History In 1932, the first gasoline tax was created and cost consumers one cent per gallon. Over the
past 80 years, this tax has constantly increased. Events such as the Korean War in 1951 and the
construction of interstate highway systems in 1959 caused the tax to raise to its current level of 18.4
cents per gallon, which is its current rate (Williams, 2005). Revenues collected have been utilized for
a variety of purposes. For almost 20 years between 1932 and 1956, taxes collected were allocated to
the general fund. The Federal Aid Highway Act of 1956 established the Highway Trust Fund, where
all monies collected were allocated until 1983 when some funds were placed in a mass transit account.
Today, 15.44 cents of every 18.4 cents is allocated to the Highway Transit Fund (Buechner, 2007).
Controversy over the federal gasoline tax has always been present, with opposition from the public to
every proposed increase. Recently, when discussions arose to use the tax to generate revenue, many

models into group intervention, there are a lot of advantages that can benefit group members in
different ways. First of all, a therapeutic alliance is seen as one of the preconditions of the CBT work,
and within a group, members can identify and develop therapeutic relationships with not only the

being successful and happy. To fulfill that dream in America you need to have courage and the
abilities to achieve your dream.
The Pursuit Of Happiness is based on a true story about a man called Chris Gardner. Chris Gardner is
a great example for somebody that wants to Pursuit his American Dream. The Pursuit Of Happiness is
about how Chris Gardner is willing to make sure he achieves his happiness. For living, Chris Gardner
is a salesman who sells bone density scanners. Eventually he figured out that nobody wanted to buy it
because it is a luxury device in the medical business. Sometime in the beginning of the movie, there is
a scene where we see Chris Gardner approaching and trying to talk to Mr.Twistle. We see a long shot
of them both which shows us that they both are dressed in suits, which means that they both might be
office workers. The long shot then drifts and became a mid shot. The director could have purposely
done that to show that Mr.Twistle is in a hurry and has no time to speak with Chris. Chris Gardner and
Mr.Twistle got in the same cab. While they are in the same taxi together, we see that Chris is talking to
Mr.Twistle but the close up of Mr.Twistle s face shows that he s not interested in what Chris is saying.
This is because Twistle is just playing with a Rubik s cube. We then see an over the shoulder shot
which shows that it could mean that Chris is desperately wanting to talk to Mr.Twistle. Mr.Twistle
then says to Chris I m sorry, this thing is impossible. The extreme close up of Chris s face shows that

The first weeks of the summer fellowship in the VDS stream was a bit hectic. They consisted of
learning protocols and techniques. Three plates of PNiC BSa4 colonies were grown by my partner and

concentration was 68ng/ul. The concentration of this sample was low ( lt;100ng/ul) because I had
skipped the first step in the cleanup process that prepares the column filter for maximum DNA
binding. PCR2 and PCR clean up were completed again and the concentration of the sample was
averaged to be 147.9ng/ul. Since the concentrations of the samples were adequate, I moved onto PNiC
BSa4 cloning and prepared a sample to be DNA sequenced. RE digest was completed on two
PNIcBSa4 prep samples, with the gel indicating that both samples were successfully cut and nano
drop results indicating high purity levels with low concentrations (~32ng/uL). Results from
sequencing were nucleotide blasted and confirmed to be PNiCBSa4. I talked to a mentor about my
samples and was told that I probably needed a higher concentration ≥100ng/uL, for cloning to be
successful. In order to increase the concentration 4 samples of 50uL each were cut and PCR clean up
into one sample that had a concentration of 123.5ng/ul. The cloning protocol was completed that week
and when the plated were checked, no colonies were present on either of the two plates. My first
