This document describes a systematic investigation into producing synthetic prions from recombinant prion protein. Approximately 20,000 unique conditions were tested involving varying solvent properties, incubation times and temperatures, as well as addition of factors like proteases, brain homogenate, lipids, and metals. Some conditions initially appeared positive by cell-based assay, but infectivity could not be reliably reproduced or propagated in cells. Published methods claiming to produce synthetic prions from recombinant prion protein were also tested but did not find significant infectious titers. In summary, while some conditions transiently showed possible infectivity, the study was unable to reproducibly produce high-titer synthetic prions suitable for further structural analysis.
