The fruits and vegetables are rich in nutritional value which make them an easy target of organisms causing spoilage and soft rot. The soft rot is one of the destructive disease of fruits and vegetables and occurs worldwide wherever fleshy storage tissues of fruits and vegetables are found. Such disease can occurred by some bacteria and fungi which are highly responsible for this soft rot causing spoilage and they are endowed with an important enzymological power, the main action of their pathogenicity. The enzyme pectinase has the power of degrading the cell wall of plant, causing the liquefaction of tissues which favour the development of phytopathogens. The understanding the role of enzymes synthesizing by pathogens causing spoilage and need to develop the new protection approach to deal with it. In fact plants defense against stresses by different ways. The role of cellular organelles is very important in this way. Cell wall and their derivatives such as oligosaccharins as biochemical defenser or for example trichomes as mechanical defenser is the frontline of the plant defense system. Also Plants have evolved a multi-layered immune system that dynamically responds to pathogens alike cell membrane that is a key mediator of communication between plants and microbes. Cytoplasm and the membrane-bounded structures (organelles) defense against different kind of stresses. The role of cellular organelles in plant defense relate to their enzymes primarily. Enzymes such as proteases, esterases and ribonucleases in cytoplasm, PM H+-ATPases in plasma membrane or β glucosidases included cyanogenic glucosides, saponins, glucosinolates or DIMBOA (2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one) glucoside in ER are responsible for plant defense. Biosynthesis and Degradation of Carotenoids in Ornamental Crops with specific...IJEAB
Carotenoids are lipophilic secondary metabolites derived from the isoprenoid pathway, accumulated in most plant organs and widely used as an antioxidant. Carotenoids synthesized in chloroplasts are essential for protecting tissues against photo-oxidative damage in the green tissues of higher plants. The importance of carotenoids for plant growth and development is evident since at least two major phytohormones, strigolactones and abscisic acid, are derived from carotenoid precursors. In flowers, carotenoids synthesized in the chromoplasts provide colour to the petals, ranging from yellow to red, in order to attract pollinators and determines the commercial value of ornamental plants. On analysis in chrysanthemum, β, ɛ-carotenoids, lutein and its derivatives, reflecting the high expression levels of lycopene ɛ-cyclase (LCYE) were found in yellow petals compared to the ratio of β, β-carotenoids to total carotenoids found in leaves reflecting the high expression levels of lycopene β-cyclase (LCYB). Petals of the yellow-flowered cultivar Yellow Paragon showed increased accumulation and drastic componential changes of carotenoids as they mature, compared to petals of the white-flowered cultivar Paragon that showed drastically decreased carotenoid content during petal development.The white petals of chrysanthemum (Chrysanthemum morifolium Ramat.) contain a factor that inhibits the accumulation of carotenoids. All the white-flowered chrysanthemum cultivars tested showed high levels of CmCCD4a transcript in their petals, whereas most of the yellow flowered cultivars showed extremely low levels indicating that in white petals of chrysanthemums, carotenoids are synthesized but subsequently degraded into colourless compounds, which results in the white colour. Studying the regulatory mechanisms underlying carotenoid accumulation in ornamental plants at the molecular level will help in producing novel coloured cultivars by plant transformation.

— Natural and synthetic estrogens have been detected in rivers, lakes and estuaries in many parts of the world. Primary sources of these compounds are domestic and industrial effluents, which are not deleted after the water treatment. Estrogen has been the endocrine disruptor most researched to be very active biologically and be the etiologic agent of diverse types of cancer and other conditions such as endometriosis, precocious puberty, feminization, masculinization, sterility. In this context, we use water of 36 natural reservoirs or dams, in a bioassay to characterize their estrogenicity in culture of MCF7 cells and obtained high concentration of estrogen in samples taken in Ibiúna and Equestrian Santo Amaro / SP. However, certain concentration in our samples for most water samples from different regions was very close to the limit of quantification by bioassay and estrogen was in fmol. Plant growth-promoting rhizobacteria (PGPR) facilitate the plant growth and enhance their
induced systemic resistance (ISR) against a variety of environmental stresses. In this study,
we carried out integrative analyses on the proteome, transcriptome, and metabolome to investigate
Arabidopsis root and shoot responses to the well-known PGPR strain Paenibacillus
polymyxa (P. polymyxa) E681. Shoot fresh and root dry weights were increased, whereas root
length was decreased by treatment with P. polymyxa E681. 2DE approach in conjunction
with MALDI-TOF/TOF analysis revealed a total of 41 (17 spots in root, 24 spots in shoot)
that were differentially expressed in response to P. polymyxa E681. Biological process- and
molecular function-based bioinformatics analysis resulted in their classification into seven different
protein groups. Of these, 36 proteins including amino acid metabolism, antioxidant,
defense and stress response, photosynthesis, and plant hormone-related proteins were upregulated,
whereas five proteins including three carbohydrate metabolism- and one amino
acid metabolism-related, and one unknown protein were down-regulated, respectively. A good
correlation was observed between protein and transcript abundances for the 12 differentially
expressed proteins during interactions as determined by qPCR analysis. Metabolite analysis
using LC-MS/MS revealed highly increased levels of tryptophan, indole-3-acetonitrile (IAN),
indole-3-acetic acid (IAA), and camalexin in the treated plants. Arabidopsis plant inoculated
P. polymyxa E681 also showed resistance to Botrytis cinerea infection. Taken together these
results suggest that P. polymyxa E681 may promote plant growth by induced metabolism and
activation of defense-related proteins against fungal pathogen.

Suillus species, in general, are edible mushrooms, and environmentally important that are associated
mostly with pine trees in the tropics regions. These fungi considered a remarkable source of phenolic
compounds that play a crucial role as antioxidants which may reduce the risk of most human chronic
diseases such as cancer, diabetes, asthma, atherosclerosis, Alzheimer, and others. On the other hand,
carotenoids (β carotene) are the most popular natural pigments which play an important role to protect
the plants from photo-oxidative reactions. In human, these compounds prevent oxidative stress and
expects to have antimicrobial activity. Here, the phenolic compounds were extracted with Ethyl acetate
from fruiting bodies of Suillus sp and analyzed by HPLC, the antioxidant activity (reducing power%) of
phenolic compounds was determined at the concentrations of 1, 2.5, and 5 mg/mL. Antimicrobial activity
of β carotene pigment was measured at a concentration of 100 mg/mL against some human pathogenic
bacteria such as Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia, and Staphylococcus
aureus. The specific DNA region ITS was amplified and sequenced using ITS1 and ITS4 primers with
some bioinformatics analyses. The phenolic extract isolated from fruiting bodies of Suillus sp showed a
remarkable antioxidant activity by increasing the reducing power percent (from F+3 ions to F+2 ions)
comparing with the industrial antioxidant (Propyl gallate) at all used concentrations. Percent of reducing
power of phenolic compounds were 75.5, 84.9 and 95.7% at concentrations of 1, 2.5, and 5 mg/mL
respectively; comparing with PG were 65.9, 81.3, and 93.3 at 1, 2.5, and 5 mg/mL respectively. The β
carotene pigment revealed a significant antimicrobial activity at a concentration of 100 mg/mL against K.
pneumonia, E. coli, and S. aureus. The highest bacterial growth inhibition was against K. pneumonia (40
mm), followed by E. coli (36 mm) and S. aureus (31 mm), while no effect showed against P. aeruginosa.
Our outcomes revealed that the phenolic bioactive compounds can be used as a natural antioxidant
instead of the industrial antioxidants, and also a β carotene pigment could be applied as a promising
natural compound rather than using the antibiotics and other manufactured compounds to inhibit
bacteria activity.
MICROORGANISMS ASSOCIATED WITH THE SPOILAGE OF CUCUMBER, GARDEN EGG AND PAWPA...paperpublications3
Abstract: A total of nine cucumbers, each of Garden egg and pawpaw samples were collected from Wurukum, High level and Wadata markets and cultured on appriopate agar, for colony count and isolation of bacteria according to their cultural and biochemical characteristics. The results revealed that garden egg from High Level Market had the highest bacterial count (1.9x105cfu/g) and the least was pawpaw from High Level Market (1.1 x 105 cfu/g). However, it was not statistically significant. The bacteria isolated were; Propianol bacteria (23.3%), Escherichia coli (16.6%), Staphylococcus aureus (36.7%), Bacillus spp (10.0%) and Corynebacteria (13.3%). The fungal isolates were Aspergillus flavus (10%), Aspergillus fumigatus (20%), Aspergillus nidulus (10%), Aspergillus terreus (20%) and mucor (40%). The result of this study shows fruits sold in Wurukum, High Level Market and Wadata Market are contaminated and may cause harm to consumers, so measures such as proper handling should be taken to control the contamination of these fruits.

Microbe-assisted phytoremediation is a recent application of bioremediation with much prospects. The genetic relatedness of culturable endophytic bacteria of wetland plants growing on a six month-old and twelve month-old petroleum-contaminated sites, and an uncontaminated site in Bayelsa State of the Niger Delta Region, Nigeria were compared. Most of the endophyte species isolated from the roots, stems and leaves were common to all the sites and belong to the phyla Proteobacteria, Bacteroidetes Firmicutes, Actinobacteria, Chloroflexi and Actinomicrobia, with the γ-Proteobacteria dominating. Pseudomonas was the most prevalent species in all three sites, but higher in the petroleum contaminated sites. Biochemical properties (API 20E) of the common isolates; Pseudomonas spp.Chryseobacterium indologenes,Bacillus and Proteusvaried with sites while only Providencia rettgeri peculiar to the petroleum-contaminated sites showed the same properties. 16S rRNA PCR-DNA fragments of forty-five species of the isolates (15 from each site) were characterized using RFLP and MspI restriction enzyme and a genetic distance tree of the restriction patterns drawn. The percentage of similarity in the genetic relatedness of isolates ranged from 11.1 – 100%. The genetic tree analysis of the 45 species of identified bacteria revealed 3 major clusters with 17 DNA fingerprinting patterns. Pseudomonas species of the root and leaves of the six month-old petroleum-contaminated site and uncontaminated site were seen to cluster together irrespective of date of isolation. The endophytes may play a role in the in situ degradation of the petroleum hydrocarbon of the sites.

Plant Growth promoting rhizobacteria are a heterogeneous group of bacteria that can be found in the rhizosphere, at root surfaces and in association with roots. They benefit plants through Production of plant hormones, such as auxins, asymbiotic N2 fixation, solubilization of mineral phosphates, antagonism against phytopathogenic microorganisms by production of antibiotics, siderophroes, Chitinase and other nutrients ability to effectively colonize roots are responsible for plant growth promotion. An experiment was conducted in the field of National Institute of Agronomic Research of Meknes. Morocco. The experiment was a completely randomized design with six replicates. There were four treatments viz. T1: (control; N0 -PGPR), T2: (N0 +2027-2), T3: (N0 +2066-7) and T4: (N0+2025-1). The results indicated that a remarkable increase in root growth, namely length, the diameter of the rod and the total chlorophyll. A total of three different bacteria colonies were isolated and proceed with in vitro screening for plant growth promoting activities; phosphate solubilization, nitrogen fixation, indole acetic acid (IAA), ammonia production and antimicrobial enzymes (cellulose, chitinase and protease) activity. Among the three bacterial strains, all bacterial strains are able to produce ammonia, IAA production and nitrogen fixation activity, one strain phosphate solubilizing activity, two strain are able to produce cellulase syntheses, Protease activity and Chitinase activity.

Enzymes such as proteases, esterases and ribonucleases in cytoplasm, PM H+-ATPases in plasma membrane or β glucosidases included cyanogenic glucosides, saponins, glucosinolates or DIMBOA (2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one) glucoside in ER are responsible for plant defense. Also ROSs plus SA and JA in chloroplast and mitochondria play an important role in immune plant system. In nucleus macromolecules including nucleoporins, importins, and Ran-GTP-related components, are essential to mount an efficient immune response in response to different pathogens. And in Golgi apparatus, peroxysomes and vacuoles, glycosyltransferases, myrosinase and hydrolytic enzymes are liable for plant defense respectively. Biosynthesis and Degradation of Carotenoids in Ornamental Crops with specific...IJEAB
Carotenoids are lipophilic secondary metabolites derived from the isoprenoid pathway, accumulated in most plant organs and widely used as an antioxidant. Carotenoids synthesized in chloroplasts are essential for protecting tissues against photo-oxidative damage in the green tissues of higher plants. The importance of carotenoids for plant growth and development is evident since at least two major phytohormones, strigolactones and abscisic acid, are derived from carotenoid precursors. In flowers, carotenoids synthesized in the chromoplasts provide colour to the petals, ranging from yellow to red, in order to attract pollinators and determines the commercial value of ornamental plants. On analysis in chrysanthemum, β, ɛ-carotenoids, lutein and its derivatives, reflecting the high expression levels of lycopene ɛ-cyclase (LCYE) were found in yellow petals compared to the ratio of β, β-carotenoids to total carotenoids found in leaves reflecting the high expression levels of lycopene β-cyclase (LCYB). Petals of the yellow-flowered cultivar Yellow Paragon showed increased accumulation and drastic componential changes of carotenoids as they mature, compared to petals of the white-flowered cultivar Paragon that showed drastically decreased carotenoid content during petal development.The white petals of chrysanthemum (Chrysanthemum morifolium Ramat.) contain a factor that inhibits the accumulation of carotenoids. All the white-flowered chrysanthemum cultivars tested showed high levels of CmCCD4a transcript in their petals, whereas most of the yellow flowered cultivars showed extremely low levels indicating that in white petals of chrysanthemums, carotenoids are synthesized but subsequently degraded into colourless compounds, which results in the white colour. Studying the regulatory mechanisms underlying carotenoid accumulation in ornamental plants at the molecular level will help in producing novel coloured cultivars by plant transformation.

— Natural and synthetic estrogens have been detected in rivers, lakes and estuaries in many parts of the world. Primary sources of these compounds are domestic and industrial effluents, which are not deleted after the water treatment. Estrogen has been the endocrine disruptor most researched to be very active biologically and be the etiologic agent of diverse types of cancer and other conditions such as endometriosis, precocious puberty, feminization, masculinization, sterility. In this context, we use water of 36 natural reservoirs or dams, in a bioassay to characterize their estrogenicity in culture of MCF7 cells and obtained high concentration of estrogen in samples taken in Ibiúna and Equestrian Santo Amaro / SP. However, certain concentration in our samples for most water samples from different regions was very close to the limit of quantification by bioassay and estrogen was in fmol. Plant growth-promoting rhizobacteria (PGPR) facilitate the plant growth and enhance their
induced systemic resistance (ISR) against a variety of environmental stresses. In this study,
we carried out integrative analyses on the proteome, transcriptome, and metabolome to investigate
Arabidopsis root and shoot responses to the well-known PGPR strain Paenibacillus
polymyxa (P. polymyxa) E681. Shoot fresh and root dry weights were increased, whereas root
length was decreased by treatment with P. polymyxa E681. 2DE approach in conjunction
with MALDI-TOF/TOF analysis revealed a total of 41 (17 spots in root, 24 spots in shoot)
that were differentially expressed in response to P. polymyxa E681. Biological process- and
molecular function-based bioinformatics analysis resulted in their classification into seven different
protein groups. Of these, 36 proteins including amino acid metabolism, antioxidant,
defense and stress response, photosynthesis, and plant hormone-related proteins were upregulated,
whereas five proteins including three carbohydrate metabolism- and one amino
acid metabolism-related, and one unknown protein were down-regulated, respectively. A good
correlation was observed between protein and transcript abundances for the 12 differentially
expressed proteins during interactions as determined by qPCR analysis. Metabolite analysis
using LC-MS/MS revealed highly increased levels of tryptophan, indole-3-acetonitrile (IAN),
indole-3-acetic acid (IAA), and camalexin in the treated plants. Arabidopsis plant inoculated
P. polymyxa E681 also showed resistance to Botrytis cinerea infection. Taken together these
results suggest that P. polymyxa E681 may promote plant growth by induced metabolism and
activation of defense-related proteins against fungal pathogen.

Suillus species, in general, are edible mushrooms, and environmentally important that are associated
mostly with pine trees in the tropics regions. These fungi considered a remarkable source of phenolic
compounds that play a crucial role as antioxidants which may reduce the risk of most human chronic
diseases such as cancer, diabetes, asthma, atherosclerosis, Alzheimer, and others. On the other hand,
carotenoids (β carotene) are the most popular natural pigments which play an important role to protect
the plants from photo-oxidative reactions. In human, these compounds prevent oxidative stress and
expects to have antimicrobial activity. Here, the phenolic compounds were extracted with Ethyl acetate
from fruiting bodies of Suillus sp and analyzed by HPLC, the antioxidant activity (reducing power%) of
phenolic compounds was determined at the concentrations of 1, 2.5, and 5 mg/mL. Antimicrobial activity
of β carotene pigment was measured at a concentration of 100 mg/mL against some human pathogenic
bacteria such as Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia, and Staphylococcus
aureus. The specific DNA region ITS was amplified and sequenced using ITS1 and ITS4 primers with
some bioinformatics analyses. The phenolic extract isolated from fruiting bodies of Suillus sp showed a
remarkable antioxidant activity by increasing the reducing power percent (from F+3 ions to F+2 ions)
comparing with the industrial antioxidant (Propyl gallate) at all used concentrations. Percent of reducing
power of phenolic compounds were 75.5, 84.9 and 95.7% at concentrations of 1, 2.5, and 5 mg/mL
respectively; comparing with PG were 65.9, 81.3, and 93.3 at 1, 2.5, and 5 mg/mL respectively. The β
carotene pigment revealed a significant antimicrobial activity at a concentration of 100 mg/mL against K.
pneumonia, E. coli, and S. aureus. The highest bacterial growth inhibition was against K. pneumonia (40
mm), followed by E. coli (36 mm) and S. aureus (31 mm), while no effect showed against P. aeruginosa.
Our outcomes revealed that the phenolic bioactive compounds can be used as a natural antioxidant
instead of the industrial antioxidants, and also a β carotene pigment could be applied as a promising
natural compound rather than using the antibiotics and other manufactured compounds to inhibit
bacteria activity.

Abstract: A total of nine cucumbers, each of Garden egg and pawpaw samples were collected from Wurukum, High level and Wadata markets and cultured on appriopate agar, for colony count and isolation of bacteria according to their cultural and biochemical characteristics. The results revealed that garden egg from High Level Market had the highest bacterial count (1.9x105cfu/g) and the least was pawpaw from High Level Market (1.1 x 105 cfu/g). However, it was not statistically significant. The bacteria isolated were; Propianol bacteria (23.3%), Escherichia coli (16.6%), Staphylococcus aureus (36.7%), Bacillus spp (10.0%) and Corynebacteria (13.3%). The fungal isolates were Aspergillus flavus (10%), Aspergillus fumigatus (20%), Aspergillus nidulus (10%), Aspergillus terreus (20%) and mucor (40%). The result of this study shows fruits sold in Wurukum, High Level Market and Wadata Market are contaminated and may cause harm to consumers, so measures such as proper handling should be taken to control the contamination of these fruits.

Microbe-assisted phytoremediation is a recent application of bioremediation with much prospects. The genetic relatedness of culturable endophytic bacteria of wetland plants growing on a six month-old and twelve month-old petroleum-contaminated sites, and an uncontaminated site in Bayelsa State of the Niger Delta Region, Nigeria were compared. Most of the endophyte species isolated from the roots, stems and leaves were common to all the sites and belong to the phyla Proteobacteria, Bacteroidetes Firmicutes, Actinobacteria, Chloroflexi and Actinomicrobia, with the γ-Proteobacteria dominating. Pseudomonas was the most prevalent species in all three sites, but higher in the petroleum contaminated sites. Biochemical properties (API 20E) of the common isolates; Pseudomonas spp.Chryseobacterium indologenes,Bacillus and Proteusvaried with sites while only Providencia rettgeri peculiar to the petroleum-contaminated sites showed the same properties. 16S rRNA PCR-DNA fragments of forty-five species of the isolates (15 from each site) were characterized using RFLP and MspI restriction enzyme and a genetic distance tree of the restriction patterns drawn. The percentage of similarity in the genetic relatedness of isolates ranged from 11.1 – 100%. The genetic tree analysis of the 45 species of identified bacteria revealed 3 major clusters with 17 DNA fingerprinting patterns. Pseudomonas species of the root and leaves of the six month-old petroleum-contaminated site and uncontaminated site were seen to cluster together irrespective of date of isolation. The endophytes may play a role in the in situ degradation of the petroleum hydrocarbon of the sites.

Plant Growth promoting rhizobacteria are a heterogeneous group of bacteria that can be found in the rhizosphere, at root surfaces and in association with roots. They benefit plants through Production of plant hormones, such as auxins, asymbiotic N2 fixation, solubilization of mineral phosphates, antagonism against phytopathogenic microorganisms by production of antibiotics, siderophroes, Chitinase and other nutrients ability to effectively colonize roots are responsible for plant growth promotion. An experiment was conducted in the field of National Institute of Agronomic Research of Meknes. Morocco. The experiment was a completely randomized design with six replicates. There were four treatments viz. T1: (control; N0 -PGPR), T2: (N0 +2027-2), T3: (N0 +2066-7) and T4: (N0+2025-1). The results indicated that a remarkable increase in root growth, namely length, the diameter of the rod and the total chlorophyll. A total of three different bacteria colonies were isolated and proceed with in vitro screening for plant growth promoting activities; phosphate solubilization, nitrogen fixation, indole acetic acid (IAA), ammonia production and antimicrobial enzymes (cellulose, chitinase and protease) activity. Among the three bacterial strains, all bacterial strains are able to produce ammonia, IAA production and nitrogen fixation activity, one strain phosphate solubilizing activity, two strain are able to produce cellulase syntheses, Protease activity and Chitinase activity.

Background and Objective Telehealth has become a well known tool for the delivery of health care in Saudi Arabia, and the perspective and knowledge of healthcare providers are influential in the implementation, adoption and advancement of the method. This systematic review was conducted to examine the current literature base regarding telehealth and the related healthcare professional perspective and knowledge in the Kingdom of Saudi Arabia. Materials and Methods This systematic review was conducted by searching 7 databases including, MEDLINE, CINHAL, Web of Science, Scopus, PubMed, PsycINFO, and ProQuest Central. Studies on healthcare practitioners telehealth knowledge and perspectives published in English in Saudi Arabia from 2000 to 2023 were included. Boland directed this comprehensive review. The researchers examined each connected study using the AXIS tool, which evaluates cross sectional systematic reviews. Narrative synthesis was used to summarise and convey the data. Results Out of 1840 search results, 10 studies were included. Positive outlook and limited knowledge among providers were seen across trials. Healthcare professionals like telehealth for its ability to improve quality, access, and delivery, save time and money, and be successful. Age, gender, occupation, and work experience also affect health workers knowledge. In Saudi Arabia, healthcare professionals face inadequate expert assistance, patient privacy, internet connection concerns, lack of training courses, lack of telehealth understanding, and high costs while performing telemedicine. Conclusions Healthcare practitioners telehealth perceptions and knowledge were examined in this systematic study. Its collection of concerned experts different personal attitudes and expertise would help enhance telehealths implementation in Saudi Arabia, develop its healthcare delivery alternative, and eliminate frequent problems. This research investigates the nexus between online discussions on Dr. B.R. Ambedkars ideals and their impact on social inclusion among college students in Gurugram, Haryana. Surveying 240 students from 12 government colleges, findings indicate that 65 actively engage in online discussions, with 80 demonstrating moderate to high awareness of Ambedkars ideals. Statistically significant correlations reveal that higher online engagement correlates with increased awareness p 0.05 and perceived social inclusion. Variations across colleges and a notable effect of college type on perceived social inclusion highlight the influence of contextual factors. Furthermore, the intersectional analysis underscores nuanced differences based on gender, caste, and socio economic status. This study investigated the impact of educational background and professional training on human rights awareness among secondary school teachers in the Marathwada region of Maharashtra, India. The key findings reveal that higher levels of education, particularly a master’s degree, and fields of study related to education, humanities, or social sciences are associated with greater human rights awareness among teachers. Additionally, both pre service teacher training and in service professional development programs focused on human rights education significantly enhance teacher’s knowledge, skills, and competencies in promoting human rights principles in their classrooms. The Role of Mentoring and Its Influence on the Effectiveness of the Teaching ...ijtsrd
This paper reports on a study which was conducted to investigate the role of mentoring and its influence on the effectiveness of the teaching of Physics in secondary schools in the South West Region of Cameroon. The study adopted the convergent parallel mixed methods design, focusing on respondents in secondary schools in the South West Region of Cameroon. Both quantitative and qualitative data were collected, analysed separately, and the results were compared to see if the findings confirm or disconfirm each other. The quantitative analysis found that majority of the respondents 72 of Physics teachers affirmed that they had more experienced colleagues as mentors to help build their confidence, improve their teaching, and help them improve their effectiveness and efficiency in guiding learners’ achievements. Only 28 of the respondents disagreed with these statements. With majority respondents 72 agreeing with the statements, it implies that in most secondary schools, experienced Physics teachers act as mentors to build teachers’ confidence in teaching and improving students’ learning. The interview qualitative data analysis summarized how secondary school Principals use meetings with mentors and mentees to promote mentorship in the school milieu. This has helped strengthen teachers’ classroom practices in secondary schools in the South West Region of Cameroon. With the results confirming each other, the study recommends that mentoring should focus on helping teachers employ social interactions and instructional practices feedback and clarity in teaching that have direct measurable impact on students’ learning achievements. This study primarily focuses on the design of a high side buck converter using an Arduino microcontroller. The converter is specifically intended for use in DC DC applications, particularly in standalone solar PV systems where the PV output voltage exceeds the load or battery voltage. To evaluate the performance of the converter, simulation experiments are conducted using Proteus Software. These simulations provide insights into the input and output voltages, currents, powers, and efficiency under different state of charge SoC conditions of a 12V,70Ah rechargeable lead acid battery. Additionally, the hardware design of the converter is implemented, and practical data is collected through operation, monitoring, and recording. By comparing the simulation results with the practical results, the efficiency and performance of the designed converter are assessed. The findings indicate that while the buck converter is suitable for practical use in standalone PV systems, its efficiency is compromised due to a lower output current. Energy becomes sustainable if it meets the needs of the present without compromising the ability of future generations to meet their own needs. Some of the definitions of sustainable energy include the considerations of environmental aspects such as greenhouse gas emissions, social, and economic aspects such as energy poverty. Generally far more sustainable than fossil fuel are renewable energy sources such as wind, hydroelectric power, solar, and geothermal energy sources. Worthy of note is that some renewable energy projects, like the clearing of forests to produce biofuels, can cause severe environmental damage. The sustainability of nuclear power which is a low carbon source is highly debated because of concerns about radioactive waste, nuclear proliferation, and accidents. The switching from coal to natural gas has environmental benefits, including a lower climate impact, but could lead to delay in switching to more sustainable options. “Carbon capture and storage” can be built into power plants to remove the carbon dioxide CO2 emissions, but this technology is expensive and has rarely been implemented. Leading non renewable energy sources around the world is fossil fuels, coal, petroleum, and natural gas. Nuclear energy is usually considered another non renewable energy source, although nuclear energy itself is a renewable energy source, but the material used in nuclear power plants is not. This paper aims to outline the executive regulations, survey standards, and specifications required for the implementation of the Sudan Survey Act, and for regulating and organizing all surveying work activities in Sudan. The act has been discussed for more than 5 years. The Land Survey Act was initiated by the Sudan Survey Authority and all official legislations were headed by the Sudan Ministry of Justice till it was issued in 2022. The paper presents conceptual guidelines to be used for the Survey Act implementation and to regulate the survey work practice, standardizing the field surveys, processing, quality control, procedures, and the processes related to survey work carried out by the stakeholders and relevant authorities in Sudan. The conceptual guidelines are meant to improve the quality and harmonization of geospatial data and to aid decision making processes as well as geospatial information systems. The established comprehensive executive regulations will govern and regulate the implementation of the Sudan Survey Geomatics Act in all surveying and mapping practices undertaken by the Sudan Survey Authority SSA and state local survey departments for public or private sector organizations. The targeted standards and specifications include the reference frame, projection, coordinate systems, and the guidelines and specifications that must be followed in the field of survey work, processes, and mapping products. Towards the Implementation of the Sudan Interpolated Geoid Model Khartoum Sta...ijtsrd
The discussions between ellipsoid and geoid have invoked many researchers during the recent decades, especially during the GNSS technology era, which had witnessed a great deal of development but still geoid undulation requires more investigations. To figure out a solution for Sudans local geoid, this research has tried to intake the possibility of determining the geoid model by following two approaches, gravimetric and geometrical geoid model determination, by making use of GNSS leveling benchmarks at Khartoum state. The Benchmarks are well distributed in the study area, in which, the horizontal coordinates and the height above the ellipsoid have been observed by GNSS while orthometric heights were carried out using precise leveling. The Global Geopotential Model GGM represented in EGM2008 has been exploited to figure out the geoid undulation at the benchmarks in the study area. This is followed by a fitting process, that has been done to suit the geoid undulation data which has been computed using GNSS leveling data and geoid undulation inspired by the EGM2008. Two geoid surfaces were created after the fitting process to ensure that they are identical and both of them could be counted for getting the same geoid undulation with an acceptable accuracy. In this respect, statistical operation played an important role in ensuring the consistency and integrity of the model by applying cross validation techniques splitting the data into training and testing datasets for building the geoid model and testing its eligibility. The geometrical solution for geoid undulation computation has been utilized by applying straightforward equations that facilitate the calculation of the geoid undulation directly through applying statistical techniques for the GNSS leveling data of the study area to get the common equation parameters values that could be utilized to calculate geoid undulation of any position in the study area within the claimed accuracy. Both systems were checked and proved eligible to be used within the study area with acceptable accuracy which may contribute to solving the geoid undulation problem in the Khartoum area, and be further generalized to determine the geoid model over the entire country, and this could be considered in the future, for regional and continental geoid model. The production of pectic enzyme in vivo is usually
proven by the removal of infected tissue from the sick
plant, purification of the crude enzymes and
determination of its ability to reproduce disease
symptoms in a healthy plant. Evidence of pectinase
activity can sometimes be obtained microscopically
as the invaded cells will be seen to separate along the
line of the middle lamella and subsequent loss of their
staining ability (Simbo Aboaba 2009).In this study,
an attempt has been made to review the relevant
available literature concerning the role of pectinase
causing the soft rot in fruits and vegetables.
Pectin –its structure and properties
Pectin is a high molecular weight heterogeneous and
acidic structural polysaccharide which is one of the
major constituents of cereals, vegetables, fruits and
fibers (O’Neil MA et al 1996). It is the component of
middle lamella and primary cell wall in plant cell wall
and within the wall forms a matrix in which a
network of cellulose and hemicelluloses are
embedded (Caffall KH et al 2009). Pectin has been
characterized as having a backbone of D-galacturonic
acid residues, linked by α (1–4) linkage with a small
number of rhamnose residues in the main chain and a
rabinose, galactose and xylose on its side chains
(Gummadi SN 2002).
Pectin contributes to the mechanical strength and
physical properties of primary cell walls there by
maintaining structural integrity. Pectic substances
represent between 0.5-4% of fresh weight plant
material (Jayani RS et al 2005 and Sakai T et al
1993)). In addition to their role as cementing and
lubricating agents in the cell walls of higher plants,
they are responsible for the texture of fruits and
vegetables during growth, maturation and their
storage (Alkorta I et al 1998 and Caffall KH et al
2009). Furthermore, pectic substances are involved in
the interaction between plant hosts and their
pathogens (Collmer A et al 1986).
Pectinase and Classification of Pectinase Enzymes
Pectinase are the group of enzymes that catalyze the
breakdown of substrates containing pectin. These are
the one which hydrolyze pectic substances. Pectic
enzymes are widely distributed in nature and are
produced by bacteria, yeast, fungi and plants. In
plants, pectic enzymes are very important since they
play a role in cellular growth as well as in fruit
ripening. Pectolytic activity of microorganisms plays
a significant role, firstly, in the pathogenesis of plants
since these enzymes are the first to attack the tissue
(Collmer A et al 1986). In addition, they are also
involved in the process of symbiosis and the decay of
vegetable residues (Lang C et al 2000).
Pectinases are the heterogeneous group of enzymes
that are classified into polygalcturonase (PG),
pectinesterase (PE), and pectin lyase (PL) based on
their mode of action on the substrate. These enzymes
break the pectin polymer into smaller fragments by
hydrolysis, trans-elimination and de-esterification
reactions, respectively. There are two main enzymes
reported, which able to hydrolyze glycosidic linkages.
First is polygalacturonase (PG), also known as pectin
depolymerise enzymes which act on α 1→4
glycosidic bond of the main chain of polygalacturonic
acid and hydrolyse it by release of a water molecule.
Second is polymethylgalacturonases (PMG) enzymes
which catalyze the hydrolytic cleavage of α (1→4)
glycosidic bonds.
Enzymes that involve in de-esterification reaction
include pectinesterases (PE), also known as pectin
methyl hydrolase. They catalyze de-esterification of
the methoxyl group of pectin and yields pectic acid.
Enzymes that cleave α(1→4) glycosidic linkage by
transelimination reaction include polymethyl
galacturonate lyases (PMGL) and catalyze trans-
eliminative cleavage of pectin and produce
unsaturated galacturonates (Heena Verma et al 2018).
Pectinolytic Enzyme Production
Pectinolytic enzymes are produced by many
organisms like bacteria, fungi, yeasts, insects,
nematodes, protozoan and plants. The microbial
world has shown to be very heterogeneous in its
ability to synthesize different types of Pectolytic
enzymes with different mechanisms of action and
biochemical properties (Anil RS 2018). Many
pathogenic bacteria and fungi are capable of
degrading pectin (Bateman DF 1972).a) Pectinolytic
Bacteria: Bacteria like Bacillus, Pseudomonas and
Micrococcus isolated from retting flax, jute, sisal and
coir and Erwinia from coffee fruits have shown to
possess the ability to degrade pectin by producing
pectinolytic enzymes (Chesson A 1980).b)
Pectinolytic fungi: Many fungal species are capable
of degrading pectin by producing different
pectinolytic enzymes. The study of Shinidia AA
(1995) showed that temperature variation during
garbage composting led to corresponding changes in
the distribution of pectin degrading fungi in the
compost and the most common pectinolytic fungi
were Aspergillus niger, Aspergillus flavus,
Aspergillus terreus, Penicillium chrysogenum,
Fusarium moniliforme, Alternaria alternate,
Cladosporium cladosporioids and Trichoderma
reesei and these fungi also formed the part of
pectinolytic microflora of coffee fruits.

Pectinase enzyme involve in the soft rot of fruits
and vegetables
The nutritional richness of fruits and vegetables
makes them an ideal target for microorganisms,
causing in particular soft rot. Certain research
revealed that pectinase enzyme synthesizing by these
pathogens causing the soft rots in fruits and
vegetables.
The study of Tarek M et al (2019) showed the
effectiveness of a biological Agent and fungicide on
the tomato rotting activity of Alternaria solani. They
determined the pectinolytic and cellulolytic activities
of the causative agents of tomato fruit rot. Their
investigation showed that A. solani produced both
cellulolytic and pectinolytic enzmes in vitro and it is
widely accepted that the production of these enzymes
is a major means by which these fungi invade the host
tissue, which agrees with the experimental findings of
Ramchandran and Kurup (2013), who showed that
pectinase-producing fungi cause spoilage of fruits and
vegetables. The activity of these enzymes decreased
with an increase in the T harzianum filtrate
percentage and chemical fungicide concentration.
Studies on pectinase activities of isolates of Erwinia
carotovora and Rhizopus sp. causes soft rot in
cabbage was studied by K.A.Bhat et al (2012) and
found that crude pectinase enzymes obtained in the
form of culture filtrate from different isolates and
were assayed by cup-plate method. Pectin lyase and
hydrolase activity of Erwinia isolates was also carried
out using viscometric assay where the percent
decrease of viscosity gives the measure of chain
splitting enzymes in the test samples.
Wakil and Oyinlola (2011) determined the diversity
of pectinolytic organisms causing soft rot disease of
five different vegetables marketed in Ibadan Nigeria.
They found that the microbial load of each vegetable
sample showed pectinolytic activity. Their results
indicated that soft rot of vegetables is caused by a
wide range of pectinolytic organisms that vary with
different vegetables andlocations of purchase.
Erwinia species were the dominating organisms
although, Pseudomonads and Xanthomonas strains
were also reported.
The fungi that cause decay and rotting of cucumber,
lemons and tomatoes in some markets was studied by
SMN Moustafa (2018). The role of CMC-ase and
polygalacturonase (PG) enzymes and its role in
corruption of these fruits were also studied.
A.alternata, P.italicum and R. stolonifer were isolated
from fruits of rotten tomato, lemon and cucumber
respectively. Enzyme production from three tested
fungi were studied and stated that enzymes play an
important role in infecting fruits and vegetables with
fungi and causing its corruption and spoilage.
Cellulase enzymes cause softening and dissolution of
primary cell walls of fruits, which are mainly
composed of cellulose and leads to easy entry of
penetration begs and haustoria of fungi into
parenchymatous cells. Poly galacturonase (PG) play
an important role in the decomposition and
disintegration of plant cells of fruits as a result of
dissolution of the middle lamella in the cell walls of
the cells. This, in turn, causes cells to disintegrate and
then to decompose and lose their cohesion until they
become rotten. Results of this study showed that
P.italicum and R. stolonier have a high ability to
produce cellulose (CMC-ases) and PG enzymes.
The study of Samuel K.C Obi et al (1981) revealed
that there is a pectic enzymes activities of bacteria
associated with rotted onion. Six bacterial genra
consisting of seven species were isolated from rooted
onions and these were a Vibrio sp.(PS1), Micrcoccus
epidermidis (PS2), P. cepacia (PS3,PS6), an
Acinetobacter sp.(PS4), a Xanthomona sp. (PS5),
Bacillus poymyxa (PS7) and Bacillus megaterium
(PS8). The enzyme extracts from the onion tissue
cultures exhibited some hydrolase activity for three
isolates, P. cepacia,(PS3), P.cepacia (PS6) and
Bacillus polymyxa (PS7).All the isolates produce both
lyase and pectinesterase enzymes and the highest
activity was recorded in Vibrio Sp. Their isolates
examined demonstrated considerable pectin lyase
activity and suggested that it may be prudent to
inhibit pectic enzymes rather than at that whole
organism.
Ishii S (1976) has observed the rapid maceration of
onion tissue by endo-pectin lyase from Aspergillus
japanicus. There is a role of pectinase enzymes in the
development of soft rot caused by Pseudomonas
fluorescens in the purple variety of onion was studied
by Simbo Aboaba (2009) and found the ability of P.
fluorescens, implicated in the soft rot of onion bulbs
to produce pectic enzymes in vitro and in vivo. The
viscometic assay confirmed this and suggests role for
the pectic enzyme in soft rot development in plants.
Determination of pectic enzyme and proteolytic
enzyme inhibitors which are known to accumulate in
tomato plants whether it also found in tomato fruits,
and if their specificity might be correlated with the
rate of rot development by three fungi G. cingulate,
C. atramentarium and B. cinerea pathogenic on
tomato fruits was reported by Brown A.E.et al (1982).
Conclusion
The various organisms such as bacteria and fungi are
responsible for soft rot of fruits and vegetables and
represent a category of phytopathogens that are very
dangerous, and this due to their ability to synthesize a

plant cell wall degrading enzymes. From these the
most important enzyme is pectinase which play
important role in rotting the fruits and vegetables.
This enzyme allow pectinolytic organism to cross the
protective barrier of fruits and vegetables, causing
soft rot in them. This is why it is also necessary to
inhibit such enzyme action causing soft rot rather than
only whole organisms.
