• CURCUMIN IS AN ORANGE–YELLOW CRYSTALLINE POWDER ESSENTIALLY
INSOLUBLE IN WATER.
• CURCUMIN IS YELLOW IN COLOR SHADE AND IS MOST PRECIOUS CONSTITUENT
OF TURMERIC.
• CURCUMIN IS ONE OF THE THREE CURCUMINOIDS THAT APPEAR IN TURMERIC,
THE OTHER TWO BEING DESMETHOXYCURCUMIN AND BIS-
DESMETHOXYCURCUMIN.
3. EXTRACTION OF CURCUMIN
• CURCUMIN IS EXTRACTED FROM TURMERIC WHICH IS DERIVED FROM RHIZOME OF
THE PLANT CURCUMA LONGA.
• THE EXTRACTION PROCESS IS CARRIED OUT IN TWO STEPS SELECTIVE REMOVAL OF
THE TURMERIC OIL AND EXTRACTION OF PIGMENT

2. THE RESIDUE IS THEN FEED FROM THE SOLVENT AND EXHAUSTIVELY EXTRACTED
WITH ANOTHER SOLVENT LIKE METHANOL TO RECOVER CURCUMIN.AFTER
REMOVING THE SOLVENT, THE CONCENTRATED EXTRACT DISSOLVED IN
ALKALI,FILTERED&ACIDIFIED WITH ACID TO PRECIPITATE TO PIGMENT.
4. ISOLATION OF CURCUMIN
• CURCUMIN IS INSOLUBLE IN WATER; AN ORGANIC SOLVENT HAS BEEN USED FOR ITS
ISOLATION.
• DEVELOPED A TECHNIQUE FOR ISOLATING CURCUMIN FROM GROUND TURMERIC.
• THEY MAGNETICALLY STIRRED THE GROUND TURMERIC IN DICHLOROMETHANE AND
HEATED AT REFLUX FOR 1H
• THE MIXTURE WAS SUCTION-FILTERED, AND THE FILTRATE WAS CONCENTRATED IN
A HOT-WATER BATH MAINTAINING AT 50°C.
• THE REDDISH-YELLOW OILRESIDUE WAS TRITURATED WITH HEXANE AND THE
RESULTING SOLID WAS COLLECTED BY SUCTION FILTRATION.
5. CHARACTERIZATION OF CURCUMIN
• THE FABRICATED CURCUMIN IS ANALYZED WITH HIGH PER-FORMANCE LIQUID
CHROMATOGRAPHY/MASS SPECTROMETRY (HPLC/MS) OF AGILENT 1260 SERIES
SINGLE QUADRUPOLE LC/MS SYSTEMS (AGILENT TECHNOLOGIES, USA.
• STOCK SOLUTIONS OF CURCUMIN (200 ΜG/ML) WAS PREPARED BY DISSOLVING
ACCURATELY WEIGHED 10 MG OF CURCUMIN IN METHANOL USING 50 ML
VOLUMETRIC FLASK.
• THIS SOLUTION WAS FURTHER DILUTED WITH METHANOL TO OBTAIN STANDARD
SOLUTIONS IN THE CONCENTRATION RANGE OF 1–20 ΜG/ML.
• THE SEPARATION WAS CARRIED OUT IN A ZORBAX ECLIPSE. XDB C18 COLUMN (4.6 ×
150 MM, 5Μ M) WITH A C18 GUARD COLUMN MAINTAINED AT 24 °C.
6. • THE ELUTION WAS PERFORMED AT FLOW RATE OF 0.5 ML/MIN WITH A GRADIENT MOBILE
PHASE FROM 10 TO 30 % ACETONITRILE IN WATER IN 5 MIN, FOLLOWED BY AN INCREASE
FROM 30 TO 70 % ACETONITRILE IN ANOTHER 20 MIN.
• THE INJEC-TION VOLUME WAS OF 5 ΜL. THE DAD ACQUISITION WAVE- LENGTH FOR
DEMETHOXYCURCUMIN, BISDEMETHOXYCURCUMINAND CURCUMIN DETECTION WAS SET AT
425 NM.
• THE MASS DETECTOR USED A MM-ES MULTIMODE SOURCE.
• OPENLAB SOFTWARE (AGILENT TECHNOLOGIES) WAS EMPLOYED TO CONTROL AND COLLECT
SAMPLE.
• THE ION PEAKS AT M/Z 339.0, 309.1, AND 369.1 ([M + H]+) WERE DETECTED FOR
DEMETHOXYCURCUMIN, BISDEMETHOXYCURCUMIN AND CURCUMIN.

AS CURCUMA LONGA.
• IT IS A FLAVOURING SPICE FROM A FLOWERING ROOT PLANT AND BELONGS TO
THE GINGER FAMILY.
• IT’S ORIGIN IS PRIMARILY INDIA AND ALSO OTHER COUNTRIES IN SOUTHEAST
ASIA AND ASIDE FROM ITS COOKING USE AND BEING USED AS A SPICE FOR
CENTURIES.
