1. CPCSEA Guidelines for Laboratory Animal
facility:- Anaesthesia , Euthanasia &
Physical facilities
SMBT College of Pharmacy, Dhamangoan, Nashik
Dr. Manoj Ramesh Kumbhare
Professor
3. CPCSEA:- means committee for the purpose of
control and supervision of experiments on animals.
• Need of CPCSEA :-
• To enhance animal well being and quality.
• To improve laboratory animal facility.
• To enhance biological knowledge.
• To ensure that lab animals are well maintained.
4. Anaesthesia
Defination:- drug which cause loss of sensation.
Types:- 1. General anaesthesia
2. Regional /Nerve block anaesthesia
3.Local anaesthesia
5. 1) General Anaesthesia:-
• Loss of sensory perception of entire body.
• unconscious person.
• Route – IV infusion/inhalation
• Eg:- Benzodiazepines( midazolin/diazepam)
6. 2) Regional / Nerve block Anaesthesia:-
• Loss of sensation over specific region of body(arm, leg,)
• No unconscious. Person remain conscious but can not feel any
pain any part of body.
• Method of administration- spinal block, Epidural block,
Regional block
• Eg :- Propofol , Ketamine .
7. 3) Local Anaesthesia:-
• Loss of sensation over a small limited area of body.
• No unconsciousness. Person remains conscious particular part
of body get loss of sensation.
• Method of administration- Topical application
• Eg :- Lidocaine , Lignocaine Hydrochloride Injection IP
solution.
8. Euthanasia
It is resorted to events where an animal is required to be sacrificed
on termination of an experiment or otherwise for ethical reasons.
Procedure carried out quickly and painlessly in an atmosphere free
from fear or anxiety.
The choice of method will depend on the nature of study, the species
of animal to be killed.
Methods :-
1. Physical methods- Electrocution, exsanguination, decapitation ,
cervical dislocation.
2. Drug administration- Overdose of chloral hydrate, Ketamine,
Chloroform.
9. • Requirements of Method:-
• Death, without causing anxiety, pain or distress with
minimum time lag phase.
• Minimum physiological and psychological
disturbances.
• Compatibility with the purpose of study and
minimum emotional effect on the operator.
• Location should be separate from animal and free
from environment contaminants.
10. Physical facilities:-
• Building material:- should be moisture proof, fire resistant,
seamless (smooth) materials most desirable for interior
surfaces including vermin and pest resistance.
• Corridor :- wide enough to facilitate the movement of
personnel as well as equipment and should be kept clean.
• Utilities :- water lines, drain pipes & electrical connections
should be outside the animal rooms
• Animal room door :- not be rust, dust proof ,vermin. fit
properly in their frames. door closures may also be provided.
• Exterior windows :- not recommended for small animals
,only for large animals.
11. • Floors :- monolithic / epoxy smooth, moisture proof,
nonabsorbent, skin-proof, resistant to wear, acid, solvents,
• Drains :- floor drains are not essential in all rooms, drainage
must be adequate to allow rapid removal of water , to prevent
the high humidity, drainage must be adequate to allow rapid
removal of water and drying surfaces.at the inlet and outlet of
the drains should be fitted with wire mesh guard to prevent
wild rodent entry
• Walls & Ceilings :- walls should be free of cracks, imperfect
junctions with doors, ceilings, floors, and corners.
• Storage areas :- separate storage areas should be designed for
feed, bedding, cages and materials not in use. Refrigerator
storage, separated from other cold storage , is essential for
storage of dead animals and animal tissue waste.
