct way to accomplish
a singular goal, while King s defiance had a more complex goal in which his civil

The books were published starting when I was a young teenager, and I was not

Attitudes Between Men And Women
In the election held in 2013, only two WC made it to the parliament, although women
made up more than 53% of the electorate. These two candidates received the highest

The third positive theme spoke to the political skills of WC as they were evidenced
in a previous parliamentary term or as they were expected to show up after they win
election. Satisfaction with past political performance was a major subtheme of this
positive theme. Praise of the communication skills of these candidates represents
another major subtheme. Tweeters, especially women, placed a special emphasis on
the communication skills of the candidates, praising them for their eloquence,
sophistication as speakers, and cogent logic while talking to the media. They
described the speaking style of one WC as making the liver of the listener cooler (a
Kuwaiti colloquial expression connoting a good mood and a positive reaction).

Al Qaeda, or the Base in Arabic, is an arrangement of extremists initially organized by
Osama Bin Laden. Al Qaeda originated throughout the uprising against the Soviet
occupation of Afghanistan. Thousands of volunteers from around the Middle East
came to Afghanistan as mujahideen, warriors fighting to defend fellow Muslims. Bin
Laden returned to his native Saudi Arabia after the Soviets withdrew from
