Interleukin 1β polymorphisms (+3954, -511 and -31) were analyzed in chronic periodontitis patients from North India. The study found that the T allele of the +3954 variant and the CT genotype were more frequent in patients compared to controls, suggesting they are risk factors for chronic periodontitis. A literature review found supporting evidence for associations of the +3954 and -511 variants with chronic periodontitis in other populations as well. The study concluded that these two IL-1β polymorphic sites may serve as potential markers for chronic periodontitis, though more research is needed in a larger sample size.

The nematodes of the genus Trichinella are known to cause the pressing foodborne parasitic disease Trichinellosis and these parasites are known to complete all stages of development in one host with the enteral and parenteral phases observed during infection. Proteomics, in general, pertains to the systematic identification and quantification of the totality of proteins, which is the proteome of a biological system, at a specific point in time. The available proteomic studies have paved the way to identify and characterize Trichinella stage-specific proteins reacting with infected host-specific antibodies. Yet, very few contributions provide any information about changes in the global proteomic serum profile of Trichinella-infested individuals. Studies demonstrate that various Trichinella species and their phases of the invasion produce a characteristic proteomic pattern in the serum of experimentally infected pigs. Recent investigations have found that T. spiralis infection induced strong regulatory T cell responses through parasite excretory-secretory (ES) products, characterized by an increase of some regulatory T cells and growth factors. T. spiralis has also been reported to induce the angiogenic molecule vascular endothelial cell growth factor (VEGF) during nurse cell formation towards the induction of angiogenesis for nutrient supply and waste disposal. Herein, the various analogs considered in these studies include the serum, excretory-secretory proteins, surface proteins, immune-reactive proteins from muscle larvae (ML) and so on. Intestinal cultures, striated muscle tissues, pigs, mice, beavers, contributions from patients are some of the major models exploited for this purpose. The current analysis focuses on recapitulating the recent findings driven on this area to create a common ground for further studies and to ease any difficulty in continuing the proteomic analysis of T. spiralis using in vitro and in vivo models.

Background: Tumor necrosis factor‑alpha (TNF‑α), a “major inflammatory cytokine,” not only plays an important role in periodontal destruction but also is extremely toxic to the host. Till date, there are not many studies comparing the levels of TNF‑α in serum and its relationship to periodontal disease.
Aim: Our study aimed to compare the serum TNF‑α among the two study groups, namely, healthy controls and chronic periodontitis patients and establish a correlation between serum TNF‑α and various clinical parameters. Hence, an attempt is made to estimate the level of TNF‑α in serum, its relationship to periodontal disease and to explore the possibility of using the level of TNF‑α in serum as a biochemical “marker” of periodontal disease. Materials and Methods: Forty individuals
participated in the study and were grouped into two subgroups. Group A – 20 systemically and periodontally healthy controls. Group B – twenty patients with generalized chronic periodontitis.
The serum samples were assayed for TNF‑α levels by enzyme‑linked immunosorbent assay method.
Results: The mean serum TNF‑α cytokines for Group B Generalized chronic periodontitis (GCP) was 2.977 ± 1.011, and Group A (healthy) was 0.867 ± 0.865. The range of serum TNF‑α was from (0.867 to 2.977). Serum TNF‑α cytokines had highly significant correlation with all clinical parameters (plaque index, probing pocket depth, clinical attachment loss, and gingival index) among all study participants (P = 0.001). Conclusion: These observations suggest a positive association
between periodontal disease and increased levels of TNF‑α in serum. It can be concluded that there is a prospect of using the estimation of TNF‑α in serum as a “marker” of periodontal disease in future. However, it remains a possibility that the absence or low levels of TNF‑α in serum might indicate a stable lesion and elevated levels might indicate an active site but only longitudinal studies taking into account, the disease “activity” and “inactivity” could suggest the possibility of using
TNF‑α in serum as an “Indicator” of periodontal disease.

Intervertebral disc diseases is otherwise known as breakdown of one or more discs that separate the bone of the spine, Which cause pain in the back, neck as well as frequent pain in legs and arms. The general role of intervertebral discs is to provide the cushioning between vertebrae and absorb the pressure that is exerted on the spine. Generally low back pain LBP is considered as common disorder from disc degenerative diseases. The main reason of the LBP is due to contribution of intervertebral disc diseases and combination of genetic and environmental factors. Most of these factor identified , But some of these remain unidentified. However recent researches identified that variations in several genes may influence the risc of intervertebral disc diseases . Most of the variations associated with genes related with immune function leads to increase risk of intervertebral disc diseases . Most of these genes plays a vital role in immune response like inflammation and water loss dehydration of the discs, Which causes their degeneration. responsible for the disease development but sometimes these changes are also
protective in nature.

29 periodontitis patients (males: females =23:6) presenting with new cases of
chronic periodontitis, evaluated in the Department of Periodontology,

The analysis of +3954 variant revealed the T-allele to be more frequent among the
patients when compared with healthy controls and the CT genotype to be a
significant risk factor for the development of the disease which is in accordance
with studies from Brazil and South Indian states

negative association for -511 variant
Shete AR et al suggested strong linkage disequilibrium of -31T/C variant with
511C/T among both patients and controls.

This study involving North-Indian population suggests the -511 and +3954 variants
are risk factors for chronic periodontitis and that these two polymorphic sites
encoded along IL-1β can be used as potential markers for chronic periodontitis, but
it needs evaluation in a larger cohort of samples.
