immunotoxic effects; …

Procedure and criteria for the approval of active substances, safeners and synergists
pursuant to Chapter II
3.6.5 An active substance, safener or synergist shall only be approved if, on the basis of
the assessment of Community or internationally agreed test guidelines or other
available data and information, including a review of the scientific literature, reviewed
by the Authority, it is not considered to have endocrine disrupting properties that may
cause adverse effect in humans …

extensive parts of the life cycle of the
organism
3.3.1.3. Human relevance
“According to the scientific criteria for determining ED properties applicable
to the BP and PPP Regulations, ‘A substance shall be considered as having
endocrine-disrupting properties that may cause adverse effect in humans [. .
.] unless there is evidence demonstrating that the adverse effects identified
are not relevant to humans’.
Note that the assessment of human relevance does not refer to adversity as
such, but rather to the question as to whether an effect elicited by a
substance in a test animal could also be elicited in a human being.
Therefore, to disprove human relevance it is necessary to demonstrate
differences in the mechanisms of action of the substance in human and in
test animals by having recourse to the MoAs.
Therefore, human relevance is addressed in the context of MoA analysis
(Section 3.5.4.4).”
3.3.1.3. Human relevance

Validity status of reprotox testing
1) Not robust (about 25% equivocal studies), [Bailey et al., 2005]
2) 74 industrial chemicals tested in New Chemical Database: 34
showed effects on offspring, but only 2 chemicals have been
classified as developmental toxic [Bremer & Hartung, 2004]
3) 55% of positives in screening studies not in multi-generation
studies [Bremer & Hartung, 2004]
4) Group size limits statistical power [Hotchkiss 2008]
5) 61% inter-species correlation [Hurrt 2003, Bailey 2005]

(b) does not have an inherent capacity to cause endocrine disrupting, neurotoxic or immunotoxic effects; … ANNEX II Procedure and criteria for the approval of active substances, safeners and synergists pursuant to Chapter II 3.6.5 An active substance, safener or synergist shall only be approved if, on the basis of the assessment of Community or internationally agreed test guidelines or other available data and information, including a review of the scientific literature, reviewed by the Authority, it is not considered to have endocrine disrupting properties that may cause adverse effect in humans … (same for the environment)

Human relevance “According to the scientific criteria for determining ED properties applicable to the BP and PPP Regulations, ‘A substance shall be considered as having endocrine-disrupting properties that may cause adverse effect in humans [. . .] unless there is evidence demonstrating that the adverse effects identified are not relevant to humans’. Note that the assessment of human relevance does not refer to adversity as such, but rather to the question as to whether an effect elicited by a substance in a test animal could also be elicited in a human being. Therefore, to disprove human relevance it is necessary to demonstrate differences in the mechanisms of action of the substance in human and in test animals by having recourse to the MoAs. Therefore, human relevance is addressed in the context of MoA analysis (Section 3.5.4.4).”

Validity status of reprotox testing 1) Not robust (about 25% equivocal studies), [Bailey et al., 2005] 2) 74 industrial chemicals tested in New Chemical Database: 34 showed effects on offspring, but only 2 chemicals have been classified as developmental toxic [Bremer & Hartung, 2004] 3) 55% of positives in screening studies not in multi-generation studies [Bremer & Hartung, 2004] 4) Group size limits statistical power [Hotchkiss 2008] 5) 61% inter-species correlation [Hurrt 2003, Bailey 2005] Moreover a) Lack of objectivity of in vivo tests b) Tests at high concentration ALTEX 2018, 35:139-162

  1. I have the pleasure to introduce this interesting topic. I’ve asked to include it in the agenda because I have the impression that there is still confusion with need for further discussion
  2. I am representing ecopa as stakeholder at EFSA. Ecopa is the european consensus-platform for alternatives to promote the 3Rs in science.
  3. Actually, I work for CAAT-Europe …. I have also a contract with TEAM mastery whoch is a private consultancy company
  4. If we want to talk about endocrine disruptors, we first need to set the correct legislative framework. Regulation on plant protection products excludes the endocrine disruptor activity in approved active substances. This concept is repealled in Annex II
  5. How to decide whether a substance has endocrine disruptors activity? The OECD guidance 150 is very helpful and precise. It has been also recently updated. There are 5 levels of investigation….
  6. In addition, last year ECHA and EFSA together published this guidance. They did it together because also aproved biocides substances should have no endocrine disruptors activity. The problem of this guidance is that it is very much focused on in vivo methods only and if you follow the instruction, it is very difficult to stop before all 5 OECD levels are explored. But this is absolutely not a guarantee of safety
  7. What is relevant is human relevance! That means how much I am guessing of the real effect of a substance on humans. Well out of 150 pages, this is the only mention of that
  8. i.e. there is only the possibility of a false positive result from in vivo. To summarize, if an in vivo study is negative, there is no concern and if it is positive I may still demonstrate that this result can be disregarded. Are we sure?
  9. The situation of the in vivo studies is not reassuring. Remember that the vast majority of in vivo studies are not validated. As far as I know, only 4 are validated and two of them in the area of endocrine disruptors. This is why we can say that 25% returns equivocal results The data of 25% disordant result arrives
  10. OECD Level 5 is the EOGRTS. This test is quite demanding… Not mentioned here, but we’re also talking of more than 2000 rats per study We don’t care about the money or the number of animals, we may ask to have this study for all substances. Well, in the EFSA databses there are 2350 reports for pesticides. This means that it is simple impossible. If we go on asking for more in vivo studies we will have less safety data. There is a solution Without considering the fact that also ECHA is now asking many of this type of study.
  11. Positive results should be adjusted with toxicokinetics False negative results are negligible and mainly due to lack of metabolism
