Exercise 9: Transfer of Bacteria: Aseptic Technique MCC- Bio 235 - Lab Evening Class Fall 2019 Dr. Jalili
Background • Inoculation- introducing to new culture media • Contamination- any unwanted microbes into the media • Aseptic technique- the technique used in microbiology to exclude contamination • Sterilization – removing any alive organisms from culture media, prior to use. This is accomplished by using autoclave.
Culture Media • Broth culture • Agar Slant • Agar Deep • Semisolid agar deep
The procedure
Materials (per group of 3-4) ➢ Culture media • 3 Nutrient broth • 3 Nutrient agar slant • 3 Nutrient agar deep ➢ Bacterial culture • Micrococcus luteus (broth) • Pseudomonas aeroginosa (broth) • Proteus vulgaris (slant)
Procedure (first period) • Instructor Demonstration • Follow P. 60-63, • and figure 3 on P. 61

Background • Inoculation- introducing to new culture media • Contamination- any unwanted microbes into the media • Aseptic technique- the technique used in microbiology to exclude contamination • Sterilization – removing any alive organisms from culture media, prior to use. This is accomplished by using autoclave.
  • 3. Culture Media • Broth culture • Agar Slant • Agar Deep • Semisolid agar deep
  • 5. Materials (per group of 3-4) ➢ Culture media • 3 Nutrient broth • 3 Nutrient agar slant • 3 Nutrient agar deep ➢ Bacterial culture • Micrococcus luteus (broth) • Pseudomonas aeroginosa (broth) • Proteus vulgaris (slant)
