2. Rifampicin
• It was isolated from Streptomyces
mediterranei in 1957 and 7 rifamycin
developed ( A,B,C,D,E,S,SV).
• They are mainly useful in treating
Tuberculosis.

3. Mode of action
• It is a group of macrocyclic antibiotics which
inhibits DNA dependent RNA polymerase.
• Mammals arent affected because it binds to
peptide chain of bacteria.

4. Vectors for DNA sequencing
• DNA sequencing:The process by which nucleic
acid sequence is determined.
• Vectors used:
These are also called as cloning
vectors which must have important features like,
1)Ori
2) selectable marker
3)Unique restriction site
4)Gene inactivation marker

4. YACs

6. Plasmid
• It can hold upto DNA size of 10kb.
• Eg:pBR322

7. Phage
• It can hold upto DNA size of 20 kb.
• Eg:M13

8. Cosmid
• This can hold upto DNA size of 45 kb.

9. YAC
• It can hold upto DNA size of 800 kb.

10. Hosts for cloning vectors
• Escherichia coli
• Bacillus subtilis
• Sacchromyces cerevisiae
• Mammalian cells
