Cytotoxicity of Blended Versus Single Medicinal Mushroom Extracts on Human Ca...Jolene1981
ABSTRACT: The use of mushrooms contributes to human nutrition by providing low lipid content of lipids and high dietary fiber content, as well as significant content of other biologically active compounds such as polysaccharides, minerals, vitamins, and polyphenolic antioxidants. This study aimed to determine the content of polyphenols and polysaccharides, as well as the cytotoxic and antioxidative properties of several medicinal mushroom preparations. The content of total phenols and flavonoids of preparations of blended mushroom extracts (Lentifom, Super Polyporin, Agarikon, Agarikon Plus, Agarikon.1, and Mykoprotect.1) was evaluated quantitatively by using ultraviolet–visible spectroscopy spectrophotometric methods. The antioxidant capacity of the preparations was evaluated using the ABTS (2,2′-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) and ferric reducing/antioxidant power assays. The content of water-soluble polysaccharides was determined using a specific gravimetric method, based on ethanol precipitation. To determine cytotoxic effects of single and blended mushroom extracts, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and neutral red assays were conducted using human small cell lung cancer, lung adenocarcinoma, colon cancer, and brain astrocytoma cancer cells. The obtained results suggest that due to the significant content of beneficial polyphenolic antioxidants and soluble polysaccharides, use of these mushroom preparations is beneficial in maintaining good health, as well as in the prevention and adjuvant biotherapy of various human pathological aberrations. These results reveal that these extracts exhibit different cytotoxic effects on tumor cells originating from different tissues. In addition, the comparison of investigated blended mushroom extracts with three well-known commercial mushroom products derived from single mushroom species or single mushroom compounds shows that blended mushroom extracts exhibit significantly stronger cytotoxic effects on human tumor cell lines.

Glutathione plays pivotal role in protecting cells against oxidative stressinduced cellular damage and in detoxifying Xenobiotics and drug metabolism. The added photo-protective effects of oral Polypodium with its antioxidant, immunoregulatory and antiinflammatory properties aid also in prevention of chronic skin damage, photoaging, and skin cancer led by UV exposure. The world population is aging, and cancer is always considered to be one of the major causes of death all over the globe. The advent of recent drug-targeted therapies undoubtedly is going to reduce the incidence of cancer over the coming years. However, the frequency of occurrence of such chronic diseases like cancer would continue to increase. Therefore, the search for a safer and cost-effective treatment is urgently needed. Phytochemicals found in plants, foods, vegetables, tea, etc. have emerged as proven therapeutic compounds modulating signaling pathways involved in cancer. We carried out a structured search of bibliographic databases for peer-reviewed research literature using the keywords: cancer chemoprevention, flavonoids, dietary polyphenols, terpenoids, bioactive, microbiota. Quality of the retrieved papers and characteristic outcomes of the articles included in the study was assessed by employing standard tools and deductive qualitative content analysis methodology. The development of personalized supplements comprising particular phytochemicals has been the key, especially dealing with chronic inflammatory disorders like cancer. Better understanding at the molecular level explains the influence of phytochemicals on human health, which has been extensively covered through this review. Moreover, the wide collection of dietary polyphenols that has significant properties in reference to human health has been highlighted. Furthermore, the etiology of end products of such phytochemicals, especially on the modulation of gut microbiota and the host-microbial interactions thereof, need to be properly understood. The present study summarizes the chemoprevention and treatment of cancer using the bioactive components, including flavonoids, dietary polyphenols, and terpenoids. Likewise, the effect of dietary polyphenols on the human gut microbiota has been realized more recently. However, more research is needed in this field, especially focused on the communications, interlinks between the gut microbiota and polyphenols with the precise mechanism of action.

RESULTS: Overall life span (LS) was 2252.1±1742.5 days and cumulative 5-year survival (5YS) reached 73.2%, 10 years – 64.8%, 20 years – 42.5%. 513 LCP lived more than 5 years (LS=3124.6±1525.6 days), 148 LCP – more than 10 years (LS=5054.4±1504.1 days).199 LCP died because of LC (LS=562.7±374.5 days). 5YS of LCP after bi/lobectomies was significantly superior in comparison with LCP after pneumonectomies (78.1% vs.63.7%, P=0.00001 by log-rank test). AT significantly improved 5YS (66.3% vs. 34.8%) (P=0.00000 by log-rank test) only for LCP with N1-2. Cox modeling displayed that 5YS of LCP significantly depended on: phase transition (PT) early-invasive LC in terms of synergetics, PT N0—N12, cell ratio factors (ratio between cancer cells- CC and blood cells subpopulations), G1-3, histology, glucose, AT, blood cell circuit, prothrombin index, heparin tolerance, recalcification time (P=0.000-0.038). Neural networks, genetic algorithm selection and bootstrap simulation revealed relationships between 5YS and PT early-invasive LC (rank=1), PT N0—N12 (rank=2), thrombocytes/CC (3), erythrocytes/CC (4), eosinophils/CC (5), healthy cells/CC (6), lymphocytes/CC (7), segmented neutrophils/CC (8), stick neutrophils/CC (9), monocytes/CC (10); leucocytes/CC (11). Correct prediction of 5YS was 100% by neural networks computing (area under ROC curve=1.0; error=0.0).
CONCLUSIONS: 5YS of LCP after radical procedures significantly depended on: 1) PT early-invasive cancer; 2) PT N0--N12; 3) cell ratio factors; 4) blood cell circuit; 5) biochemical factors; 6) hemostasis system; 7) AT; 8) LC characteristics; 9) LC cell dynamics; 10) surgery type: lobectomy/pneumonectomy; 11) anthropometric data. Optimal diagnosis and treatment strategies for LC are: 1) screening and early detection of LC; 2) availability of experienced thoracic surgeons because of complexity of radical procedures; 3) aggressive en block surgery and adequate lymph node dissection for completeness; 4) precise prediction; 5) adjuvant chemoimmunoradiotherapy for LCP with unfavorable prognosis.

▪ Animal and epidemiological studies reveal that consuming food and beverages rich in polyphenols
(e.g., catechins, flavones, and anthocyanines) is associated with a lower incidence of cancer, and
several molecular mechanisms have been proposed for explaining this effect.
▪ However, because most of these mechanisms were observed only under specific and
nonphysiological conditions, and in most cases, with practically irrelevant concentrations, there is
still no clear-cut or universal explanation for the major events that underlie the anticancer effects
of polyphenols.
▪ In this study we present clear in vitro and in vivo evidence that the inhibition of the cancer-
associated enzyme telomerase is a key mechanism involved in cancer inhibition by
epigallocatechin gallate (EGCG), a major tea polyphenol.
▪ We demonstrate that EGCG and other selected polyphenols undergo structural rearrangements at
physiologically permissible conditions that result in remarkably increased telomerase inhibition. In
nude mice models bearing both telomerase-dependent and -independent xenograft tumors cloned
from a single human cancer progeny, only the telomerase-dependent tumors responded to
prolonged oral administration of EGCG.
▪ Thus, EGCG and likely other structurally related dietary polyphenols seem to act as prodrug-like
molecules that, once ingested and distributed, undergo structural changes that favor potent
activity against telomerase.

▪ Plant-derived ceramides are functional natural products with vari-ous physiological effects that improve water retention and skin
permeability functions.
▪ Objective:
▪ Herein, we isolated peach (Prunus persica)-derived glucosylceramide with high purity and evaluated its effects on moisture and
permeability of skin.
▪ Methods:
▪ Three separate experiments were performed: evaluations of the effect of peach-derived glucosylceramide application on the ceramide
content in a three-dimensional hu-man cultured skin model, water retention effect of the oral administration of the peach-derived
glucosylceramide on human skin functionality, and safety of the long-term admin-istration of the peach-derived glucosylceramide in
human subjects.
▪ Results:
▪ Treatments of three-dimensional cultures of human skin with peach-derived gluco-sylceramide led to dose-dependent increases in
human ceramide contents. Additionally, af-ter oral administration of ceramide to humans, dose-dependent improvements in water re-
tention functions of skin, suppression of trans-epidermal water loss and improvements in skin texture were observed. Furthermore, no
significant changes in subjective/objective symptoms, physical characteristics, or laboratory test values were observed in human sub-
jects following long term oral administration of ceramide at doses that were 4.5-9-times greater than recommended. Taken together, the
present data indicate the positive effects of peach ceramide on skin function and no adverse effects.

40. Peach Tree Pollen and Prunus persica Sensitisation and Allergy
in Children and Adolescents (2019)
▪ Background: Although plant and fruit pollens are entomophilous and relevant in exposed workers,
we have shown a high frequency of sensitisation and symptoms induction of peach tree pollen
(PTP) and Prunus persica 9 (Pru p 9) in adults from areas of peach cultivars.
▪ Methods: We studied the sensitisation and clinical relevance of PTP and Pru p 9 in a large group of
children and adolescents aged 3-19 years. A detailed questionnaire plus skin prick testing to
prevalent allergens, PTP, and Pru p 9 were carried out. The clinical relevance was established by
nasal provocation test (NPT) and symptom score index.
▪ Results: We evaluated 685 children (mean age 8.75 ± 3.3 years, median 9 years), 52% of them
female. Sensitisation to PTP occurred in 20% of the cases following olive tree (33%) and Phleum
pratense (26%). In a randomly selected subgroup of subjects sensitised to PTP, 30% were skin prick
test-positive to Pru p 9. Most cases had rhinitis or rhinoconjunctivitis. NPT showed the relevance of
PTP and Pru p 9 in the induction of symptoms.
▪ Conclusion: PTP and Pru p 9 are relevant in the induction of sensitisation and respiratory symptoms
in children and adolescents. This allergen should be evaluated in children living in regions of peach
tree cultivars.
