El ácido acetohidroxi sintasa (AHAS) es una enzima tiamina dependiente que cataliza los primeros pasos de la ruta biosintética de los aminoácidos de cadena ramificada valina, leucina e isoleucina. Esta enzima se encuentra en plantas, hongos y bacterias por lo que es el principal blanco de acción de herbicidas, fungicidas y agentes antimicrobianos. El ciclo catalítico de la AHAS está constituido por cuatro pasos principales. En el primer paso el intermediario iluro realiza un ataque nucleofílico sobre una molécula de piruvato para dar paso a la formación del intermediario (2S)-2-Lactil-ThDP (L-ThDP). La formación de este intermediario necesita además la presencia de un hidrógeno el cual en la AHAS es de procedencia desconocida, debido a la inexistencia de grupos ionizables cercanos a las zonas del sitio activo de la enzima. Por este motivo en la presente tesis se estudió el mecanismo de reacción para la formación del L-ThDP, postulando que el grupo encargado de promover la transferencia protónica es el grupo amino del anillo de pirimidina cuando este se encuentra bajo la forma APH⁺. Para ello se realizó una exploración de la superficie de energía potencial (SEP) de la reacción considerando a los sustratos aislados en fase gas. Además se realizaron cálculos de las funciones de Fukui para el sustrato piruvato, para el estado de transición y para el intermediario iluro bajo las formas AP y APH⁺ con el fin de contrastar la reactividad de ambas formas tautomericas. Por último se realizaron cálculos termodinámicos utilizando medios de distintas constante dieléctrica, con el propósito de verificar la estabilidad de formación del intermediario iluro bajo la forma APH⁺ debido a la protonación del anillo pirimidínico por parte del aminoácido glutamato. Los cálculos computacionales se realizaron con el paquete computacional GAUSSIAN 03, empleando la teoría de los funcionales de densidad (DFT) a nivel B3LYP/6-31++G(d,p) para la exploración de la SEP y B3LYP(C-PCM)/6-311++G(d,p)//B3LYP/6-31++G(d,p) para los cálculos termodinámicos, en tanto los estudios de reactividad se realizaron con el paquete computacional Jaguar 7.7, empleando B3LYP/6-31++G(d,p). Los resultados obtenidos indicaron que la formación del L-ThDP ocurre de manera concertada, mostrando una barrera de activación de aproximadamente 21 Kcal/mol. Además se observó que es el iluro bajo la forma AP quien realiza el ataque nucleofílico sobre la molécula de piruvato.
