3. What is callus?
callus is a growing mass of
unorganized plant parenchyma cells.
In living plants, callus cells are those cells that cover
a plant wound.
4. History of ovary and ovule culture
• Larue was probably the first to rise aseptic culture of
angiosperm flower.
• Technique of ovary culture was develop by Nitsch in
1951.
• The first report of gynogenesis was given by San Noem in
1976.(in case of barley)
5. Definition of ovary and ovule culture :-
Ovary culture:-
Ovary culture is the in-vitro technique which is carried outside on a
suitable nutrient medium in an unpollinated flower to grow new
haploid plants.
The process of ovary culture refers as “Gynogenesis” .
Ovule culture:-
Ovule culture is an elegant experimental system by which ovules are
aseptically isolated from the ovary .
Aseptically grown on chemically defined nutrient medium under
controlled conditions.
7. Selection of either opened or unopened of flower
Wash it with distilled water
Dip in 5% teepol solution/ or any detergent
Wash it 4-5 times with distilled water
Suface sterilization 5% sodium hypochloride (NaoCl)
8. Wash with distilled water
with the of Scalpel separate the ovary
Incubate the culture for 16 hours at 25⁰C
10. Collect the open flower where the anther
are decised and pollination has taken place.
Remove sepals,petals etc from the ovaries
containing either fertilized or unfertilized ovuleA
Soak it in 6% NaoCl solution
Rinse the ovaries 3-4 times times with sterile
distilled water
11. Ovules are gently prodded with the help of spatula by
breaking the funicles at its junction with placental tissue.
Spatula with ovule is gently lowered into the sterile solid or
liquid medium as the culture vial is slanted about 45⁰.
Incubate the ovule either in dark or light for 16 hrs at 25⁰C
14. Application of ovary and ovule culture :-
It is useful to study the early development of embryo
development .
To study the effect of Phytohormones on parthenocaroic fruits
development.
To study the fruit development and also different type of fruit
physiology
