ABSTRACT
Background:The main objective of the study is to determine the anti-arthritic effect of whole plant ethanolic extract of Polygonum glabrum
belonging to the family Polygonaceae in Female wistar rats using the Freund’s Complete Adjuvant (FCA) model . Methods:The plants areal
parts were collected near Tirupathi hills, Chittoor district of Andhra Pradesh in India. The Phytoconstituents were identified through the
chemical tests. Ethanol (95%) was used to obtain the whole plant extraction through Soxhlet extractor. Female SD rats were used for antiarthritic
screening. Arthritis was induced using FCA, and the anti-arthritic effect of the ethanolic extract of P.glabrum was studied at doses
of 250 and500 mg/kg. The effects were compared with those of indomethacin (10 mg/kg). At the end of the study, theliver enzyme levels were
determined and a radiological examination was carried out. Results and Discussion:The preliminary phytochemical analysis of the ethanolic
extract of Polygonum glabrum showed the presence of alkaloids, tannins, flavonoids and saponins. P. glabrum at 250 and 500 mg/kg
significantly inhibited the FCA-induced arthritis in the rats. This was manifested by as a decrease in the paw volume. The arthritic control
animals exhibited a significant decrease in body weight compared with control animals without arthritis. P. glabrum treated animals showed
dose dependent reduction in decrease in body weight and arthritis.At the same time, P.glabrum significantly altered the biochemical and
haematological changes induced by FCA (P < 0.05). The anti-arthritic effect of P.glabrum was comparable with that of Indomethacin.
Conclusion:The whole plant extract of P.glabrum showed significant anti-arthritic activity against FCA-induced arthritis in female Wistar
rats.

alcoholic (AlE) and aqueous extract (AqE) from Commelina benghalensis root (CB) on EtOH-induced hepatic
injury in Wistar rat. Hepatotoxic parameters studied in vivo include serum transaminases (AST, and ALT),
ALP, bilirubin, protein, lipid profile (Cholesterol, triglyceride, VLDL and HDL) and level of antioxidants
together with histopathological examination. Liv 52® was used as a reference hepatoprotective agent
(5ml/kg-1b.w.). AlE and AqE (200 mg/kg-1b.w.) on oral administration decreased the level of AST, ALP, ALT,
bilirubin, cholesterol, triglyceride, VLDL, MDA and increased the level of protein, HDL and antioxidants (SOD,
GSH and CAT) in rats being treated with ethanol (EtOH). Pentobarbitone -induced sleeping time study was
carried out to verify the effect on microsomal enzymes Histopathological observations confirmed the
beneficial roles of MF against EtOH-induced liver injury in rats. Possible mechanism may involve their
antioxidant activity

The Effects of Rauwolfia Vomitoria Extract on the Liver Enzymes of Carbon Tet...IOSR Journals
Rauwolfia vomitoria is a natural medicinal plant which has been used over the years for the treatment of various ailments. The effects of extract of rauwolfia vomitoria on liver enzymes of carbon tetrachloride induced hepatotoxicity were observed in adult wistar rats weighing between 120g and 190g. They were divided into four groups A,B, C and D of six rats each. Group A served as the control and received 0.41ml of distilled water. The experimental groups B, C and D received different doses of drugs as follows : group B received 0.50ml of rauwolfia vomitoria extract, group C received 0.5ml of carbon tetrachloride and group D received 0.41ml of carbon tetrachloride + 0.4ml of rauwolfia vomitoria extract. The drugs were administered once in a day using intubation method for a period of twenty one days. Twenty four hours after the last administration, the animals were anaesthetized under chloroform vapour and dissected . liver tissues were removed and weighed. Blood samples were collected by cardiac puncture and Serum samples were separated from clot by centrifugation using bench top centrifuge. Activities of serum aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase were determined using randox kit method. The relative liver weight for carbon tetrachloride treated group were significantly higher (p<0.001)><0.001) than the control. The extract exhibited a liver protective effect against carbon tetrachloride induced hepatotoxicity

In this study, the innate immuno-modulatory effects and anti-cancer action of arabinogalactan (AG), a derivative of a well-known orchid, Anoectochilus formosanus, were investigated. The innate immunomodulatory effects of AG were determined in vitro using RAW 264.7 cells for microarray analysis, and in vivo using BALB/c mice administrated with AG at 5 and 15 mg/kg intra-peritoneally for 3 weeks. The anti-cancer activity of AG was evaluated by CT26 colon cancer-bearing BALB/c mice. The microarray
analysis was performed to evaluate the innate immunity and demonstrated that AG significantly induced the expression of cytokines, chemokines, and co-stimulatory receptors, such as IL-1, CXCL2, and CD69.
An intraperitoneal injection of AG in mice increased the spleen weight, but not the body weight. The treatment of mitogen, LPS significantly stimulated splenocyte proliferation in AG treated groups. The AG treatment also promoted splenocyte cytotoxicity against YAC-1 cells and increased the percentage
of CD3+CD8+ cytotoxic T cells in innate immunity test. Our experiments revealed that AG significantly decreased both tumour size and tumour weight. Besides, AG increased the percentage of DC, CD3+CD8+ T cells, CD49b+CD3− NK cells among splenocytes, and cytotoxicity activity in tumour-bearing mice. In addition, the immunohistochemistry of the tumour demonstrated that the AG treatments increased the tumour-filtrating NK and cytotoxic T-cell. These results demonstrated that AG, a polysaccharide derived
from a plant source, has potent innate immuno-modulatory and anti-cancer activity. AG may therefore be used for cancer immunotherapy.
Comparison of the Acute Hypervolemic Capacities of Erythropoietin and U-74389...CrimsonpublishersITERM
Aim: This study compared the hyper volemic capacities of erythropoietin (Epo) and antioxidant drug U-74389G based on 2 preliminary studies. The provided results at mean corpuscular volumes (MCV) levels augmentation were co-evaluated in a hypoxia re oxygenation protocol of an animal model.
Materials and methods: MCV levels (MCVl) were evaluated at the 60th reoxygenation min (for groups A, C and E) and at the 120th reoxygenation min (for groups B, D and F) in 60 rats. Groups A and B received no drugs, rats from groups C and D were administered with Epo; whereas rats from groups E and F were administered with U-74389G.
Results: The first preliminary study of Epo non-significantly increased the MCVl by 0.30%+0.39% (p-value=0.4430). However, the second preliminary study of U-74389G significantly rised the MCVl by 1.60%+0.43% (p-value=0.0005). These 2 studies were co-evaluated since they came from the same experimental setting. The outcome of the co-evaluation was that U-74389G has 4.352528-fold hypervolemic potency than Epo (p-value=0.0000).
Conclusion: The anti-oxidant capacities of U-74389G accelerate the acute hypervolemic properties; presenting 4.352528-fold rise on MCVl than epo (p-value=0.0000)

Traditional medicine still remains the main recourse for a large majority of people for treating health problems in African. Therefore, the aim of this work is to assess the toxicological effect of the fresh ripe fruits using two solvents for extraction. The toxicological evaluation of aqueous-methanol extract of Dennettia tripetala fresh ripe fruits at 100, 200 and 400 mg/kg body weight for 14 days on some biochemical parameters in wistar rats was investigated. The extract at all the doses tested show non-significant (p > 0.05) increase from the control in ALT, AST, ALP, total protein, albumin, direct bilirubin, creatinine, Na+ and K+, while the level of total bilirubin and urea show significant (p < 0.05) increase from the control at 400mg/kg body weight. The levels of SOD, GPx, GST, and GSH in the serum were significantly (p < 0.05) decrease in the treated rats at 200 and 400mg/kg body weight, whereas the level of MDA and CAT showed non-significant (p > 0.05) increase in all the animals. The results of this finding indicated that the aqueous-methanol extract may not have serious effect on the liver and the kidney at 100 mg/kg b.d., but may be toxic at high doses as observed in the acute toxicity, sub-acute results and antioxidant parameters where it shows a dose-specific effects.

In recent studies Teucrium polium(T. polium ) was known as a hypoglycemic plants. But further research is needed to better understand the effect of Teucrium polium and biological active part of it. The purpose of this investigation is to examine the effect of different chromatographic fractions of aqueous and alcoholic extract of this plant on the level of insulin secretion and glucose content in hyperglycemic rat model. Also, our aim is determination of biological active fraction of aqueous and alcoholic extract of this plant. This study was carried out on the 36 rats. Hyperglycemia induced by administrating of 50 mg/kg alloxan intraperitoneally and glucose level was monitored for hyperglycemic status. Hyperglycemic was confirmed by blood glucose measurement. In each experiment 100 grams of Teucrium polium aerial parts powder were boiled with 2 Litter of distilled water for 36 h. The decoction preparation was then filtered through a gauz cloth followed by filtration through filter paper. The extract was evaporated to one-fifth of its original volume and kept at 4oC until its use. Determination of different fraction aqueous extract effect of Teucrium polium on glucose level and insulin secretion was carried out. Blood was collected from the tail of the rats. Then glucose and insulin level was evaluated. The hyperglycemic animals showed significant decrease in the blood glucose level in rats administered with fourth fraction compared with other factions. Administration of fourth fraction Teucrium polium aerial parts extract cause increase in insulin levels in alloxan-treated rats. Results suggest that treatment of fourth fraction Teucrium polium aerial parts extract may be useful in preventing the increase of glucose level in hyperglycemic rats. The interesting phenomenon of our results has shown that fourth fraction given parenterally possesses a hypoglycemic effect in alloxan hyperglycemic rats. Fourth fraction was found biological active and to be responsive to glucose challenge as evidenced by increase in insulin secretion.

Objective:Many of the available therapies for diabetes have a number of serious adverse effects; therefore the search for more effective and safer hypoglycaemic agents becomes a paramount. This research was set to investigate the antidiabetic potential and cytoprotective effect of ethanolic root extract of Salacianitida on alloxan induced hyperglyceamic rats. Method:Acute toxicity and phytochemical constituents were evaluated using standardized methods. The study lasted for a period of thirty days comprising of two phases: induction phase and treatment phase. Thirty animals were grouped into five groups of six rats each. Group 1 and 2 serve as normal and toxic control respectively while groups 3, 4, and 5 were treated with 750, 1500mg of S.nitidaand 5mg glibenclamide respectively. The BGL, food and fluid intake was monitored daily while the body weight was measured on weekly. The last day of the study after an overnight fast, the animals were loaded with glucose and the OGTT measured prior to and at 30minutes interval for two hours after which the animals were sacrificed under ether anaesthesia and the organs isolated for histological examination. Results/Conclusion:The results of the present study indicate that S.nitida possessantidiabetic potentials and cytoprotective effect noted in its significant percentage reduction in the glucose level, slight increase in body weight and restoration of the toxic organs to normal.

The aqueous extract of Celosia argentea showed no evidence of single dose toxicity 2000 mg kg when studying acute toxicity on biochemical, hematological or histological parameters. The aim of the study is to evaluate the acute toxicity effect of Celosia argentea leaf extract for 14 days in female albino rats. The aqueous extract of Celosia argentea showed no evidence of single dose toxicity 2000 mg kg when studying acute toxicity on biochemical, hematological or histological parameters. Liver diseases are still a worldwide health problem due to drug-induced hepatotoxicity. It may occur as an unexpected idiosyncratic reaction, or nontoxic drug or it may be an expected consequence of the intrinsic toxicity of a drug, taken in a sufficiently large dose to cause liver injury. A highly potential therapeutic agent or a medicinal extract is necessary for the preventive action of the hepatic disorders leading to the inflammation and drug inducing liver injury. The present study proved the medicinal plant with supportive therapeutic efficacy. Albino wistar rats of either sex are induced by Rifampicin and D-Galactosamine orally at a dose of 1g/kg and 400 mg/kg for a period of 28 days, and were treated with ethanolic extract of the stems of Andrographis lineata Nees (EEALN) orally at a dose of 200 and 400 mg/kg/day. The biochemical parameters such as serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), alkaline phosphatase (ALP),Total protein, and serum bilirubin (Total and Direct) were estimated to assess the liver function. Ethanolic extract of the stems of Andrographis lineata Nees (EEALN) showed the hepatoprotective activity by decreasing the levels of serum hepatic marker enzymes. Silymarin is used as a Standard drug. Histopathological studies were performed to confirm the biochemical changes in the hepatocytes. Toxicological studies were carried out with the extract and 2000 mg/kg b.wt. is considered as the safe dose with no mortality and adverse effects.
Comparative Cytotoxic Activities of the Flavonoid-Rich Ethyl Acetate Fruit Ex...inventionjournals
The fruit of Pouteriacampechianahas been previously reported to contain flavonoids and polyphenolic substances with high in vitro anti-oxidative effects. Its anti-tumorigenic activities have not been previously demonstrated. This study seeks to demonstrate the cytotoxic effects of the flavonoid-rich ethyl acetate fraction of the fruit extract of P. campechiana(EAFFPC) against K562 leukemic cell lines and healthy human whole blood cells. The standardized MTT-dye cell viability assay was carried out to measure the cytotoxicity of EAFFPC against the aforementioned cell lines cultured in RPMI. The assay showed that at a 60 µg well concentration, EAFFPC exhibited a 55.4% cell viability which is significantly lower than the cell viability obtained with 10 µg of vincristine. In contrast, EAFFPC demonstrated concentration-dependent cytoprotective properties in healthy human whole blood cells (HHWBC). This study confirms specific nonconcentration-dependent cytotoxic effects on K562 leukemic cell lines while exhibiting a concentrationdependent cytoprotective effects in HHWBC
Objective: Ischemia-reperfusion (I/R) leads to reactive oxygen species formation and cell death in kidney tissue with injury and organ transplantation. Simvastatin (SIM) is an antioxidant, anti-inflammatory, and anticoagulant agent. Alterations in I/R-induced acute kidney injury model with SIM treatment were analyzed.
Study Design: Wistar rats (n=28) were grouped into Sham, Ischemia, I/R, and I/R+SIM treated. Left rat kidney renal vessels were clamped for 60 minutes for ischemia, and the I/R group had 6 hours of reperfusion. 10 mg/kg SIM was given orally for 28 days. MDA, GSH, and MPO were analyzed. Kidney tissues were paraffin embedded, and primary antibodies TNF-α and caspase-3 were applied for immunohistochemistry.
Results: In the I/R group, intense inflammatory cell infiltration around the vessels and necrosis in the glomerular structures were observed. In the treated group, proximal and distal tubular cells were found to be close to normal. Immunoexpression of caspase-3 in the ischemia group was positive in degenerative glomeruli. In the treated group, TNF-α expression was negative in the glomerular structures. MDA and MPO levels were significantly increased in ischemia and I/R.
Conclusion: We suggest that SIM treatment improved kidney tissue structure and function in a model of I/R injury.
